Unlocking more lipid nanoparticle insights with Zetasizer Advance

Quality particles need quality data

Particle Works, a sister brand of Dolomite Microfluidics, develops and manufactures expertly engineered nanoparticle synthesis platforms for the pharmaceutical and biotech industries. Its innovative systems are used around the world to produce lipid nanoparticles (LNPs), important delivery vectors for mRNA-based medicines such as vaccines and gene therapy.

Developing mRNA-based therapeutics is complex work, requiring highly skilled scientists, high-tech microfluidics platforms and sophisticated analytical equipment, and of course the mRNA itself.

As Ben Knappett, Head of Science and Applications at Particle Works, points out: “You don't want to be constantly re-ordering highly expensive mRNA because your process is inefficient. We need ways of reproducibly and reliably generating the particles that encapsulate the mRNA.” 

LNP developers need reliable, high-quality data to optimize processes and product performance, and this only comes from analytics that provide a full and accurate understanding of their particles. In 2018, Particle Works added the Zetasizer Advance Ultra to its lab to deliver that data, robustly and quickly.

The Zetasizer significantly reduced dependence on transmission electron microscopy (TEM), a reliable yet expensive, time-consuming and low-throughput process which became a limiting factor in production efficiency.

The solution: measurement and data guidance in one place

The company turned to the Zetasizer Advance Ultra to produce detailed data across multiple quality attributes of its LNPs. The team was also impressed by the insights the system generated and the robustness of the data it delivers.

Producing LNPs from 40 to 800 nm, particle size consistency is a key parameter for Particle Works. Using the Zetasizer Advance, scientists at the company can determine the average particle size across this full range, as well as the size distribution.

“The most important pieces of information we look at are the average size and the PDI (polydispersity index),” Dr Knappett explains. PDI is a key indicator of batch uniformity and the stability of the LNPs.

Using the Multi-Angle Dynamic Light Scattering (MADLS) and particle concentration functions, drug loading efficiency and therapeutic encapsulation, which directly impact treatment delivery and efficacy, can be determined. 

Dr Knappett also sees opportunities to take further advantage of Zetasizer Advance capabilities such as zeta potential measurements to monitor LNP surface characteristics. Surface characteristics such as charge have direct implications for tissue targeting and cellular uptake.

“As we move on to increasingly complex formulations,” he adds, “things like surface charge are going to be really important for us.”

Physicochemical analysis functions are crucial to Particle Works, and the data quality guidance provided by the Zetasizer Advance software means they can interpret data and see issues with samples that may not be obvious. 

Dr Knappett explains that the Zetasizer Advance software provides advice on how to better run samples, reducing the chance that they might inadvertently discard samples based on a misinterpretation of the data.

Saving time without sacrificing quality

As LNPs grow more complex, vaccine developers have to monitor an ever-growing list of attributes related to production and clinical use. As Dr Knappett suggests, being able to do much of that in one instrument – the Zetasizer Advance – helps ensure LNP quality while reducing production time and costs.

“Organizations working in this space need to build out their characterization suite so that they can effectively analyze and understand their particles” Knappett continues. “We want to see our customers be successful and that’s why I have recommended the Zetasizer as a suitable solution for our customers, who are developing LNP encapsulated mRNA-based drug products and vaccines.”

Data analysis can also add time to a project, particularly if you need to collate data from numerous sources.  

“The data processing capabilities of the Zetasizer, post sample collection, are really powerful and allow us to do a lot,” says Knappett. “Being able to analyze data within the Zetasizer’s software is really convenient – you can directly compare data collected without needing to import it into any other software, which is just great.”
