Use DSC to select antibody with lowest propensity to aggregate. An important consideration during the engineering process of an antibody is both the inherent conformational stability of the antibody as well as long-term stability. Understanding these stability parameters and what trends they follow will be critical throughout the biotherapeutic development. Differential scanning calorimetry (DSC) is a potent tool in probing these qualities. As a stability indicating assay, DSC provides insights for the selection of stable protein construct candidates. In the case of multi-domain proteins, DSC uniquely allows the investigation of the stability of individual domains from multi-domain proteins like antibodies and Fc-conjugated proteins. In this application note, we compare stability data predicted by DSC of different engineered antibodies with SEC-HPLC data from accelerated stability studies. For the engineered proteins studied, there is a correlation between decreased thermostabilty based on DSC data, and greater aggregation formation during accelerated stability studies. This study demonstrates how DSC can be a useful tool in stability screening of engineered proteins.

Battery manufacture relies on the electrode materials being applied as a slurry, to form a film. The size and shape of particles within the slurry are critical to the production of the film and its final uniformity, via the viscosity of the slurry. In this application note, the Morphologi automated image analysis system is used to characterize efficiently the shape of particles in high and low viscosity electrode slurries. IntroductionBatteries are ubiquitous in modern life and our reliance on them has never been greater. Therefore, ensuring optimum battery performance through manufacturing control is of increasing significance. In previous application notes we have discussed the importance of controlling the size of particles used in the manufacture of battery materials [1] and the impact of carbon microstructure in graphite electrodes on battery performance [2]. Shape is also an important factor to consider and control, as irregular shaped particles not only reduce packing density, but they can lead to the formation of a high viscosity electrode slurry. In this third application note on batteries, we consider the role of size and shape on the viscosity of the electrode slurry. Electrode compositionThe typical structure of a battery electrode is given in Figure 1. The electrode is usually fabricated by applying a slurry of particles in suspension onto a metal foil. Figure 1 – typical structure of the Li-ion battery. The slurry in question is composed of electrode particles (anode or cathode), small carbon particles to aid conduction, and binder material (composed of solvent and polymer) to hold the structure together. The concentration of particles in the slurry is high, representing between 20-40 % of the total by weight. Consequently, the particle properties have a significant impact on the physical properties of the resultant slurry. Particle properties and slurry viscosityThe viscosity, dispersibility, concentration and compactability of the slurry are important parameters in determining how effective the slurry will be during application. A high viscosity slurry causes difficulties in the coating process, and poor dispersibility results in low film uniformity; the concentration and compactability of the slurry controls the film density. Uniformity of the coating thickness and the layer density are important to ensure control over the ion transfer rate and life time (recharge cycle time) of the battery, while controlling the layer thickness enables a smaller battery to be produced. As illustrated in Figure 2, the presence of a significant proportion of irregularly shaped particles will lead to a higher viscosity slurry due to the increased impact of particle friction and interlocking but also due to the extra flow energy required for the fluid to circumvent the particles. Figure 2 – irregularly shaped particles experience greater interlocking and friction, leading to a higher viscosity. Particle shape also impacts on packing density since irregular particles pack less efficiently than spheres. Hence, fewer particles can be added to the liquid before the viscosity starts to increase, as illustrated in Figure 3. Further, a polydisperse sample will pack more efficiently than a monodisperse sample, at the same concentration, lowering viscosity. Smaller irregular particles, however, may increase viscosity due to their higher surface area, which will accentuate particle-particle and particle-liquid interactions. Therefore, it is important to be able to monitor and control the proportion of irregularly shaped particles and fine material within an electrode material sample to minimize the viscosity. Quantification of the M1 and M3 polymorphs of alite in OPC clinker Практическая аннотация

This data sheet demonstrates that the M1 and M3 polymorphs in clinker can be quantified individually using the Rietveld method. The repeatability of the method is such that it allows accurate process control of cement production.. The main component in industrial clinkers for the cement production is alite with the ideal composition Ca3SiO5, also written as C3S in the short cement notation. This compound can occur in crystallographic polymorphs. Their formation depends mainly on the burning conditions (temperature, burning time, cooling rates) and minor or trace elements in the crystal structure, introduced by the raw material and fuels. Most relevant in clinkers are two monoclinic polymorphs of alite, the M1 and M3 forms. In recent years the quantification of these two polymorphs has become of more interest because of publications claiming differences in the reactivity of synthetic M1 and M3 alite. .

