Aggregation of both biological and synthetic materials is a source of serious concern, posing problems at various stages of the discovery and formulation processes. Liposomes have been used in drug discovery and drug delivery for some time, and the biophysical characterization of these systems and their payloads is critical to understanding their function. One such payload is the membrane-associated receptor tyrosine kinase (RTK) targets which continue to be the focus of many discovery campaigns. International standard ISO16332 Diesel engines — Fuel filters — Method for evaluating fuel/water separation efficiency (the US equivalent being SAE J1488) is currently undergoing revision and its publication is imminent. This new version standardizes the testing of filters between manufacturers and looks at the effectiveness of the test rig and how potential droplet recirculation could affect particle size distribution (PSD). It outlines testing of filter efficiency and gives a model particle size distribution for this. Laser diffraction is specified in the standard as the only method for sizing this distribution. This white paper examines how laser diffraction particle sizing, already an established tool in fuel filter testing, is stepping up to the challenges posed by modern fuels and the latest testing requirements. Analyses of a variety of sample types, including fused beads, pressed powders, metals, liquids and plastics, illustrate the performance of Omnian. This note demonstrates the flexibility of Omnian and illustrates numerous advanced features designed to enhance accuracy. All measurements were carried out on a Zetium WDXRF spectrometer. In conventional XRF analysis multiple standard reference samples are used to make calibration lines for each of the elements to be analyzed. Preferably, these reference samples should have compositions similar to the unknown samples and when a large number of different sample types are to be analyzed, different calibrations need to be maintained for each type. This can be very time-consuming and expensive in terms of costly certified reference materials. Under such circumstances, standardless analysis with Omnian is a very valuable solution. With one measurement program, Omnian covers all elements from F to U and quantitative results can be obtained for a wide variety of sample types.

The recent changes to the United States Pharmacopeia (USP) regarding requirements for elemental impurity testing has peaked interest into new ways to perform this testing. This study demonstrates that the X-ray Fluorescence (XRF) spectrometry is capable of performing elemental analysis of a wide range of pharmaceutical (liquid, powder and solid) materials with high sensitivity, precision and accuracy for many of the elements required by the United States and the European pharmacopeias. Introduction This white paper outlines the main benefits of using hard radiation for material analysis studies. X-ray powder diffractometers are generally equipped with X-ray sources using Cu anodes. For specific applications, however, switching to shorter wavelength, ‘harder’ radiation, like obtained from X-ray tubes with Mo or Ag anodes, gives significant improvements over Cu, or even allows experiments that are impossible with Cu radiation, like experiments where a large Q range needs to be probed. Often, people make use of synchrotron radiation, but new developments on source, optics and detector technologies allow a variety of hard radiation experiments in the home lab. In this white paper we present six key advantages of hard radiation studies. Larger penetration depth For inorganic materials, the penetration depth of Cu K-alpha is often only a few micrometers (see Table 1). In order to extract information from a larger sample volume, hard radiation is an advantage in transmission experiments. Thanks to the larger penetration depth, it is possible to observe crystallographic changes in working devices, such as Li-ion batteries, and perform highpressure experiments with diamond anvil cells. In this study, small spot analysis and elemental distribution mapping of the Northwest-Africa 2086 (NWA 2086) CV3 meteorite were conducted. Qualitative mapping and semi-quantitative spot elemental composition analyses were compared to phase identification using X-ray diffraction for cross verification. The qualitative mapping results show that elements such as Al and Ca are abundant in the inclusion region while Fe and Cr are more prominent in the meteorite body. Combined XRF analysis and XRD phase identification show the presence of sodalite in addition to variations of olivine, pyroxene, and spinel in the rim and the core of calcium-aluminum rich inclusions (CAIs). The sodalite is an indication of early Na-Cl metasomatic processes in the solar nebula. John Carpenter is a Professor of Pharmaceutical Sciences in the Department of Pharmaceutical Sciences at the University of Colorado, Anschutz Medical Campus. His laboratory is involved in researching the causes and control of aggregates and particles in therapeutic protein products which occur during manufacturing, shipping, storage and delivery to patients. This research is important as it contributes to improvements in product quality and therefore to reductions in adverse reactions of patients to aggregates and particles within biopharmaceutical products, which can include immune responses and dangerous infusion reactions. Also, by investigating the roles of key biophysical properties in protein stability and degradation, rapid and rational formulation strategies can be developed to help ensure that a product’s stability is properly optimized.

