DSC is an established analytcal tool used during process development and manufacturing of biopharmaceuticals. Rank ordering of thermal transition midpoints (TMs) and other parameters from DSC is commonly used to screen process conditions like buffers, pH and ionic strength. The typical correlation is the higher the TM, the more stable the protein is and less likely to aggregate. DSC is also used as a comparability assay to make sure different lots of the the same protein drug are "highly similar" in thermal stability.

Dynamic Light Scattering (DLS) is a mature and proven technology for nanomaterial characterization. Nanoparticle Tracking Analysis (NTA) is a more recent addition to the spectrum of particle characterization techniques and has many similarities to DLS, leading to some confusion as to their relative capabilities. The broad range of measurement capabilities for each technique is complementary in nature, providing a wealth of valuable information. The differences between the two techniques allow the data from one technology to validate and support the other.

DSC is an established analytical tool used for higher order structure (HOS) comparability studies of biopharmaceuticals, and for HOS analysis during development of biosimilars. Comparison of DSC thermograms from a reference protein with a new batch of the protein is used to assess the similarity of the conformational stability of the two proteins. Comparability studies are also performed if there is a process change, to ensure the change does not affect protein stability. DSC is also utilized as an analytical tool during the development of biosimilars, where DSC thermograms for the biosimilar are compared to the parent (innovator) drug product.

International standard ISO16332 Diesel engines — Fuel filters — Method for evaluating fuel/water separation efficiency (the US equivalent being SAE J1488) is currently undergoing revision and its publication is imminent. This new version standardizes the testing of filters between manufacturers and looks at the effectiveness of the test rig and how potential droplet recirculation could affect particle size distribution (PSD). It outlines testing of filter efficiency and gives a model particle size distribution for this. Laser diffraction is specified in the standard as the only method for sizing this distribution. This white paper examines how laser diffraction particle sizing, already an established tool in fuel filter testing, is stepping up to the challenges posed by modern fuels and the latest testing requirements. We consider the technique’s application within the context of the new methods being written for ISO16332, focusing on its implementation for measurements at high concentration, high pressure and high flow rates.

Zetium and EmpyreanMeteorites are records of the processes which occured in the early stages of the formation of the solar system. What makes them interesting is that they typically preserve the original chemistry and mineral phases over billions of years since they typically do not undergo geological transformations in space. By studying their elemental composition and mineral phases a better understanding of how the solar system was formed can be obtained. X -ray diffraction (XRD) is a common technique used to investigate phase composition of meteorites but does not provide elemental analysis. X-ray fluorescence (XRF) spectroscopy is a well-recognized technique used to determine the elemental composition of rocks.

In this final issue of X'Press for 2012 I would like you to get acquainted with some of our automation solutions for industrial applications and on-line elemental analyses.

The recent changes to the United States Pharmacopeia (USP) regarding requirements for elemental impurity testing has peaked interest into new ways to perform this testing. Combining these new requirements with the introduction of USP chapter <735> X-ray fluorescence spectrometry1 permits a new alternate method for quantifying elemental impurities in pharmaceutical materials. This study demonstrates that the X-ray Fluorescence (XRF) spectrometry is capable of performing elemental analysis of a wide range of pharmaceutical (liquid, powder and solid) materials with high sensitivity, precision and accuracy for many of the elements required by the United States and the European pharmacopeias. Simple sample preparation, non-destructive analysis, no chemical waste, low running costs with a wide dynamic concentration range and good to excellent detection limits are some of the advantages of this technique.

Sample homogeneity is an aspect that significantly affects several physical properties - strength, robustness, and lifetime of a particular product. Often, the inhomogeneity can be traced back to an error in the production process of a product. By knowing the sample homogeneity, manufacturers can trace back and optimize their process in order to create better quality products. Small spot analysis and elemental distribution mapping was conducted using a wavelength dispersive (WB) spectrometer with an energy dispersive (ED) small spot mapping (SSM) functionality. The latest advances in WDXRF instruments include the addition of an ED in the same spectrometer. The ED-SSM combination allows small spot analysis and element distribution mapping with spot sizes of 0.5 mm (FWHM) in diameter.