Products, quality is often defined by a range of functional and sensory attributes, some of which may be easily measured - such as color - and others which may not. Visual or textural appearance and ease of application exemplify properties in this second category. Ultimately these attributes are assessed subjectively via user feedback, but relying on such information to guide formulation is somewhat impractical, particularly in the early stages of product development. Metrics that reliably correlate with product performance are therefore essential.. Rheology adds value in product formulation and benchmarking studies by providing these product performance metrics. The resulting data can be used either to replicate the quality of an existing product, to compare formulations or competitor products, or to develop a specification for a brand new product. Rheological properties such as viscosity, yield stress and viscoelasticity can be used to determine how easy it is to squeeze toothpaste or ointment from its packaging, to spread a body lotion over skin, or the feel of skin cream when the user dips their fingers into the pot. In addition it can also be used to assess and predict product stability and help to resolve process issues. In this article we look specifically at the application of rheological tests to quantify aspects of consumer product performance focusing on which tests are best for which application and the selection of appropriate test conditions..

Differential scanning calorimetry (DSC) is an established biophysical tool for the characterization of protein stability . A protein's stability is a critical quality attribute (CQA) which is characterized throughout biopharmaceutical drug discovery and development, to help decide if it should be chosen to advance in the pipeline, and also to help guide selection of the process and formulation conditions needed to maintain its stability. In a DSC system, the protein in solution is placed in the DSC cell and heated at a constant rate, resulting in the protein unfolding due to thermal denaturation. The protein’s heat capacity (Cp) changes as the protein denatures. The peak of a DSC thermogram represents the protein's thermal transition midpoint (TM). For a protein which reversibly denatures, TM is the temperature where 50% of the protein is in its native (folded) conformation, and 50% is in its denatured (unfolded) conformation. TM is considered a good indication of thermal stability – the higher the TM, the more thermally stable the protein. Even if a protein is irreversibly denatured during a DSC experiment, the TM still provides important insights into its thermal stability. Multi-domain proteins (like antibodies) typically have more than one peak on a DSC thermogram, so more than one TM can be determined. The first automated DSC system for biopharmaceutical applications, MicroCal VP-Capillary DSC, was launched in 2002. With automation, DSC quickly performs TM screening for biopharmaceutical preformulation and formulation development. Data from MicroCal VP-Capillary DSC systems are used to evaluate protein drug products for: candidate selection/protein engineering; process development; higher order structure (HOS) characterization during comparability studies and manufacturing support; HOS characterization for biosimilar development. In 2017, Malvern Instruments launched the MicroCal PEAQ-DSC Automated system. Compared to the MicroCal VP-Capillary DSC, the MicroCal PEAQ-DSC Automated system has new features and improvements designed to improve productivity during biopharmaceutical discovery and development, generating high-quality, reproducible, and reliable DSC data which can be used for regulatory submissions. MicroCal PEAQ-DSC Automated systems are essential additions to the 'biophysical toolbox' in biopharma, CROs and CDMOs, and core facilities at academic and government research laboratories.

Introduction This white paper outlines the main benefits of using hard radiation for material analysis studies. X-ray powder diffractometers are generally equipped with X-ray sources using Cu anodes. For specific applications, however, switching to shorter wavelength, ‘harder’ radiation, like obtained from X-ray tubes with Mo or Ag anodes, gives significant improvements over Cu, or even allows experiments that are impossible with Cu radiation, like experiments where a large Q range needs to be probed. Often, people make use of synchrotron radiation, but new developments on source, optics and detector technologies allow a variety of hard radiation experiments in the home lab. In this white paper we present six key advantages of hard radiation studies. Larger penetration depth For inorganic materials, the penetration depth of Cu K-alpha is often only a few micrometers (see Table 1). In order to extract information from a larger sample volume, hard radiation is an advantage in transmission experiments. Thanks to the larger penetration depth, it is possible to observe crystallographic changes in working devices, such as Li-ion batteries, and perform highpressure experiments with diamond anvil cells. Hard radiation allows using transmission geometry for inorganic samples. Table 1. Sample homogeneity is an aspect that significantly affects several physical properties - strength, robustness, and lifetime of a particular product. Often, the inhomogeneity can be traced back to an error in the production process of a product. By knowing the sample homogeneity, manufacturers can trace back and optimize their process in order to create better quality products. Small spot analysis and elemental distribution mapping was conducted using a wavelength dispersive (WB) spectrometer with an energy dispersive (ED) small spot mapping (SSM) functionality. The latest advances in WDXRF instruments include the addition of an ED in the same spectrometer. The ED-SSM combination allows small spot analysis and element distribution mapping with spot sizes of 0.5 mm (FWHM) in diameter. Biopolymers are widely used in the pharmaceutical and food industries. Rheological testing of biopolymers used in these industries is necessary to provide information about their behaviour in physiological environments as their functionality within the body may be reliant on a particular mechanical performance. Some rheological measurements therefore, need to be undertaken in hydrochloric acid (HCl) to mimic particular physiological conditions. Simulated gastric fluid, for example, contains HCl possessing a pH which can be as low as pH 1. Moreover, certain biopolymers, depending on the industrial application, require preparation in HCl. This can cause problems when using conventional geometries due to the corrosion of stainless steel. Corrosion occurs on contact with HCl causing damage to the geometry and consequently influencing measurements and results. Therefore, it is necessary to develop geometries resistant to HCl corrosion. In this study, we have investigated using a geometry manufactured from poly(ether-ether-ketone) (PEEK) to measure the rheological properties of different biopolymers in HCl simulated gastric fluid..