There is often a necessity to measure, or at least estimate, true viscosity values using non-standard measuring systems on a rotational rheometer. This may be to replicate a mixing or manufacturing process on a lab scale, to keep a sample dispersed and uniform during a measurement or to measure some rheological property that would be difficult or impossible with a standard configuration. Such measurements can be made easily enough, but without a process for converting torque to shear stress and angular velocity to shear rate only these raw data variables can be reported. In this poster a simple and novel empirical method for determining strain/strain rate (C1) and stress (C2) constants for non-standard measuring systems on a rotational rheometer is presented. This method uses relative torque measurements made with a Newtonian and non-Newtonian material and their corresponding power law fitting parameters to determine C1 and C2 using a non-linear regression analysis. Equilibrium flow curves generated for two non-Newtonian fluids using two non-standard mixing geometries show very good agreement with data generated using a standard cone and plate configuration, therefore, validating the approach.

Since the 1990s, the automotive industry has become the largest consumer of platinum (Pt), palladium (Pd), and rhodium (Rh) on account of the arrival of catalytic converters in car exhaust systems. Approximately 40% of the total Pt, 74% of the total Pd and 80% of the total Rh were consumed by the auto catalyst industry in 2012. Malvern Panalytical has developed a simple sodium peroxide fusion method, for the dissolution of automotive catalytic converters substrates samples. Combined with a simultaneous ICP-OES, it yields good accuracy and precision for the determination of Pt, Pd and Rh contents in autocatalysts. The Claisse® fusion method is quick, allows total sample dissolution, eliminates the use of dangerous acids and demonstrates good accuracy, precision and recovery for Pt, Pd and Rh. The analysis and determination of metals and trace metals in high silica and alumino-silicate samples such as catalysts and Zeolites are never easy, mainly because of the dissolution process. Samples containing silica often need hazardous chemicals (HF), strong acids (HClO4) or microwave digestions. This application note describes a new approach using lithium borate fusion as the sample preparation method as well as the optimization of ICP-OES parameters. Many elements showed a signal suppression varying from 0 to 40 % while Plasma ionization effects from the borate solution enhanced the K, Na, and Rb signals. High levels of Si, Al, Ca and Na from the sample and Li from the matrix required a strict scheme to correct all spectral interferences. The PerkinElmer® WinLab 32 software for the Optima™ ICP-OES allowed for three spectral correction techniques and they were used as follows: • Background correction if no wing lines nearby; • Multi-component spectral fitting ( MSF ) when there is a partial overlapping; • Inter-element corrections ( IEC ) when lines are completely overlapped. It is essential to quantify nutrients such as vitamins and minerals in animal feed products, including dog food, to avoid legal and financial consequences related to misbranding. Dog food samples are difficult to analyse since they have a high volatile organic content. To overcome this challenge, special attention must be paid to the pre-treatment of samples prior to the lithium borate fusion step. A previous paper portrayed sample preparation by fusion methodology and the X-Ray Fluorescence (XRF) spectrometry analytical conditions for the calibration of cement materials[1]. Experiments were conducted according to the guidelines of two well-known cement chemical analysis International Standard Methods and results were presented. The results proved that this robust analytical method is able to qualify by the ASTM C 114[2] and ISO/DIS 29581-2[3] norms. This analytical method was developed using a Claisse® M4TM fluxer, an automated fusion instrument for the sample preparation and a Wavelength Dispersive X-Ray Fluorescence (WDXRF) spectrometer, for the determination of all the elements of interest relating to the cement industry. The method was used to prepare finished products, process materials, as well as a very large range of raw materials which will be described in this second paper. Cement, blended cement, cement with additions, aluminate cement, clinker, kiln feed, raw mix, limestone, gypsum, sand, clay, bauxite, silica fume, slag, fly ash and iron ore are among the raw materials covered by this analytical application. This sample preparation was also transferred and optimized for TheOx® fluxer, a fully electric fusion instrument, by executing a robust validation of the method’s performance. Due to the fact that powders are affected by mineralogy and particle size effects[4, 5], it is almost impossible to use a single XRF calibration curve for the analysis of such a wide range of different materials using the pressed powder preparation method. When fused in a borate glass, all mineralogy and particle size effects are eliminated. A single XRF calibration curve covering the whole range of concentrations for all elements of interests for the cement industry raw materials analysis can be made. This paper examines all the XRF analytical conditions for the calibration of the entire range of raw materials using the robust borate fusion sample preparation methodology as well as the numerous Reference Materials (RMs) used for this analytical application. The results of this general and unique XRF raw materials calibration will also be presented in terms of precision, accuracy, and limit of detection. Fuel oil is one of the main by-products of petroleum refining. It is used as fuel for ocean liners and cargo ship engines. It is also used in power plants and industrial plants. Fluid catalytic cracking (FCC) is often found in the petroleum refining process to maximize the distillation of hydrocarbon with a high octane rate. Some contaminants such as nickel, iron, vanadium, silicon, aluminium, sodium and calcium are found in FCC and are transferred in fuel oil during the refining process. It is important to find the concentration of these elements because they are adhesive at certain temperatures and they cause corrosion on the reheater tube surfaces of boilers as well as on the combustion chamber and components of fuel oil engines. Corrosion should be avoided as much as possible because it causes wear and entails additional costs for maintenance or replacement.

