Starburst Dendrimers Automated Isoelectric Point Measurements Практическая аннотация

A Zetasizer Nano ZS is demonstrated to make automated measurement of zeta potential as a function of pH for a starburst dendrimer, showing the existence of titratable amino groups at low pH. Dendrimers are densely branched, treelike, star-shaped macromolecules with potential for drug delivery, encapsulation, and gene delivery applications. The pH behavior of the effective charge of the polymer is of interest in the design for delivery complexes, and is shown here for a generation 6.5 amidoamin dendrimer with temrinal carboxylate groups.The isoelectric point was found to be at pH 5.8 suing 100mM NaOH and 100mM HCl as titrants.

Surfactant micelle characterization using dynamic light scattering Практическая аннотация

Dynamic Light Scattering is an ideal method for studying micelles as it is a non-invasive technique, and can be used to study size, CMC and aggregation number. NIBS (non-invasive back scatter) technology enables characterization with little or no sample preparation combining high sensitivity with expanded concentration range. Examples of surfactant micelle sizes (diameters) are presented: Triton X-100 at 7.5nm, Tween-20 at 8.5nm, Tween-80 at 11.7nm, Nonidet P40 at 15.4nm. The polydispersity depends on surfactant chemistry, and critical micelle concentration cmc is also specific to pH, buffer, temperature and even surfactant concentration itself. Dynamic light scattering is an ideal technique for the characterization of surfactant micelles. The results reported here illustrate the application of DLS in the study of various aspects of micelle characterization.

Using A2 and kD to assess protein interactions in formulations Практическая аннотация

Two different proteins [Immunoglobulin G IgG and human serum albumin HSA] are measured using a series of static (SLS) and dynamic (DLS) light scattering measurements. The molecular weight, 'true' hydrodynamic diameter, second virial coefficient, and DLS interaction parameter are measured.The results show that both SLS and DLS measurements can be used to study interactions between proteins, as well as simply their molecular weight and hydrodynamic size. The values of A2 and kD can be seen to change in response to a change in buffer pH.

In Quality Control (QC) procedures many pharmaceutical companies rely on microscope-based analyses to identify, qualify and or quantify solid particles meeting specified size criteria. A specification for QC analysis is agreed with the competent authority in the country where the product is to be marketed. For example the specification may limit the number of solid particles to 90% < 25 µm in length and 100% less than 100 µm in length. The method will also generally define the quantity of product to be used in the test. In practice, the sample is scanned by the analyst and an estimate of the total number of particles is made along with the percentage of solid particles meeting the specification. The samples must pass the analyses for the batch of product to be released for further processing or sale. Often such manual microscope methods involve time-consuming procedures for the analyst and can be open to human subjectivity variations from analyst to analyst. The Morphologi can automate such methods thus speeding up the analysis time, freeing-up the operators time and reducing error due to human subjectivity and/or fatigue. Additionally the system can distinguish particles according to their shape and classifications can be set up to identify the number or proportion of, for example, spherical and needle-shaped particles in a sample.

Use DSC to follow the stability of antibodies during the critical steps of manufacturing. There are multiple challenges to overcome while developing formulations for proteins intended as therapeutics. Most notably, project timelines and often limited initial availability of material drive the need for rapid, robust, and sensitive formulation development tools. Thermodynamic analysis has proven to be an invaluable approach to aid in the characterization of physical protein instabilities. However, the generated thermodynamic parameters, such as melting temperature (Tm), are typically apparent rather than absolute. Even with this caveat, apparent thermodynamic parameters are indispensable in rapidly evaluating protein instabilities. Differential Scanning Calorimetry (DSC) is a key tool in the analytical arsenal of a formulation development scientist. DSC allows for the determination of calorimetric melting temperatures and enthalpies of unfolding that permit an assessment of protein solution behavior under a variety of conditions. Several examples of pre-formulation and later-stage formulation applications of DSC, as well as their limitations and other special considerations, are discussed. These examples illustrate the crucial role of DSC and thermodynamic analysis during therapeutic protein development.

See how thermodynamics can be used to improve your lead optimization process. Renin is an aspartyl protease involved in the production of angiotensin II, a potent vasoconstrictor. Renin inhibitors can prevent blood vessel constriction and could therefore be useful for the treatment of hypertension. High throughput screening efforts identified a small molecule renin inhibitor with a core substituted diaminopyrimidine ring. Parallel medicinal chemistry efforts based on this lead resulted in compound 1. A complex of 1 bound to renin was crystallized and structural data obtained by X-ray diffraction. The structure indicated there were adjacent unoccupied binding pockets. Synthetic efforts were initiated to extend functionality into these pockets to improve affinity and adjust pharmacokinetic parameters. Thermodynamics data for inhibitor binding to renin were also collected using isothermal titration calorimetry. These data were used to help guide inhibitor optimization by suggesting molecular alterations to improve binding affinity from both a thermodynamic and structural perspective. Addition of a methoxy-propyl group extending into the S3 subpocket improved inhibitor affinity and resulted in greater binding enthalpy. Initial additions to the pyrimidine ring template that extended into the large hydrophobic S2 pocket did not improve affinity and dramatically altered the thermodynamic driving force for the binding interaction. Binding of the core template was enthalpically driven whereas binding of initial inhibitors with S2 extensions were both enthalpically and entropically driven but lost significant binding enthalpy. Additional electrostatic interactions were then incorporated in the S2 extension to improve binding enthalpy while taking advantage of the favorable entropy.

Polyol-induced increases in thermal stability of antibodies by DSC Практическая аннотация

See how DSC can be used to assess the effects of polyol excipients on antibodies. Differential scanning calorimetry was used to monitor polyol-induced increases in thermal stability across seven monoclonal antibodies. The increases in thermal stability were measured as a function of polyol concentration and orientation of hydroxyl groups. It was found that increases in thermal stability were linear with respect to alcohol concentration. Further, the increase in thermal stability was dependant on the transition temperature; antibodies with lower transition temperatures exhibited a greater increase due to the addition of polyols than antibodies with a higher transition temperature.

Learn how to use DSC to screen for physical stability of proteins in solution. The utility of microcalorimetry as a rapid screening tool for assessing the solution stability of high molecular weight pharmaceutical proteins was evaluated using model recombinant antibodies, Protein I and Protein ii. Changes in the transition midpoint, Tm, were monitored as a function of pH and/or in the presence of excipients, and results were compared with traditional accelerated stability data from samples that were analyzed by size exclusion chromatography (SEC). The data from microcalorimetry were well correlated with those from SEC for predicting both optimal solution pH as well as excipient effects on solution stability. These results indicate that microcalorimetry can be an efficient screening tool useful in identifying optimal pH conditions and excipients to stabilize pharmaceutical proteins in solution formulations.