To guide soil fertility investment programmes in sub-Saharan Africa, better understanding is needed of the relative importance of soil and crop management factors in determining smallholder crop yields and yield variability. Spatial variability in crop yields within farms is strongly influenced by variation in both current crop management (e.g. planting dates, fertilizer rates) and soil fertility. Variability in soil fertility is in turn strongly influenced by farmers’ past soil and crop management. The aim of this study was to investigate the relative importance of soil fertility and crop management factors in determining yield variability and the gap between farmers’ maize yields and potential yields in western Kenya. In this study, 522 samples taken from the farms were analysed by diffuse reflectance spectroscopy, using an ASD FieldSpec FR spectroradiometer.

International standard test methods used for the determination of Mg, P, S, Cl, Ca, Cu, Zn and Ba in unused lubricating oil samples by means of WDXRF include ASTM D4927 and ASTM D6443. In this study we have set up calibrations in accordance with the methods stipulated in both ASTM norms. XRF is an excellent method for the multi-elemental analysis of new lubricating oils for the purposes of quality control, product development and product performance classification. Small Sample Size Evaluation using LabSpec 4 Standard-Res i Практическая аннотация

Many analytic applications benefit from the ability to measure very small samples non-destructively. Typical applications of near-infrared analysis require sample sizes of 5 to 50 grams. The purpose of this evaluation was to determine if sub-gram quantities of sample could be used with the ASD LabSpec 4 Standard-Res i. A set of dried ground corn stover samples was used for this study to evaluate this small sample size capability. ASD’s spectrometer system was used with very small samples in micro-centrifuge tubes to allow higher throughput in the lab thus reducing costs associated with the NIR analysis.

X-ray micro-diffraction in wall painting research Практическая аннотация

A study by the Academic Laboratory of Materials Research of Paintings (ALMA) in Prague, Czech Republic has shown that X-ray powder micro-diffraction can be used for the investigation of salt damage. The use of X-ray microdiffraction enabled the research team to identify the probable causes of damage to an ancient fresco. Pigment degradation, often resulting in color changes, causes serious damage to wall paintings. Many factors lead to pigment degradation including sunlight, air pollution, salts and microorganisms. Of these, salts are one of the most damaging agents, especially for artworks of a porous nature such as wall paintings.

In this paper we present how SAXS measurements can be performed on a multi-purpose X-ray diffractometer platform. Application examples for nanopowders, colloids, nanocomposites and porous materials will be given. A multi-purpose X-ray diffractometer platform was configured for small-angle X-ray scattering measurements. The FieldSpec 4 spectroradiometer we received during the Goetz Instrument Support Program award period was used for the non-invasive analysis of a selection of medieval and Renaissance illuminated manuscripts (10th-16th century) in the collection of the Fitzwilliam Museum in Cambridge, UK. Since sampling or contact with the artwork is not required, FORS is a very useful method for the technical analysis of manuscripts, for which sampling is very rarely allowed. We analysed selected folios within 23 bound manuscripts, as well as 22 manuscript cuttings and a 17th century colour chart (Trinity College 310.b.6A.8). Green pigments were surveyed on 7 additional manuscripts, and spectra were acquired on 5 paintings at the Hamilton Kerr Institute. Almost 2500 reflectance spectra were collected on the manuscripts alone, and in-depth data processing is still ongoing. In order to correctly identify the different pigments and paint binders used on the manuscripts, we also created a database by acquiring spectra on ‘test samples’ made of historic pigments bound in different media and painted on glass and parchment.

This paper present the results of a preliminary study to assess the potential of the visible, NIR and SWIR energy of the EMR in differing iron ores of different grades in a rapid manner using hyperspectral radiometry. Using different iron ore samples from Noamundi mines, Jharkhand, India, certain spectro-radiometric measurements and geochemical analysis were carried out and the results have been presented. It was observed that the primary spectral characteristics of iron ores lie in the 850 to 900nm region. Various methods to evaluate the parameters for each curve such as: the radius of curvature of the maximum absorption trough, position of the trough with respect to wavelength in the NIR region and distance of the trough from a reference line, were adopted. Comparison of the spectral parameters and the geochemistry of the samples indicates that the depth of absorption (distance of trough from a reference line) in the NIR region for a sample has a strong positive correlation with the iron oxide content in the sample, while the radius of curvature of the absorption trough has a strong negative correlation with the iron oxide content. Such strong correlations indicate that hyperspectral radiometry in the visible and NIR regions can give a better estimate of the grade of iron ore. This study has demonstrated that generation of empirical models using hyperspectral radiometric techniques is helpful to quantify the grade of iron ores with limited geochemical analysis.