Whilst being able to measure particles of below 1 nm size, DLS is preferentially sensitive to larger particles due to the 6th power relationship between particle radius and scattering intensity. This means that sample preparation typically needs to be scrupulous, especially for low scattering samples such a proteins and biological molecules. The contribution to contaminants such as dust and aggregates can be mitigated by filtering, however this may not always be practical or possible depending on the volume and fragility of the sample. Filtration of samples can also constitute a financial burden, both in terms of additional sample preparation time and consumables costs. This application note introduces a new DLS data capture process called Adaptive Correlation which helps address these issues. Adaptive Correlation uses a statistically driven approach to produce the best correlation data, which in turn gives more reliable DLS size data. It can reduce the need for filtering and give added confidence in you DLS results.

Liposomes are nowadays frequently used as drug delivery systems, and as such requires that they are well characterized for manufacturing and batch release. This characterization includes their size and size distribution. For these parameters, dynamic light scattering (DLS) is commonly used to characterize size and size distribution and also nanoparticle tracking analysis (NTA) has previously been shown to be useful for measuring liposomes size and concentration.1 This application note will demonstrate how the multi-angle DLS (MADLS®) based particle concentration measurement in the Zetasizer Ultra is capable of measuring the particle concentration as well as size and size distribution of a range of dilutions of a liposome sample. A comparison will be made with NTA measurements of the same samples.

Polyvinyl chloride (PVC) is a common polymer used in countless applications from pipes, to flooring, and electrical insulation. As one of the world’s most widely produced polymers, it is very much a commodity polymer where tight margins mean little room for detailed analysis. However, even in this case, high level analytical tools can be used to characterize the polymer for appropriate grading. For example, the glossiness of a PVC sample has a significant effect on its finish and grade, and for which applications it can be used. Applying appropriate characterization tools can help grade different PVC batches, assigning them to appropriate applications to maximize the value of high quality batches, saving lower grades for others. In this application note, four PVC samples of known differences in glossiness were measured with both multi-detector GPC and rotational rheometry. The results both explain the differences in glossiness and the likely cause of those differences.

Gisèle Usanase, a Masters Student at C2P2 laboratory (Chemistry Catalysis Polymers and Processes) based at University of Lyon describes her experience of working with the Kinexus and how it provided a quick and inexpensive means for quantifying co-monomer content in LLDPE. C2P2 is a research center with a major focus on fundamental chemistry, catalysis and chemical engineering to enhance the ability to produce organic, inorganic and composite materials, and to propose the processes used for this purpose. C2P2 brings together a range of unique skill sets and capabilities which enable them to perform high level research, based on original concepts, and backed up by a scientific approach. As part of her Masters project, Gisèle Usanase wanted to use rheology as a tool for probing the physical properties and composition of LLDPE and approached Malvern Panalytical for assistance with this project. Dr Frédéric Fraϊsse, Malvern Panalytical’s Rheology Specialist in France, was able to provide access to a Kinexus Ultra+ rheometer along with training and support so that Gisèle could complete her project.

The most common approach for measuring viscoelastic properties using a rotational rheometer is to perform small amplitude oscillatory shear (SAOS) testing within the samples linear viscoelastic region (LVER). An alternative approach for obtaining time dependent viscoelastic information is by means of a creep test in which a constant and unidirectional shear stress is applied to the sample and the resultant shear strain measured with time. While creep and oscillatory tests, and the results generated, are often considered independently they are in fact intrinsically linked. As with spectroscopic techniques such as NMR the time domain data can be transformed to the frequency domain using mathematical procedures such as the Fourier transform or equivalent algebraic procedures. One such algebraic procedure is based on an approach widely used in the field of microrheology for converting the mean square displacement to complex modulus which has also been adapted for converting creep compliance data also. In this technical note we demonstrate how such an approach can be used to generate comparable data to oscillatory testing but in a fraction of the time.