Tack or tackiness in the context of material behavior is associated with stickiness and may result from adhesive forces between two materials in contact or cohesive forces in a material bridging two substrates. For pressure sensitive adhesives including tapes and labels, tack is defined as the ability to form an adhesive bond to a substrate under slight pressure and brief contact and is an essential requirement for such products. For other materials and applications tack may be an unwanted property, an example being bone cements, which according to ISO5833 must be tack-free, to allow the user to shape and apply the cement without adhering to gloves or application aids. Tack may also influence the behavior and perception of consumer products, examples being the extrusion of thick viscoelastic products such as toothpaste from tubes or chewing or handling of sticky foods. It may also be used to assess the properties of a surface and judge whether it is clean or not. Therefore, a qualitative assessment of tack can be made simply through touch or feel, however, such assessments are subjective, difficult to quantify and may be influenced by other additional factors. For many research and development activities it can be useful to screen, compare and quantify ‘tack’ or ‘stickiness’ using a simple objective test. For the adhesives industry there are many such standard tests available depending on the product type, including tests such as loop tack, quick stick, inverted probe and rolling ball tests. In this application note we show how a Kinexus rotational rheometer can be used to assess tack and adhesion using axial force measurements.

Liposomes have been widely used as model membranes and in drug delivery, as their basic properties can easily be varied to target desired properties (lipid composition, concentration, water content, media (different buffers, varying ionic strength, pH, etc.) as well as lamellarity (unilamellar, large or small (Large Unilamellar Vesicles, LUVs, and Small Unilamellar Vesicles, SUVs), and multilamellar vesicles, MLVs)). In these studies, one must characterize the pure liposomes as well as their mixtures with other molecules (proteins, peptides, drugs), to ascertain the influence of the added component in the liposome phase behavior. DSC is a fundamental technique for the characterization of membrane behavior, providing all thermodynamic parameters for temperature-induced transitions. With the increasing demands on water supplies and tighter regulations, much research is being done on water treatment processes. Polystyrene (PS) and poly(methyl methacrylate) (PMMA) are two of the most widely used synthetic polymers. Polystyrene is used as a protective packaging for anything from foods to CD cases and disposable cutlery, while PMMA is regularly used as a polymeric alternative to glass and even in some medical technologies, and countless other consumer products. In both cases, melting and molding are regular processes that these polymers must endure. It is therefore important to understand how they will respond to this kind of treatment. It is well recognized that the bulk properties of a polymer such as strength, toughness, flexibility, etc. are strongly dependent on molecular properties such as molecular weight and branching. If the molecular properties of these polymers change as a consequence of the processing, then it is likely that the final properties of the molded product will vary from those expected of the virgin polymer. A study was therefore undertaken to explore how these polymers respond to the stresses of molding. This application note describes the molecular and rheological changes that occurred as samples of PS and PMMA were repeatedly extruded through a capillary rheometer, simulating molding. The capillary rheometer was used to measure changes in melt viscosity, while multi-detector GPC (Gel Permeation Chromatography) was used to characterize changes in molecular weight and structure in the samples after each cycle through the rheometer.

Products, quality is often defined by a range of functional and sensory attributes, some of which may be easily measured - such as color - and others which may not. Visual or textural appearance and ease of application exemplify properties in this second category. Ultimately these attributes are assessed subjectively via user feedback, but relying on such information to guide formulation is somewhat impractical, particularly in the early stages of product development. Metrics that reliably correlate with product performance are therefore essential. Rheology adds value in product formulation and benchmarking studies by providing these product performance metrics. The resulting data can be used either to replicate the quality of an existing product, to compare formulations or competitor products, or to develop a specification for a brand new product. Rheological properties such as viscosity, yield stress and viscoelasticity can be used to determine how easy it is to squeeze toothpaste or ointment from its packaging, to spread a body lotion over skin, or the feel of skin cream when the user dips their fingers into the pot. In addition it can also be used to assess and predict product stability and help to resolve process issues In this article we look specifically at the application of rheological tests to quantify aspects of consumer product performance focusing on which tests are best for which application and the selection of appropriate test conditions.

Tracking down and halting counterfeiting operations is a substantial and complex activity and calls for highly effective analytical strategies. Morphologically-Directed Raman Spectroscopy (MDRS) is a powerful technique that combines automated particle imaging with Raman spectroscopy to provide particle size, shape and chemical identification. These capabilities make MDRS a novel and valuable tool for the investigation of counterfeit pharmaceuticals and subsequent adjudications.

The standard measurement mode for Malvern’s NanoSight Nanoparticle Tracking Analysis (NTA) technique takes place with the sample static. In that case, the only motion in the analysis is the Brownian motion of the particles. Since NTA is a single-particle technique, the statistical representation of the analysis is increased as more particles are included in the measurement. Precision and repeatability of concentration measurements are significantly improved when a slow flow is imparted on the sample to ensure that a greater number of new particles flow through the measurement zone and are analyzed during an experiment.

When producing particles of a known size, the monodispersity of the samples in terms of both size and shape is important and often requires a high-resolution technique such as TEM to look at these sample properties. For example, Particle Works, a brand of Blacktrace Holdings Ltd, produce gold nanoparticles which must be highly monodisperse and have high batch-to-batch consistency, with CV values as low as 5% for the size distribution and 2.5% for the batch consistency. Currently, Particle Works use TEM as the primary technique for characterizing their samples for QC. However, TEM is a time consuming and expensive technique that requires an experienced user to perform analysis. In this study we show how the Zetasizer Ultra can be used to identify the majority of 'out of specification' batches of gold nanoparticles and therefore significantly reduce the amount of TEM analysis required.

Gel permeation / size exclusion chromatography (GPC/SEC) is the most well-known method for polymer and macromolecular analysis. As the technique has become more pervasive, there has been a push toward greater efficiency, by way of reduced run times and minimal solvent and sample consumption. The development of Advanced Polymer Chromatography (APC™) systems has accommodated these issues, by employing low volume columns and pumps with the ability to handle higher pressure generated by these columns. These features allowed for more efficient polymer analyses, but unfortunately, the technique still relies on the need for column calibration to provide relative molecular weight values. To determine if the efficiency of the ACQUITY APC unit and the power of the advanced detection of OMNISEC REVEAL were additive, two polymer samples with different structural features were analyzed. The goal of this experiment was to obtain enough characterization data to differentiate between linear and branched polystyrene samples – all in a matter of minutes.