Remote Sensing from 700 km: Challenges and Potential Практическая аннотация

Remote sensing from 700 km, the nominal height for broad coverage, earth observation satellites, has advanced over the decades such that now, with adequate atmospheric correction, the instrument acquired radiance can be converted to spectral reflectance with sufficient accuracy for many applications. Industrial applications require quantitative analyses of material properties and abundances that far exceed those so far obtainable from orbital sensors using the sun as a source, viewing through a changing atmosphere. Although the physics of reflectance spectroscopy is the same for measurements from orbit and over the conveyor belt, the needs for illumination, sample presentation and signal-to-noise ratio drive different approaches for close-up remote sensing. This paper will attempt to illuminate the accuracy, illumination, calibration and stability requirements as well as solutions for systems used to make quantitative, real-time measurements of the properties of rocks on conveyors in industrial applications. Quantitative reflectance spectroscopy has the promise of making a step change in the efficiency of materials processing in factory settings.

This study demonstrates the accuracy and precision that the Axios FAST can deliver for controlling the composition of stainless steel, as well as the speed of analysis so important for controlling the cost of production. Quantifying Bitumen in Natural Oil Sands using NIR Technology Практическая аннотация

Oil sand and oil shale deposits are found all over the world. The two largest are the Athabasca Oil Sands in northern Alberta and the Orinoco River deposit in Venezuela. Oil sands are very different from conventional oil and natural gas deposits. Oil sands are a mixture of bitumen, water, quartz, clay and other gangue minerals. The bitumen, a particularly thick and tar-like hydrocarbon, coats the mineral grains in the sand matrix. The major challenge in developing oil sands is separating the bitumen from this sand matrix. Once separated, bitumen can then be upgraded into high-quality oil called "synthetic crude." The objective of this study was to demonstrate the capability of ASD’s field-portable FieldSpec spectroradiometer and NIR technology to perform field assessment of the concentration of bitumen contained in oil sand samples. Previous studies have also demonstrated the ability to quantify clay content as well as clay mineralogy using the FieldSpec spectroradiometer. Use of the TerraSpec® mineral analyzer has also proven successful in these applications.

Prediction of soil content using near-infrared spectroscopy Практическая аннотация

Near-infrared spectroscopy can be used to accurately predict certain soil properties, making it a valuable tool in precision farming. Modern technological developments in positioning, sensing, and control systems have opened a new era in which many traditional agricultural practices are being left behind. Replacing them are ‘precision farming’ techniques that manage variability within a field by applying agronomic inputs in the right place, at the right time, and in the right quantity to reduce the environmental impact of crop production. Visible and near-infrared (NIR) spectroscopy has emerged as one of these techniques, enabling the rapid and nondestructive analytical correlation of diffusely reflected NIR radiation with chemical and physical properties of soil components.

Specifically, the intent is to determine Protein (ds), Softness (avg.), Volume (avg.) and Moisture (%) properties.. A LabSpec® 4 with a wavelength range of 350 to 2500 nm and a contact probe were used for this study. The LabSpec 4 system features three modular spectrometers, each collecting a portion of the entire wavelength range. The contact probe is designed for contact measurements of solid raw materials such as minerals, grains, and other granular materials. .

Forests are a natural resource of major economic significance to Canada and many other countries. For example, Atlantic Canada forest industries exported over $3.9 billion in wood products and had a direct employment of 37,900 persons in 2003. Secondary and value added products, particularly that of Canadian northern bleached softwood kraft market pulps represent revenues reaching $7.5 billion in 2007. Canada leads the world in wood pulp exports. In addition to direct employment, the industry has an indirect effect through demand for goods and services and diversifies income sources for many communities.. More recently, wood is recognized as a renewable resource and can sequester CO2. There is increasing emphasis on using wood for energy, bio-fuels and bio-chemicals production. Timely knowledge of wood characteristics is essential to ensure that the right fiber is directed to the appropriate wood manufacturing company at the right time and right cost to produce the right product.. Agenda 2020 has identified that a number of key variables that need to be measured accurately along the wood processing, from trees to logs to lumber and to secondary processing such as pulp and paper products. Measurements of wood and processing parameters will allow process monitoring and control, leading to consistent high product quality at minimal costs and energy consumption.. Reflectance spectroscopy, particularly visible-NIR spectroscopy, has shown much promise as an adaptable technology that would meet the requirements for fast, non-contact, and process capable as online and at-line analysers for monitoring wood and wood products properties. This paper will discuss the application of reflectance spectroscopy for measurements of standing tree wood properties, lumber properties during sawmilling operation, OSB flake properties and its use in panel production, and other pulp and paper applications. The paper will focus on adapting reflectance spectroscopy for on-line and at-line measurements for 24/7 operations..