By combining measurement data, sequentially collected, from three measurement angles it can provide improvements in resolution, typically from 3:1 to 2:1. It also provides improvements in accuracy by reducing signal to noise ratio, allowing lower smoothing coefficients to be used, and by reducing angular dependence of the final particle size result, as illustrated in the following Mie solution plot for a single protein macromolecule measured at 45, 90, and 173 degree scattering angles. The increased demand for portable electronic devices, including mobile phones, laptops and new ‘wearables’, has required advances in battery technology to provide a low-cost, lightweight, long-lasting and stable power source. With fossil fuels dwindling and CO2 regulations becoming more stringent, battery technology is increasingly being used in applications such as renewable energy storage and electric vehicles, which require ever more lightweight, safe, high-power and fast-charging batteries. During discharge, lithium ions (Li+) move from the anode to the cathode, conducted via the electrolyte, with flow in the reverse direction occurring when the battery is charged. Anodes are typically graphite-based, while cathodes are often manufactured using lithium iron phosphate, lithium cobalt oxide, lithium-manganese oxide, lithium-nickel cobalt manganese oxide, etc. The cornerstones of battery performance are power, which impacts current and discharge characteristics, and energy storage capacity. Battery power is determined by the rate of reaction between the electrodes and the electrolyte, while storage capacity is a function of the volume of electrolyte within the cell. These properties are intrinsically linked to the intercalation structure and primary particle size of the electrode particles, which determine how well the mobile ions are taken up and released by the electrode1. Particle size distribution and particle shape also influence particle packing and hence the volume of electrolyte that can be accommodated within the interstitial voids of the electrode, which affects storage capacity. Sodium alginate, the sodium salt of alginic acid, is a polysaccharide harvested from algae, specifically brown seaweeds. Due to its similar function to cellulose in plants, sodium alginate possesses a structural integrity which results in a viscous gum that forms when it interacts with and absorbs water. Because of this, sodium alginate lends itself to applications in the food industry where it thickens solutions, dehydrates and/or expands upon the presence of water, or serves as a protective gel or coating for drug delivery. To produce these end products, manufacturers and researchers must be able to identify and relate the physical properties of sodium alginate samples with specific molecular properties, such as molecular weight, intrinsic viscosity, and molecular size, that are appropriate for each application. As such, the accurate analysis and molecular weight characterization of sodium alginate is of paramount importance.

The white micas are a diverse group of common fine-grained phyllosilicate minerals that include the true micas paragonite, muscovite, and phengite, as well as the K-deficient mica, illite. This group often serves as a useful exploration vector as they provide an indicator of geothermal and geochemical conditions at the time of their formation. Using the Al-OH scalar reported by the ASD TerraSpec® Halo mineral identifier it is possible to track geochemical conditions while the Illite Spectral Maturity (ISM) scalar (Doulblier et al. 2010) reported by Halo provides an indicator of thermal maturity. The white micas contained in the mineral library used by the ASD TerraSpec Halo mineral identifier are organized by composition. The true white micas are represented by the potassium white micas muscovite and phengite, and the sodic white mica paragonite. The muscovite-phengite series is characterized by increasing substitution of Mg+2 or Fe+2 and Si+4 for Al+3. The illites are classified similarly. Thus, a shift in mineralogy from muscovite and/ or illite (labeled as K-Illite in the Halo mineral library) to paragonite and/or paragonitic illite (labeled as Na-Illite in the Halo mineral library) is indicative of increasingly sodic conditions. Similarly, a shift in mineralogy to phengite and/or phengitic illite (labeled as Mg-Illite in the Halo mineral library) is usually indicative of increasingly magnesium-rich conditions. The ASD TerraSpec Halo mineral identifier is able to track geothermal gradients both by detecting mineralogy and through the reporting of the ISM scalar. With increasing metamorphic grade, smectites convert to illites, which then covert to muscovite (and other true micas). Within the illites, subtle geothermal trends are easily detected and mapped using the Halo’s ISM scalar which tracks the illite dehydration process as the illite converts to true mica.