For virus development and production, it is important to know the virus concentration at different stages of the process, to optimize the clone used as well as the production yields. For example, instances such as clone screening, multiplicity of infection (MOI) optimization and adaptations of cell culture methods are instances when virus concentration, or virus titre as it is also commonly called, is of interest. Dynamic light scattering (DLS) is often used in virus development, in a screening function to separate good and stable samples from those with contaminants or aggregates. With the new multi-angle DLS (MADLS) based particle concentration measurement in the Zetasizer Ultra, it is now possible to get both size and size distribution as well as particle concentration per population, within a few minutes. In this application note, Allergan are kindly sharing data from their evaluation of the Zetasizer Ultra, examples of three adeno-associated virus (AAV) samples are shown. The concentration results are compared to results from capsid ELISA based virus titre assays.

Measuring the rheology of thermoplastic polymer melts Тематическая публикация

Rheology is the science of studying the flow and deformation of materials rooted in the laws of elasticity and viscosity proposed by Hooke and Newton in the late 17th Century. Thermoplastic polymer melts are widely used in many modern industrial processes to manufacture a multitude of objects. Polymers are used because they are relatively cheap to form into complex shapes in the molten state and therefore, we need to understand how they flow when being processed. Polymers are complicated materials to characterize rheologically because there are many factors that influence their flow properties, such as, processing temperature, rate of flow, residence time etc. Furthermore the rheological properties of polymers lie between those of a liquid and a solid. This leads to time dependence of the flow properties and other important characteristics. Various rheological instruments are available for characterizing the flow and viscoelastic properties of polymers. Rotational rheometers are the preferred choice when the requirement is to obtain information concerning the molecular structure and how this affects processing characteristics. In particular, the ability to easily extract information about the average molecular weight and molecular weight distribution via measurement of the viscoelastic properties makes the rotational rheometer a powerful tool. The capillary rheometer extends the shear rate range attainable in the laboratory beyond that available in a rotational instrument and allows the flow properties to be measured under typical processing conditions. In addition, the ability to readily determine both the shear and extensional properties under real life conditions provide the polymer producer and processor with information that is vital to the successful use of a polymer melt. Finally, the capillary rheometer enables processing problems to be investigated in a controlled environment without the need to stop production on the factory floor. In this white paper we discuss and compare rotational and capillary rheometry and the various tests that can be performed with each system to provide comprehensive insight into the structure, properties and processing characteristics of polymer melts.

Under current ICH Q10 guidelines, companies should be pursuing continuous improvement to their pharmaceutical products. Changing the supplier of a given excipient can be a question of quality risk management and cost. By having alternative suppliers, the company can reduce risks in the supply chain, but will need to demonstrate compatibility with the existing manufacturing process. This application note shows comparable particle morphology of an excipient from two suppliers, which supported the similar performance seen in tablet disintegration tests. The proposed alternative supplier priced the excipient significantly lower than the current supplier potentially leading to a significant reduction in cost of manufacture of the pharmaceutical product if the risk of change is found to be minimal.

Field Characterization of White Micas Using the ASD TerraSpec® Halo Тематическая публикация

The white micas are a diverse group of common fine-grained phyllosilicate minerals that include the true micas paragonite, muscovite, and phengite, as well as the K-deficient mica, illite. This group often serves as a useful exploration vector as they provide an indicator of geothermal and geochemical conditions at the time of their formation. Using the Al-OH scalar reported by the ASD TerraSpec® Halo mineral identifier it is possible to track geochemical conditions while the Illite Spectral Maturity (ISM) scalar (Doulblier et al. 2010) reported by Halo provides an indicator of thermal maturity. The white micas contained in the mineral library used by the ASD TerraSpec Halo mineral identifier are organized by composition. The true white micas are represented by the potassium white micas muscovite and phengite, and the sodic white mica paragonite. The muscovite-phengite series is characterized by increasing substitution of Mg+2 or Fe+2 and Si+4 for Al+3. The illites are classified similarly. Thus, a shift in mineralogy from muscovite and/ or illite (labeled as K-Illite in the Halo mineral library) to paragonite and/or paragonitic illite (labeled as Na-Illite in the Halo mineral library) is indicative of increasingly sodic conditions. Similarly, a shift in mineralogy to phengite and/or phengitic illite (labeled as Mg-Illite in the Halo mineral library) is usually indicative of increasingly magnesium-rich conditions. The ASD TerraSpec Halo mineral identifier is able to track geothermal gradients both by detecting mineralogy and through the reporting of the ISM scalar. With increasing metamorphic grade, smectites convert to illites, which then covert to muscovite (and other true micas). Within the illites, subtle geothermal trends are easily detected and mapped using the Halo’s ISM scalar which tracks the illite dehydration process as the illite converts to true mica.

2D WAXS measurements of polymer fibers Практическая аннотация

Here we present results of 2D wide angle X-ray scattering measurements (WAXS) obtained on two PVDF fiber series synthesized byelectrospinningfrom (a) water and (b)LimanolLB25 (15%) at a spinning speed of 195 m/s and a temperature of 120 °C that were stretched to different ratios ranging from 1.42 to 4.3. The goal was to study the effect of the applied stretching of the phase composition and the alignment of polymer chains. A total of 22 individual fiber samples were analyzed. Both series independently of used solvent behave very similarly and are hence not discussed separately during the discussion of the results.

Determine particle density with Resonant Mass Measurement Техническая аннотация

The Archimedes' Resonant Mass Measurement technique measures a particle’s buoyant mass which can be used to calculate its mass and size. This requires a value for the density of the particle being measured, as well as the density of the fluid in which it is suspended, to be known. The density of a particle can be obtained using the Archimedes instrument even if its size is unknown. This is done by measuring the buoyant mass of the particle in different fluids of varying densities. This method has been shown to be very effective at accurately measuring lower density particles such as latex. Measuring particle density allows for the accurate calculation of mass and size and can give structural information for particles which are composed of more than one material.

ω-texture analysis on metals Практическая аннотация

The preferred orientation of crystallites constituting polycrystalline solid materials such as metals, ceramics or composites significantly influences the intrinsic properties of the respective solid. In material sciences this preferred orientation is commonly known as texture and describes the orientation of all crystallites in a sample with respect to a reference frame defined by the sample orientation. Using X-ray diffraction (XRD) the texture of a sample is usually measured using the approach developed by Schulz (1949) where the intensity of certain (hkl) reflections is measured as a function of the sample orientation. This is conventionally done by rotating (ϕ) and tilting the sample in axial plane (𝜒). Hence, point focus optics as well as an Eulerian cradle are required. In contrast to the conventional Schulz-technique, in the ω-texture approach different sample tilts in the equatorial plane are accessed by applying a ω-offsett to the goniometer while keeping the sample horizontal and rotating it in ϕ (Fig. 1). Therefore ω-texture allows measurements of complete pole figures in line focus without the need for an Eulerian cradle and point focus optics.