The closed-cycle helium cryostat for X-ray powder diffraction applications was evaluated for temperature accuracy. Two methods were used to test the cryostat: thermal expansion and phase transition, and both methods confirmed the instrument’s accuracy and its suitability for low temperature powder diffraction. The PheniX cryostat is a valuable extension to the non-ambient capabilities of the Empyrean diffractometer. The Phenix from Oxford Cryosystems is a closed-cycle helium powder cryostat. Designed for cooling flat-plate powder samples, the PheniX enables data collection close to liquid helium temperatures and combines fast cooldown and warm-up times with ease of use. The PheniX is equipped with a closed-cycle cryostat; a continuous supply of liquid helium is not required. It can be used to conduct X-ray powder diffraction experiments on θ-θ and θ-2θ type systems.

This application note shows the possibilities of Empyrean in the field of nanomaterial characterization, by combining SAXS (small-angle scattering), LPA (line profile analysis) and PDF (pair distribution function), also in non-ambient conditions. The Empyrean allows measurements of many types of samples and supports many different applications. Small-angle scattering is used to determine particle and cluster dimensions and is insensitive to length scales below 1 nm i.e. those comprising small domains and crystal structure. Small-angle scattering is therefore useful in nanotechnologies because it is measuring directly the particle and cluster dimensions that are often most significant in the exploitation of nanoparticle properties.

Sample holders for air-sensitive materials Практическая аннотация

Air-sensitivity of a sample material (change of properties in direct contact with oxygen or/and water) is an important issue for many research fields. Besides other materials, a large number of pharmaceuticals, catalysts, materials for gas storage applications, battery research, etc. requires an oxygen-/water-free atmosphere for their characterization. Adequate protection of a sample material from air during an X-ray experiment is an important factor for a successful analysis. Here we present a comparative study of different sample holders for air-sensitive materials (based on air-tightness, contribution to background and transparency to X-rays).. In this application note it is shown that Anton Paar domed sample holder with a PEEK dome is the most air-tight option, which comes at a cost of the strongest contribution to the XRD background. However, PEEK dome background contribution can be reduced by placing the detector anti-scatter slit closer to the sample. In the figure below we compare the XRD background of two air-sensitive sample holder types, measured using two instrument configurations for X-ray diffraction in reflection: 1. Instrument configuration with the detector anti-scatter slit at the standard position 2. Instrument configuration with the anti-scatter slit for transmission geometry placed in front of the 1D detector. For the standard setup with programmable slits background contribution of the Anton Paar domed sample holder with a PEEK dome (green line) is quite significant, much stronger than that of the PANalytical sample holder with a Kapton foil (red line). Similar result is observed for the standard instrument configuration with the fixed slits (not shown here). For the non-standard configuration, with the detector anti-scatter slit for transmission, the PEEK dome background contribution (blue line) is significantly reduced whilst background of the PANalytical sample holder (brown line) is virtually the same. The non-standard instrument configuration with the detector anti-scatter slit for transmission geometry is undoubtedly beneficial when using Anton Paar domed sample holder. However, considering the narrow opening of the detector anti-scatter slit, incident beam divergence should be optimized to ensure correct peak intensity over the entire measured 2Theta range. We advice to use fixed divergence slit mode with rather narrow slit opening. .

In this study the improved performance of PX10 relative to LiF200 is shown by comparing the measured sensitivities, calculated detection limits and decreased measurement times for the K-lines of elements between potassium and cerium. For low-level trace element analysis by wavelength dispersive XRF (WDXRF), it is essential to maximize the analyte peak to background ratio and maintain a high spectral resolution to minimize line overlap issues. This study demonstrates that Zetium is capable of providing accurate results for the analysis of FeCr alloys prepared using the fused bead method. Compared to classic analytical methods, XRF offers a competitive advantage with short throughput time, making Zetium highly suitable in process control environments, especially in combination with a fusion sample preparation machine. Ferrochrome is the most widely used ferroalloy. It is a very common practice to determine the chemistry of ferroalloys by ‘wet chemistry methods’. However, this technique involves high labor costs and it requires long sample preparation and analysis time. X-ray fluorescence spectroscopy is an excellent alternative technique, offering easy routine sample preparation and high throughput. However, the analysis of pressed pellets and loose powders presents imitations in precision, due to severe metallurgical as well as particle size effects. To overcome these problems, PANalytical has developed a method to analyze ferroalloys as fused beads, eliminating in this way metallurgical and particle size effects.

This application note shows the possibilities of Empyrean for the analysis of gas storage materials, illustrating two examples of interactions with hydrogen.. Such studies are normally performed at large-scale facilities like synchrotrons and neutron sources, or by using dedicated instruments. It is essential to quantify nutrients such as vitamins and minerals in animal feed products, including dog food, to avoid legal and financial consequences related to misbranding. Dog food samples are difficult to analyse since they have a high volatile organic content. To overcome this challenge, special attention must be paid to the pre-treatment of samples prior to the lithium borate fusion step. In fact, special thermal pre-treatments are necessary to remove the organic materials and keep volatile elements of interest. An oxidizer is also needed to complete the oxidation of the partly reduced material during the fusion process. .