The purpose of this project is to demonstrate that accuracy, recovery and precision criteria in standard methods such as ASTM D 51843, IP 3774 and IP 5015 can be met by using borate fusion as a dissolution method for inductively coupled plasma emission spectroscopy (ICP-OES) analysis. This preparation step will be facilitated by using LeNeo fusion instrument. A lithium metaborate with 1.5% lithium bromide integrated (LiM/1.5% LiBr) flux is used instead of the lithium tetraborate blended with 10% lithium fluoride (LiT/LiF 90/10) flux suggested in the standards. This avoids the loss of silicon ensued by the reaction with fluoride and increases the solubility of the analytes found in fuel oil.

Due to the fact that powders are affected by mineralogy and particle size effects, it is almost impossible to use a single XRF calibration curve for the analysis of such a wide range of different materials using the pressed powder preparation method. When fused in a borate glass, all mineralogy and particle size effects are eliminated. A single XRF calibration curve covering the whole range of concentrations for all elements of interests for the cement industry raw materials analysis can be made. This paper examines all the XRF analytical conditions for the calibration of the entire range of raw materials using the robust borate fusion sample preparation methodology as well as the numerous Reference Materials (RMs) used for this analytical application. The results of this general and unique XRF raw materials calibration will also be presented in terms of precision, accuracy, and limit of detection.

Crystalline phase analysis of Lithium ion battery electrode materials Практическая аннотация

Modern batteries like Li-ion have revolutionized our day to day life from smart mobile devices to pollution free electric cars and intelligent power management solutions. Batteries also hold the potential to being economical alternatives for mass energy storage to compliment renewable energy resources for power grid applications. Despite these successes, gaps in the battery technology remain in terms of safety as well as performance. Many new materials and battery chemistries are in research and development phase to enhance the energy density, discharge capacity and safety of new generation batteries. Electrodes used in Li-ion batteries have a defining influence on their electrochemical performance and are typically manufactured by coating a metal foil substrate with a multicomponent slurry made up of active electrode particles and conductive additives suspended in a binder solution. Primary particle size and crystalline phases of electrode materials play a significant role in the diffusion of Li-ions and impact key battery performance parameters such as ion transfer rate and battery recharge time. Crystalline phase can broadly be characterized in terms of phase composition and crystallite size. Phase composition (e.g. Ni concentration in NCM cathode materials) governs energy density and materials stability during cycling. So, a careful control over phase composition is needed while working on new cathode materials. Having the correct chemical composition of starting materials is not enough, though, to ensure correct phase composition of final cathode materials, as the reactants may not fuse to a single crystalline phase, mandating crystalline phase analysis of finished cathode materials. Another important parameter associated with crystalline phase, the crystallite size, represents coherent crystalline domain, which in general, may be different from the particle size as shown in Figure 1. However, in processes where primary particles nucleate and later agglomerate to form large secondary particles, a correlation may exist between the crystallite size and the primary particle size. X-ray diffraction (XRD) is routinely used technique to analyze crystalline phase properties and thus the quality of synthesized powder materials. It can measure phase purity, phase composition, and the crystallite size. Phase analysis using XRD can also be used to derive parameters like degree of graphitization or the orientation index in graphite anode materials. Case studyIn this study, five LMFP (LiMnxFe1-x(PO4)) cathode materials with Mn composition varying from 0 to 80%, and some synthetic graphite (anode) samples were investigated using X-ray diffraction for the determination of phase composition and crystallite size in the cathode, and the degree pf graphitization in anode materials. To find the correlation between crystallite size and the primary particle size, few samples were also measured using Ultra-Small Angle X-ray Scattering (USAXS) to determine the primary particle size.

Enzymes are proteins that function as biological catalysts, which play crucial roles in the biochemical processes that occur in living organisms. Understanding how enzymes function, and how to activate or inhibit their activity, is a core research focus for biochemists. Since many drug targets are enzymes, the development of new therapies requires understanding of how the target enzyme binds and catalyzes its natural substrate. Several drugs are therapeutic enzymes, which are injected into the patient to treat genetic disorders characterized by missing or defective enzymes. Enzymes are also important in other industries including food science, biofuels, and detergents. Although enzymes were discovered in the mid-1800s, isolated and purified since the early 20th century, and cloned and expressed in recombinant systems since the 1970s, there is a continued need for efficient and detailed enzyme analysis to exploit the potential of enzyme-driven catalytic reactions. New enzymes are also being discovered. Reliable enzyme kinetics data are crucial to understand and control enzyme effectiveness and create next generation drugs. Isothermal titration calorimetry (ITC) techniques have been successfully applied to study enzyme kinetics and inhibition. ITC is a well-established, versatile technique that is widely used for measuring reaction thermodynamics. In this review, we discuss how ITC generates real-time, enzyme kinetics data, comparable to other enzyme assays. Relating zeta potential to jar testing Тематическая публикация

Jar testing simulates the coagulation / flocculation process in a water treatment plant. In a set of beakers, a defined amount of raw plant water is mixed with varying amounts of coagulant. Each beaker receives an increasing incremental dose of coagulant and is then stirred for a set amount of time to mimic plant operation. The formation of floc during this test simulates plant operation. Observations from the floc formation indicate a preferred range of coagulant dosage. Thus, jar testing can help identify the parameters that promote an appropriate final water quality. While this jar test is simple to perform, it does require time, typically about 2-3 hours, including preparation and wait for settlement. Since water quality can change seasonally (summer algae growth, flood events, forest fires) the jar test has to be repeated regularly. A significantly faster method is the measurement of zeta potential expressed in milli Volt (mV). Here, the water is exposed to an electric and scientifically supported field, and the movement of colloidal particles indicates their net charge under the specific water conditions. We present details of the zeta potential method and compare results with jar testing. At the example of alum (Aluminium Sulfate, Al2(SO4)3) as coagulant, we show that the optimal dose is related to a favorable zeta potential range of -8mV to +3mV. ​ Not shown here, the use of ferric (Iron Chloride or Iron Sulfate) typically correlates with the same zeta potential range to optimize water treatment plant operation. Each coagulant chemistry has an optimal pH range where it is most effective, so it is also important to monitor pH in the plant and maintain the similar pH during the jar testing.