Experiments were conducted according to the guidelines of two well-known cement chemical analysis International Standard Methods and results were presented. This data sheet proves the capability of the Air Quality edition of Epsilon 4, a benchtop EDXRF spectrometer, as an analytical tool for the analysis of air filters according to the US EPA method IO-3.3. The presence of toxic pollutants in the air has been a subject of research for many years in countries around the world. In the United States, air quality standards are governed by the ‘Clean Air Act’ and administered by the US Environmental Protection Agency (EPA). One of the key areas of concern for the US EPA is the Suspended Particulate Matter (SPM) of air. Research on the health effects of SPM in ambient air has focused increasingly on particles that can be inhaled into the respiratory system, i.e. particles of aerodynamic diameter of < 10 μm. These particles are referred to as PM10 (2.5 – 10 μm) and PM2.5 (<2.5 μm). Despite chemical toxicity, these particles are a significant threat to health.

The data presented in this application note clearly demonstrate that the Epsilon 4 EDXRF spectrometer can measure elemental impurities at the levels required by ICH Q3D and USP <232> using only 100 mg of sample. The EDXRF data compares favorably with ICP data. The repeatability results illustrate the stability and robustness of the Epsilon 4. The combination of excellent detector resolution, high sensitivity and powerful software deconvolution models contribute to the accuracy and precision of the results. The non-destructive nature of the method makes it possible to measure the samples subsequently by other analytical techniques, if required.

Dynamic Light Scattering (DLS) is a powerful and sensitive technique for characterizing particles or macromolecules in dispersion, due to its ability to resolve particle or molecular sizes ranging from sub nanometer to several microns. This sensitivity also means that DLS is a useful technique to characterize aggregated material, which may occur in far smaller quantities but is of a significant importance in many applications. Typically, however, the presence or dust (including filter spoil, column shedding, tracer aggregates or material from dirty lab ware), can have detrimental effects on the measurement of smaller sized particles and algorithms exist to suppress these effects. In this application note we present a new approach to handling DLS data which prevents the skewing of data for small particles whilst retaining insight into the presence of aggregates that may otherwise be lost, whereby the relative abundance and size of aggregates can be deduced.

The economy and efficiency of Waters’ ACQUITY APC combined with the powerful advanced GPC/SEC detectors of OMNISEC REVEAL allowed the complete characterization of the low molecular weight polystyrene and polycaprolactone samples. The individual oligomers of both samples could be observed and characterized separately due to the resolution of the APC columns and sensitivity of the OMNISEC REVEAL detectors – with standard 30 cm analytical GPC/SEC columns it is not possible to resolve the oligomers at all. The greatly reduced quantity of solvent and time required to obtain comprehensive characterization of samples using the APC-REVEAL system presents obvious benefits to both researchers and manufacturers working with polymeric materials.

For virus development and production, it is important to know the virus concentration at different stages of the process, to optimize the clone used as well as the production yields. For example, instances such as clone screening, multiplicity of infection (MOI) optimization and adaptations of cell culture methods are instances when virus concentration, or virus titre as it is also commonly called, is of interest. Dynamic light scattering (DLS) is often used in virus development, in a screening function to separate good and stable samples from those with contaminants or aggregates. With the new multi-angle DLS (MADLS) based particle concentration measurement in the Zetasizer Ultra, it is now possible to get both size and size distribution as well as particle concentration per population, within a few minutes. In this application note, Allergan are kindly sharing data from their evaluation of the Zetasizer Ultra, examples of three adeno-associated virus (AAV) samples are shown. The concentration results are compared to results from capsid ELISA based virus titre assays.

Gisèle Usanase, a Masters Student at C2P2 laboratory (Chemistry Catalysis Polymers and Processes) based at University of Lyon describes her experience of working with the Kinexus and how it provided a quick and inexpensive means for quantifying co-monomer content in LLDPE. C2P2 is a research center with a major focus on fundamental chemistry, catalysis and chemical engineering to enhance the ability to produce organic, inorganic and composite materials, and to propose the processes used for this purpose. C2P2 brings together a range of unique skill sets and capabilities which enable them to perform high level research, based on original concepts, and backed up by a scientific approach. As part of her Masters project, Gisèle Usanase wanted to use rheology as a tool for probing the physical properties and composition of LLDPE and approached Malvern Panalytical for assistance with this project. Dr Frédéric Fraϊsse, Malvern Panalytical’s Rheology Specialist in France, was able to provide access to a Kinexus Ultra+ rheometer along with training and support so that Gisèle could complete her project.