The 1Der for crystallography studies Практическая аннотация

Fluorescence is commonly a problem in mineralogical phases like Hematite- Eskolaite, because these minerals contain the strongly fluorescing Cr and Fe ions. This data sheet shows how the 1Der detector excludes both Cr and Fe fluorescence. Empyrean’s unique optical configuration provides truly clean data leading to an excellent Rietveld refinement and resulting in an accurate crystal structure determination of this natural mineral. For X-ray diffraction (XRD) measurements on the Empyrean system, it is beneficial to be able to choose an X-ray tube to suit the requirements of a material. Standard Empyrean tubes offer a choice of Cr, Mn, Fe, Co, Cu, Mo and Ag, and other anode emission energies can be provided on request. Cu is the most commonly used anode and the Cu Kα emission peak is the most commonly used energy for diffraction experiments. In an X-ray diffraction experiment, the optics and the detector are together tuned to the narrow energy window required by the experiment. The 1Der strip detector can be automatically tuned to provide a narrow energy window around any chosen diffraction energy. This provides full flexibility across the whole range of Empyrean 1D applications. All elements fluoresce when irradiated with a certain X-ray energy. In a diffraction measurement, fluorescence from some elements can give rise to unwanted background intensity in a diffraction pattern. A high background reduces the possibility of identifying the low intensity peaks that often provide the final clues in a Rietveld crystal structure refinement. Figure 1 illustrates the elements that emit the greatest fluorescence interference in a diffraction measurement using a Cu X-ray tube. This application note describes the texture analysis of a rolled copper specimen. The experimental data were measured on a multi-purpose X-ray diffraction system equipped with a texture cradle. In polycrystalline solid materials, such as a wide class of metals and ceramics, usually the orientation of the crystallites, constituting the matrial, is not distributed randomly, like in an ideal powder specimen. In most cases a preferred orientation of the crystallites with respect to the sample reference frame is present. In materials science this preferred orientation is referred to as texture.

Measurement of Soil Mineralogy and Total Carbon Using NIR Практическая аннотация

Analysis of metals in pharmaceuticals with Epsilon 3XLE Практическая аннотация

Epsilon 3XLE is capable of determining metal concentrations in pharmaceutical materials, with minimal sample preparation. The repeatability results of this study illustrate the stability and robustness of the Epsilon 3XLE. The calculated detection limits (LLD) and analysis accuracy (RMS) clearly demonstrate that the Epsilon 3XLE can reliably measure most elements of concern at the 1 to 3 μg/g (ppm) level. The performance demonstrated in this work can be reproduced with either the Rh or Ag X-ray tube configurations. Furthermore, the compact size and low weight of the spectrometer make it an ideal tool for the pharmaceutical industry. Examples of use include residual catalysts investigations in API’s, raw material screening and regulatory reporting of finished products.

Visible/Infrared Reflectance Spectroscopy (VIRS) measures absorption in the 350 nm to 2500 nm region of the electromagnetic spectrum, where many minerals have distinct responses that allow their identification, even within mixed parageneses. The VIRS method characterizes a range of hydrothermal and supergene alteration minerals, and also some primary igneous and metamorphic minerals. Data acquisition is simple and rapid, but interpretation is more complex, requiring a large database of reference spectra. Automated computer programs can simplify and expedite this process, but human analysis and reasoning are also required, and VIRS data are most useful where combined with other data, notably petrography and geochemistry. Several pilot studies, summarized here, illustrate the versatility of the VIRS method. Studies of alteration, associated with epithermal gold mineralization, easily recognized key indicator minerals such as pyrophyllite, alunite, dickite and topaz; such information may be useful in mapping zoned alteration systems, and vectoring toward the most favourable areas. A study of footwall alteration in a VMS deposit did not support previous visual identifications of pyrophyllite, but gave ambiguous results with respect to kaolinite. Modelling showed that subordinate kaolinite could be masked by interference from more abundant white mica (sericite). The VIRS data from a mesothermal vein-type gold deposit indicate subtle alteration signatures, on a scale of several metres, around auriferous veins, even though visual evidence for such effects is muted. The mineralogical causes for this effect are as yet unresolved, but this does not prevent its use as a proximity indicator in exploration. Studies of porphyry-style Mo–Cu deposits, associated with sheeted veins, document the progressive overprinting of regional chloritedominated propylitic alteration by focused potassic to phyllic alteration associated with the mineralization. A later stage of advanced argillic alteration, typified by kaolinite, was also documented in one deposit. A reconnaissance investigation of rareearth element (REE) mineralization in Labrador suggests potential for the VIRS method in studies of host rocks and mineralization. The VIRS data can distinguish Na–Fe-rich chain silicates characteristic of peralkaline igneous rocks from common pyroxenes and amphiboles, facilitating identification of potential host suites. The REE also generate unusual absorption features in the visible and near infrared, which directly indicate specific REE ions. Although there are few reference spectra for complex REE-bearing minerals, the VIRS data recognize mineralized samples, and may help to identify samples collected on the basis of other criteria, for geochemical assays. Finally, VIRS analysis of samples from central Labrador now provides the first reference spectra for the REE-enriched mineral eudialyte.

In this application note we will show a robust calibration for the analysis of solid alternative fuel materials with Epsilon 5 Alternative Fuels using a series of CRMs with different compositions. To indicate the applicability of our approach, we have analyzed plant CRMs as routine samples against our calibrations and compared certified vs. measured values. Alternative fuels commonly originate from widely varying sources and tend to be heterogeneous in terms of both material type (e.g. oil, plastic, biomass) and chemical composition. For XRF this is less of a problem than for other analytical techniques.

The power of reflectance spectroscopy as an analytical tool for mineralogical studies has been known since the first half of the 20th century. However, it took several decades until this technology was widely adopted by the mineral exploration community as an operational tool, either in the laboratory or in the field. As a result, the identification of minerals associated with ore deposits using reflectance spectra has become one of the major applications of the technology in recent years. This paper summarizes some of the studies developed at the Institute of Geosciences, University of Campinas (Brazil), in which reflectance spectroscopy in the visible, near-infrared and shortwave infrared played a key role in unveiling subtle mineralogical information related to different types/styles of mineral deposits. The applications to be reviewed include the characterization of the hydrothermal alteration mineralogy in a gold-bearing epithermal deposit in Patagonia, Argentina, in a porphyry copper deposit in Chile, and in pegmatites of the Borborema Pegmatitic Province in northeastern Brazil.

Environmental monitoring over land surfaces has various complexities caused by highly heterogeneous surface structures with vegetation layer, soil surfaces, and atmospheric disturbance. The vegetation biophysical parameters must be provided with sufficient accuracy to be used as an input of general circulation models, where the satellite remote sensing also plays an important role. Satellite remote sensing is the use of vegetation index (VI) as an intermediate variable to quantify status and amount of vegetation from satellite images. Various VI products currently exist and more VI products are expected to be available simultaneously, the estimation of biophysical parameters through VI has the be compatible by various sensors to ensure continuity of global environmental simulation over generations. Therefore, it is important to investigate the compatibility of VI products for new sensors with existing and old satellite sensors every time a new satellite will be launched. A soon to be launched optical sensor on board Japanese ADEOS-II platform, named GLobal Imager (GLI), is the focus of this study.

Operational impacts of Glen Canyon Dam on downstream resources of the Colorado River ecosystem have been the subject of intense study and discussions among adaptive management stakeholders that include Federal, State and tribal entities, as well as recreational businesses, environmentalists, power companies, and others ( U.S. Bureau of Reclamation 1995, Schmidt et al. 1998). The Grand Canyon Monitoring and Research Center (GCMRC within the U.S. Department of Interior) has identified the following key objectives for long-term monitoring of vital components of the ecosystem between Lakes Powell and Mead, while providing stakeholders with information for decision making: 1) lowest possible impact on critical resources; 2) highest possible return of integrated scientific information; and 3) long-term, repeatable data collection. Airborne hyperspectral image data are being considered as a possible long-term monitoring tool, which hold the promise of being able to fulfill the above requirements. The present study is a pilot project which is intended to explore the extent and variety of information, relevant for long-term monitoring, that can be extracted from images of both high spectral and spatial resolution. The Colorado River ecosystem in Glen Canyon and Grand Canyon poses special challenges for remote monitoring with respect to logistics required to support field-based studies and relative to spatial scales at which monitoring is needed. The narrow corridor of the river is confined by tall vertical cliffs, yet provides a habitat for a great variety of both native and non-native plant species. Riparian vegetation consists of stands that often cover only very small areas. Often, these stands consist of intermixed native and non-native species, and spectral signatures of the various plants sometimes display only subtle differences. Adequate change detection mapping requires both high spectral and high spatial resolution, as well as advanced processing algorithms that can detect subtle spectral differences. Change detection for evolving sand bars, rapids and debris fans is a critical part of the GCMRC's long-term monitoring program. Additionally, changes in the aquatic ecosystem's food base, such as abundance and distribution of algae and other benthic organisms are of great interest. Presently, the need to lower river stage for 2- 3 days during annual monitoring overflights for conventional aerial photography and traditional surface measurements introduces artificial and undesirable impacts on the ecosystem (e.g., Blinn et al. 1999). With hyperspectral imagery, we expect to map riparian vegetation, terrestrial sediment deposits, as well as sand bars and benthic organisms through some depth of water, obtaining greater compositional information than can be obtained through the use of aerial photography.

Spectral Signatures of Nighttime Lights Практическая аннотация

For the past fifteen years NGDC has provided satellite derived maps of the spatial distribution and brightness nighttime lights for environmental, energy, and economic applications. The consistent response of the nighttime lights user community is that they want higher spatial resolution and an ability to discriminate lighting types. We recently completed a high spectral resolution survey of the emission characteristics of a wide range of lighting types (incandescents, fluorescents, metal halides, high pressure sodiums, low pressure sodium, LEDs and others). The laboratory spectra have been used to guide the classification of lighting types observed in airborne hyperspectral imagery. The results are being used in the design of a Nightsat sensor.

Analyzing powder diffraction data of nanocrystalline and amorphous materials using the atomic pair distribution function (PDF) method, provides useful information about the long- and short-range ordering of the atoms in the materials. Recent years have shown an increased interest in the study of nanocrystalline materials due to their specific properties for application in e.g. semiconductors, pharmaceuticals and polymers.

Small Sample Size Evaluation using LabSpec 4 Standard-Res i Практическая аннотация

Many analytic applications benefit from the ability to measure very small samples non-destructively. Typical applications of near-infrared analysis require sample sizes of 5 to 50 grams. The purpose of this evaluation was to determine if sub-gram quantities of sample could be used with the ASD LabSpec 4 Standard-Res i. A set of dried ground corn stover samples was used for this study to evaluate this small sample size capability. ASD’s spectrometer system was used with very small samples in micro-centrifuge tubes to allow higher throughput in the lab thus reducing costs associated with the NIR analysis.

X-ray micro-diffraction in wall painting research Практическая аннотация

A study by the Academic Laboratory of Materials Research of Paintings (ALMA) in Prague, Czech Republic has shown that X-ray powder micro-diffraction can be used for the investigation of salt damage. The use of X-ray microdiffraction enabled the research team to identify the probable causes of damage to an ancient fresco. Pigment degradation, often resulting in color changes, causes serious damage to wall paintings. Many factors lead to pigment degradation including sunlight, air pollution, salts and microorganisms. Of these, salts are one of the most damaging agents, especially for artworks of a porous nature such as wall paintings.

Field based spectroscopy for ecological and environmental monitoring has become an indispensable part of complete observational systems as defined in GEOSS (Global Earth Observation System of Systems). Spectral scaling approaches are currently ranging from molecular to ecosystem or even biome scales. We discuss the use of field spectroscopy in relation to supporting large-scale ecosystem and ecotone inventorying, in particular the retrieval of biochemical and structural attributes of vegetation. First, attention will be put on using an object-relational database for the structured compilation of field spectral measurements and corresponding metadata. Spectral libraries have been collected over a wide variety of natural and man-made targets and their sampling scheme is discussed. Second, recent advances in reflectance and radiance terminology for field spectrometers are discussed, propagating the use of spectral albedo. Advanced measurement instrumentation and facilities are presented that complement solar reflective measurements with the angular, thermal and plant fluorescence domains. Further, experiments measuring leaf albedo, transmittance and absorbtance are discussed using examples from a wide range of ecosystems and their ecotones. Finally, we discuss scaling approaches from field measurements to airborne and spaceborne mapping methods, demonstrating the wide use of field spectrometers for environmental applications. We conclude on the importance of using field spectrometers over the past one and a half decades at our institutions.

In this paper we present how SAXS measurements can be performed on a multi-purpose X-ray diffractometer platform. Application examples for nanopowders, colloids, nanocomposites and porous materials will be given. A multi-purpose X-ray diffractometer platform was configured for small-angle X-ray scattering measurements. This technique was applied to nanoparticle and pore size distribution determination in nanopowders, colloidal dispersions, nanocomposites and porous materials in a range of 1-100 nm.