Dynamic light scattering (DLS) is a technique that is widely used to measure dispersed particle size (specifically the hydrodynamic diameter) via direct measurement of the translational diffusion coefficient within the continuous phase. Chocolate is a complex multi-phase product consisting of fat, milk, cocoa powder and sugar, as well as other additive such as emulsifiers and flow modifiers. Of critical importance for both the sensory characteristics (texture) and processing of chocolate is its rheology or viscosity, which is influenced by many factors including particle characteristics such as size and shape, as well as fat composition and structure. To date the Casson model has been widely used to characterise the flow properties of Chocolate. This model is used to assess differences in chocolate flow properties that can be appropriated to the behavior during processing such as pump-ability and slump characteristics. In this application note, chocolate samples were analysed using the aforementioned method and the results discussed.

The increased demand for portable electronic devices, including mobile phones, laptops and new ‘wearables’, has required advances in battery technology to provide a low-cost, lightweight, long-lasting and stable power source. With fossil fuels dwindling and CO2 regulations becoming more stringent, battery technology is increasingly being used in applications such as renewable energy storage and electric vehicles, which require ever more lightweight, safe, high-power and fast-charging batteries. During discharge, lithium ions (Li+) move from the anode to the cathode, conducted via the electrolyte, with flow in the reverse direction occurring when the battery is charged. Anodes are typically graphite-based, while cathodes are often manufactured using lithium iron phosphate, lithium cobalt oxide, lithium-manganese oxide, lithium-nickel cobalt manganese oxide, etc. The cornerstones of battery performance are power, which impacts current and discharge characteristics, and energy storage capacity. Battery power is determined by the rate of reaction between the electrodes and the electrolyte, while storage capacity is a function of the volume of electrolyte within the cell. These properties are intrinsically linked to the intercalation structure and primary particle size of the electrode particles, which determine how well the mobile ions are taken up and released by the electrode1. Particle size distribution and particle shape also influence particle packing and hence the volume of electrolyte that can be accommodated within the interstitial voids of the electrode, which affects storage capacity. Monitoring degradation of sodium alginate using GPC/SEC Практическая аннотация

Sodium alginate, the sodium salt of alginic acid, is a polysaccharide harvested from algae, specifically brown seaweeds. Due to its similar function to cellulose in plants, sodium alginate possesses a structural integrity which results in a viscous gum that forms when it interacts with and absorbs water. Because of this, sodium alginate lends itself to applications in the food industry where it thickens solutions, dehydrates and/or expands upon the presence of water, or serves as a protective gel or coating for drug delivery. To produce these end products, manufacturers and researchers must be able to identify and relate the physical properties of sodium alginate samples with specific molecular properties, such as molecular weight, intrinsic viscosity, and molecular size, that are appropriate for each application. As such, the accurate analysis and molecular weight characterization of sodium alginate is of paramount importance.

Field Characterization of White Micas Using the TerraSpec Halo Тематическая публикация

The white micas are a diverse group of common fine-grained phyllosilicate minerals that include the true micas paragonite, muscovite, and phengite, as well as the K-deficient mica, illite. This group often serves as a useful exploration vector as they provide an indicator of geothermal and geochemical conditions at the time of their formation. Using the Al-OH scalar reported by TerraSpec Halo mineral identifier it is possible to track geochemical conditions while the Illite Spectral Maturity (ISM) scalar (Doulblier et al. 2010) reported by Halo provides an indicator of thermal maturity. The white micas contained in the mineral library used by the TerraSpec Halo mineral identifier are organized by composition. The true white micas are represented by the potassium white micas muscovite and phengite, and the sodic white mica paragonite. The muscovite-phengite series is characterized by increasing substitution of Mg+2 or Fe+2 and Si+4 for Al+3. The illites are classified similarly. Thus, a shift in mineralogy from muscovite and/ or illite (labeled as K-Illite in the Halo mineral library) to paragonite and/or paragonitic illite (labeled as Na-Illite in the Halo mineral library) is indicative of increasingly sodic conditions. Similarly, a shift in mineralogy to phengite and/or phengitic illite (labeled as Mg-Illite in the Halo mineral library) is usually indicative of increasingly magnesium-rich conditions. The TerraSpec Halo mineral identifier is able to track geothermal gradients both by detecting mineralogy and through the reporting of the ISM scalar. With increasing metamorphic grade, smectites convert to illites, which then covert to muscovite (and other true micas). Within the illites, subtle geothermal trends are easily detected and mapped using the Halo’s ISM scalar which tracks the illite dehydration process as the illite converts to true mica.