There has been growing interest in the use of diffuse infrared reflectance as a quick, inexpensive tool for soil characterization. In studies reported to date, calibration and validation samples have been collected at either a local or regional scale. For this study, we selected 3768 samples from all 50 U.S. states and two tropical territories and an additional 416 samples from 36 different countries in Africa (125), Asia (104), the Americas (75) and Europe (112). The samples were selected from the National Soil Survey Center archives in Lincoln, NE, USA, with only one sample per pedon and a weighted random sampling to maximize compositional diversity. Applying visible and near-infrared (VNIR) diffuse reflectance spectroscopy (DRS) to air-dry soil (< 2 mm) with auxiliary predictors including sand content or pH, we obtained validation root mean squared deviation (RMSD) estimates of 54 g kg− 1 for clay, 7.9 g kg− 1 for soil organic C (SOC), 5.6 g kg− 1 for inorganic C (IC), 8.9 g kg− 1 for dithionate–citrate extractable Fe (FEd), and 5.5 cmolc kg− 1 for cation exchange capacity (CEC) with NH4 at pH = 7. For all of these properties, boosted regression trees (BRT) outperformed PLS regression, suggesting that this might be a preferred method for VNIR-DRS soil characterization. Using BRT, we were also able to predict ordinal clay mineralogy levels for montmorillonite and kaolinite, with 88% and 96%, respectively, falling within one ordinal unit of reference X-ray diffraction (XRD) values (0–5 on ordinal scale). Given the amount of information obtained in this study with not, vert, similar4 × 103 samples, we anticipate that calibrations sufficient for many applications might be obtained with large but obtainable soil-spectral libraries (perhaps 104–105 samples). The use of auxiliary predictors (potentially from complementary sensors), supplemental local calibration samples and theoretical spectroscopy all have the potential to improve predictions. Our findings suggest that VNIR soil characterization has the potential to replace or augment standard soil characterization techniques where rapid and inexpensive analysis is required.

Growing concerns regarding the impact of global carbon emissions have led to rigorous studies to find effective methods and processes to sequester atmospheric carbon back into a terrestrial phase of the carbon cycle. A number of innovative mechanical sequestration solutions have been developed including geologic sub surface and deep ocean storage, while research into passive natural systems have led to a better understanding of the potential for certain soils to act as carbon sinks. Soil Organic Carbon (SOC) sequestration, in which carbon dioxide present in the atmosphere is transferred into the soil through crop residues and other organic solids is not only an important source of carbon in the soil but also provides an off-set from fossil-fuel combustion and other carbon emitting activities (OSU, SOC Factsheet). Understanding the mechanics of SOC is important for determining soil health, productivity, and developing land management strategies, as well as carbon dioxide fluxes in the atmosphere. Accurately measuring current soil organic carbon (SOC) concentrations, understanding the contributing factors, and being able to assess a given soil’s potential to sequester additional carbon (Iowa EPSCoR) are at the core of understanding these dynamics. SOC sequestration potential is defined as the maximum possible storage of Total Below-Ground Carbon Allocation (TBCA) under a specific soil-climate-land management regime. Traditional lab-based methods for determining TBCA are costly, time-consuming and tend to destroy the sample in which it is measured (Iowa EPSCoR). Near-infrared (NIR) reflectance spectroscopy provides an efficient cost-effective alternative to traditional lab-based SOC analysis. With NIR reflectance analysis, rapid non-destructive measurements can be taken in the field or in a controlled laboratory environment. Quantitative calibration models can be developed for rapid characterization of soil nutrients and other physical properties. Coupling this technology with hyperspectral imagery and improved spatial statistical methodologies breaks the bottleneck of sample collection and lab analysis and facilitates large-area soil characterization assessments. The goal of this evaluation was to create a calibration model for percent SOC using near-infrared spectra of soil samples collected on the ASD LabSpec 2500 by Kenneth Wacha at the University of Iowa.

Mineral physics dictates the appearance of rocks and soils across the electromagnetic spectrum. In the Visible/Near-Infrared (VNIR) and Short Wave Infrared (SWIR), many materials absorb radiation at specific wavelengths, allowing their identification by the position and character of absorption features. Electronic processes at wavelengths less ~1.0 micrometers allow identification of minerals containing Fe+3. Molecular vibrational features at wavelengths between ~1.0 and 2.5 micrometers are diagnostic of minerals containing anion groups such as Al-OH, Mg-OH, Fe-OH, Si-OH, CO3, NH4, and SO4. Small differences in absorption band position and shape are correlated with mineral compositional differences and variability. Imaging spectrometry has been used since the early 1980s to perform 2-dimensional mapping of mineral distribution based on spectroscopic characteristics. Geologic applications include areas such as lithologic mapping; exploration for precious and base metals; and oil, gas, and geothermal energy exploration. Field spectroscopy plays a critical role in the calibration, analysis, and validation of imaging spectrometer data. Spectral libraries have been measured for a variety of minerals. Imaging spectrometer datasets have been acquired around the world using airborne platforms and recent satellite systems provide spectral measurements for selected areas. Case histories will be presented demonstrating the link between laboratory, field, and imaging spectrometer data. Selected physics-based analysis methodologies will be discussed and the level of information available from the data will be demonstrated. Examples will include mineral identification and mapping in the context of hydrothermal alteration associated with active and fossil hot springs and mineral deposits. Specific mineral mapping examples will include hematite, goethite, and jarosite in the VNIR and identification and separation of calcite, dolomite, and calc-silicates along with phyllosilicates and sulfate minerals such as alunite and jaorsite using SWIR spectroscopy. Spectral variability caused by processes such as anion substitution in illite/muscovite, crystallinity in kaolinite-group minerals, and spectral mixing will also be discussed.

Leafy spurge (Euphorbia esula L.) is a noxious invasive weed that infests over 1.2 million hectares of land in North America. One of the fundamental needs in leafy spurge management is cost-effective, large-scale, and long-term monitoring of plant populations. Leafy spurge has distinctive yellow-green flower bracts which are spectrally unique when compared to co-occurring green vegetation; the spectral signature is caused by a reduction of chlorophyll allowing various carotenoids to influence the reflectance spectrum. In 1999, Airborne VisibleInfrared Imaging Spectrometer (AVIRIS) data were acquired over Devils Tower National Monument in Northeastern Wyoming. Accuracy of Spectral Angle Mapper classification for flowering leafy spurge was highest using a threshold of 3.5°, which is consistent with both reflectance spectroscopy data and simulations using a canopy radiative transfer model. The minimum detectable cover of flower bracts is about 10%, which is about 35% total cover of leafy spurge, and is ideal for biological control with flea beetles (Aphthona spp.).

The X’Pert3 MRD X-ray diffraction system is a versatile tool for studying residual lattice strain in solar cell structures grown on relaxed buffer layers. Offcut structures in the form of single crystal AlGaAs/GaAs multilayers on GaAs and Ge substrate wafers and on Ge-on-Si pseudo-substrates were analyzed. The physical properties of a single crystal solar energy collecting device are dependent upon the unit cell and thin film structure of the layers that form the device. During growth, the thin film unit cell is constrained to an in-plane lattice parameter provided by the substrate.

This application note shows that the Semyos micro X-ray fluorescence spectrometer is easily capable of analyzing CoNiFe films used in the manufacturing of read/write heads for hard drives. Since the last decade, the read/write head technology used in hard drives has been advancing at an incredible pace, resulting in formidable information densities that are currently achieved for state-of-the-art hard drives. This paper present the results of a preliminary study to assess the potential of the visible, NIR and SWIR energy of the EMR in differing iron ores of different grades in a rapid manner using hyperspectral radiometry. Using different iron ore samples from Noamundi mines, Jharkhand, India, certain spectro-radiometric measurements and geochemical analysis were carried out and the results have been presented. It was observed that the primary spectral characteristics of iron ores lie in the 850 to 900nm region. Various methods to evaluate the parameters for each curve such as: the radius of curvature of the maximum absorption trough, position of the trough with respect to wavelength in the NIR region and distance of the trough from a reference line, were adopted. Comparison of the spectral parameters and the geochemistry of the samples indicates that the depth of absorption (distance of trough from a reference line) in the NIR region for a sample has a strong positive correlation with the iron oxide content in the sample, while the radius of curvature of the absorption trough has a strong negative correlation with the iron oxide content. Such strong correlations indicate that hyperspectral radiometry in the visible and NIR regions can give a better estimate of the grade of iron ore. This study has demonstrated that generation of empirical models using hyperspectral radiometric techniques is helpful to quantify the grade of iron ores with limited geochemical analysis.

An algorithm based on the physics of radiative transfer in vegetation canopies for the retrieval of vegetation green leaf area index (LAI) and fraction of absorbed photosynthetically active radiation (FPAR) from surface reflectances was developed and implemented for operational processing prior to the launch of the moderate resolution imaging spectroradiometer (MODIS) aboard the TERRA platform in December of 1999. The performance of the algorithm has been extensively tested in prototyping activities prior to operational production. Considerable attention was paid to characterizing the quality of the product and this information is available to the users as quality assessment (QA) accompanying the product. The MODIS LAI/FPAR product has been operationally produced from day one of science data processing from MODIS and is available free of charge to the users from the Earth Resources Observation System (EROS) Data Center Distributed Active Archive Center. Current and planned validation activities are aimed at evaluating the product at several field sites representative of the six structural biomes. Example results illustrating the physics and performance of the algorithm are presented together with initial QA and validation results. Potential users of the product are advised of the provisional nature of the product in view of changes to calibration, geolocation, cloud screening, atmospheric correction and ongoing validation activities.

Astronomers began the use of multi-wavelength studies of Solar System objects in the solar spectrum region, first by eyeballing the objects’ colors, such as for Mars, and quantitatively when the photographic process was developed in the last half of the 19th century. After the time of Galileo, astronomers came to emphasize gas and plasma studies, while solar system studies included dirt and rocks as subjects. I am more the dirt and rocks type and I will emphasize this history. I arrived on the scene in the 1960s when electronic detectors first began to trickle out to civilian scientists. These allowed far more accurate quantitative measurements, as well as measurements at infrared wavelengths. We, who had access to these new detectors, began using the astronomers’ telescopes to observe solar system objects almost at random during the nights we could get precious telescope time. My own early emphasis was to demonstrate that there are spectral expressions of rock-forming minerals (basalts) on the Moon. This seems primitive by today’s standards, but it is where we were in the1960s. Solar System studies grew from there, first with telescopes and then using spacecraft to carry our spectrometers to the objects. Spacecraft history developed slowly due to the dominance of geologists and picture taking over geochemists and the less visual spectroscopy. The delay of the launch of the Galileo spacecraft, due to the Challenger disaster, the first deep space mission to carry a visual-infrared imaging spectrometer, was a further setback. But we (and our graduate students) eventually succeeded at this. Today, we still find the telescope is an important tool but, nearly every spacecraft to Solar System object carries imaging spectrometers operating in the UV-visual-IR spectral region, where reflected solar radiation and emitted thermal radiation is detectable.

Brilliant Blue is often used to trace water movement in soils. Its concentration in soil samples is usually determined by extraction – a laborious procedure with varying accuracy. We show that Brilliant Blue can be estimated directly by visible diffuse reflectance spectroscopy. We build a PLSR (partial least squares regression) model for the concentration range of 0.1 to 15 mg Brilliant Blue per g soil with an RMSE (root mean square error of prediction) of 1 mg g-1 and an R2 adj of 0.9. However, as the method is based on visible spectra, prediction accuracy can be altered by variations in soil colour between calibration and prediction data sets.

EnMAP – An Advanced Optical Payload for Earth Observation Практическая аннотация

The potential of imaging spectroscopy to provide more and better information about the Earth system than traditional multispectral instruments is currently not counterbalanced by an equivalent availability of space-borne spectroscopy data. The Environmental Mapping and Analysis Program (EnMAP), a German hyperspectral mission scheduled for launch in 2013, is intended to cover this gap. The primary goal of EnMAP is to offer accurate, diagnostic information on the state and evolution of terrestrial ecosystems on a timely and frequent basis, and to allow for a detailed analysis of surface parameters with regard to the characterisation of vegetation canopies, rock/soil targets and coastal waters on a global scale. EnMAP is designed to record bio-physical, bio-chemical and geo-chemical variables to increase our understanding of biospheric/geospheric processes and to ensure the sustainability of our resources. EnMAP will sample areas of 30 x 30 km2 with a ground sampling distance (GSD) of 30 m, measuring in the 420-2450 nm range by means of two separate spectrometers covering the visible to near-infrared (VNIR) and short-wave infrared (SWIR) spectral regions. The mean spectral sampling distance and resolution is of 6.5 nm at the VNIR, and of 10 nm at the SWIR. Accurate radiometric and spectral responses are guaranteed by a required signal-to-noise ratio (SNR) of about 500:1 in the VNIR and about 150:1 in the SWIR, a radiometric calibration accuracy better than 5% and a spectral calibration uncertainty of 0.5 in the VNIR and 1 nm in the SWIR. An off-nadir pointing capability of up to 30° enables a target revisit time of 4 days. Recent scientific activities have been focused on the development of a forward scene simulator to support hardware developments and the consolidation of the mission concept.