As a (future) user of a Malvern Panalytical X-ray diffractometer you can perform almost all types of diffraction applications, depending on the configuration of your system. To get the best possible results out of your system, this book gives you an introduction to X-ray powder diffraction (XRPD or, mostly used, XRD) and provides helpful information. It gives a simple explanation of how a diffractometer works and how XRD analysis is done. This book is intended both for people new to the field of XRD analysis and for more experienced users to find new applications which might be helpful in daily work. This book avoids complex mathematical equations and understanding its contents only requires a basic knowledge of crystallography, mathematics and physics. It is not dedicated to specific diffractometer configurations or application areas, but aims to give a global overview of the various possibilities in XRD. Chapter 2 briefly explains XRD and its benefits. General application fields are presented. Chapter 3 gives an introduction to the physics and crystallography of XRD, while Chapter 4 describes how this is used for diffraction experiments. Chapter 5 explains the XRD data collection. This includes presentation of sample preparation and measurement procedures to get the best possible results. In Chapters 6-9 the procedures to perform qualitative and quantitative phase analysis are presented. Chapter 10 deals with non-ambient XRD and describes its methods and applications together with other techniques. Chapter 11 lists recommended literature for further information on XRD analysis. Since this book is intended to give an introduction to standard powder diffraction techniques, it does not cover other X-ray techniques like single crystal diffraction, high-resolution XRD of heteroepitaxial layers, or small-angle X-ray scattering (SAXS).

Isothermal Titration Calorimetry (ITC) is a powerful and versatile technique that is widely used for measuring the binding affinity and thermodynamics of equilibrium association reactions. ITC is also an accepted, universal label-free assay technique that can be applied to any enzyme-catalyzed system, provided that the reaction is associated with a change in enthalpy – see the companion whitepaper and other review articles [1-8]. ITC does not rely on a spectroscopic readout and labelled reagents, and the experiments are quick – ITC data for a Michaelis-Menten curve can be generated in 30 -90 minutes with current instrumentation – and only small amounts of material are required, making it an appealing alternative for enzyme kinetics and inhibition assays. Reference Measurements: The What, Why, When and How Практическая аннотация

IntroductionAccurate field spectral reflectance measurements are important in the calibration and validation of airborne and space-borne hyperspectral and multispectral imagery. The field portable ASD FieldSpec® 4 spectroradiometer was designed with these requirements in mind. However, even the best instrument can deliver inaccurate data if the measurements are not carried out properly. One of the most important actions a researcher takes in the field is frequent reference measurements. This paper serves to explain what a reference measurement is in relation to reflectance measurements, why reference measurements are important, and how to properly perform them in the field. What is a reference measurement?Before discussing reference measurements, it is important to provide a definition of a reflectance measurement. Reflectance is not a direct measurement. A reflectance measurement is actually a ratio derived from two separate radiance measurements. It is calculated by dividing the radiance of a sample by the radiance from a reference panel of known reflectance, usually having nearly 100% reflectance. This reflectance panel measurement is known as a reference measurement. This relative reflectance spectrum is then sometimes corrected to absolute reflectance by multiplying it be the known reflectance of the reference panel. Why is a reference measurement so important?The solar spectrum contains many absorption features, the strength of which are a function of solar elevation angle, altitude, and atmospheric conditions (Fig. 1). By ratioing the radiance of the observed sample to the radiance observed when viewing the reference panel, these features are removed from the resulting reflectance spectrum. An Inherent assumption in this method of ratioing radiances is that the solar irradiance and light scattering effects from the local surroundings is the same for both the sample and reference measurements. To obtain an accurate reflectance spectrum, the irradiance illuminating the reference panel must be the same as the irradiance illuminating the sample. Any change in the irradiance between the measurement of the reference panel and the sample will result in atmospheric absorption features being imprinted on the measured reflectance spectrum. This is what drives the need to frequently re-measure the reference panel. Figure 1. Solar radiance reflected from a 99% reflective reference panel. We present the use of Multi-angle Dynamic Light Scattering (MADLS®) for the measurement of nanoparticle concentration. We describe the theory of the method and its application to nanoparticles made of gold, silica and polystyrene, with diameters ranging from 30 to 400 nm, and demonstrate some of the limitations with particles of sizes 500 nm and above. We evaluate the method accuracy, linearity and reproducibility, as well as the operational nanoparticle concentration and size range. We show that the concentration working range depends on the material’s optical properties, size and concentration. Here it is shown that the level of accuracy that can be expected for the concentration of particles is typically within 50% of the nominal value across a range of materials and sizes and, for some samples, within 20%. The repeatability of the measurements, in terms of relative standard deviation, is typically below 30%. A linearity of within 40% across a concentration range of 3·108 to 2·1011 mL−1 for concentration measurements was also demonstrated by using gold nanoparticles and gravimetric dilutions for method validation. Overall, we show that MADLS® is a rapid and straightforward method for the reproducible measurement of particle concentration, as well as size, requiring minimal sample preparation, without the need to calibrate using a pre-determined concentration series, and applicable to a broad range of materials. These features make it an ideal tool to support both development and quality control of particle materials for a broad range of applications.