ω-texture analysis on metals Note d'application

The preferred orientation of crystallites constituting polycrystalline solid materials such as metals, ceramics or composites significantly influences the intrinsic properties of the respective solid. In material sciences this preferred orientation is commonly known as texture and describes the orientation of all crystallites in a sample with respect to a reference frame defined by the sample orientation. Using X-ray diffraction (XRD) the texture of a sample is usually measured using the approach developed by Schulz (1949) where the intensity of certain (hkl) reflections is measured as a function of the sample orientation. This is conventionally done by rotating (ϕ) and tilting the sample in axial plane (𝜒). Hence, point focus optics as well as an Eulerian cradle are required. In contrast to the conventional Schulz-technique, in the ω-texture approach different sample tilts in the equatorial plane are accessed by applying a ω-offsett to the goniometer while keeping the sample horizontal and rotating it in ϕ (Fig. 1). Therefore ω-texture allows measurements of complete pole figures in line focus without the need for an Eulerian cradle and point focus optics.

The whole analytical chain, including the weighing step in sample preparation prior to fusion, is of crucial importance to get precise and accurate x-ray fluorescence (XRF) results. As a (future) user of a Malvern Panalytical X-ray diffractometer you can perform almost all types of diffraction applications, depending on the configuration of your system. To get the best possible results out of your system, this book gives you an introduction to X-ray powder diffraction (XRPD or, mostly used, XRD) and provides helpful information. It gives a simple explanation of how a diffractometer works and how XRD analysis is done. This book is intended both for people new to the field of XRD analysis and for more experienced users to find new applications which might be helpful in daily work. This book avoids complex mathematical equations and understanding its contents only requires a basic knowledge of crystallography, mathematics and physics. It is not dedicated to specific diffractometer configurations or application areas, but aims to give a global overview of the various possibilities in XRD. Chapter 2 briefly explains XRD and its benefits. General application fields are presented. Chapter 3 gives an introduction to the physics and crystallography of XRD, while Chapter 4 describes how this is used for diffraction experiments. Chapter 5 explains the XRD data collection. This includes presentation of sample preparation and measurement procedures to get the best possible results. In Chapters 6-9 the procedures to perform qualitative and quantitative phase analysis are presented. Chapter 10 deals with non-ambient XRD and describes its methods and applications together with other techniques. Chapter 11 lists recommended literature for further information on XRD analysis. Since this book is intended to give an introduction to standard powder diffraction techniques, it does not cover other X-ray techniques like single crystal diffraction, high-resolution XRD of heteroepitaxial layers, or small-angle X-ray scattering (SAXS).

Since its establishment in 1920, Rand Refinery (close to Johannesburg, South Africa) has refined nearly 50,000 tons of gold – almost one third of all the gold mined worldwide. The molecular characteristics of a macromolecule are important parameters to be determined as they all can affect the material’s physical properties and behavior in a final product as well as during processing. In order to measure and evaluate these parameters (molecular weight and molecular weight distribution, molecular size and structure) size exclusion chromatography (SEC), or gel permeation chromatography (GPC) as it is also known, is one of the most useful tools that can be used. SEC is a liquid chromatography method that separates macromolecules according to their size (hydrodynamic volume) thus allowing further macromolecular characteristics to be derived from analysis of these separated fractions using various detectors. The number of parameters derivable from this technique strongly depends on the number and type of detector used or included in the SEC system. Colloidal gold is used in molecular recognition and targeting, electronics and nanotechnology. The appearance of the sample is determined by the size and shape of the gold nanoparticles present. Accurate and fast elemental analysis during the production process of titanium and its base materials is a critical requirement. Measurements were performed using an Axios FAST simultaneous X-ray fluorescence spectrometer equipped with a 4 kW Rh SST-mAX X-ray tube.

Volcanogenic massive sulphide deposits generally have zoned mineral alteration systems related to the hydrothermal circulation cells that led to their formation. In ideal situations it is possible to use field observations of such alteration to vector toward potential ore horizons. However, in many cases, such reasoning relies on subsequent detailed petrographic and lithogeochemical investigations; but the fine-grained and cryptic nature of much of the alteration may be an obstacle. A relatively new technique that may alleviate such delays is the use of portable reflectance spectrometers that provide visible/infrared spectroscopy (VIRS). This allows field identification of alteration minerals and compositional inferences that may constitute vectors to potential ore horizons. Visible/infrared spectroscopy methods were applied to two of the volcanogenic massive sulphide deposits in the Tulks Volcanic Belt of central Newfoundland, namely the Daniels Pond and Boomerang deposits. Both deposits have ubiquitous white mica and chlorite alteration, and mineralogical zonation in the field is poorly developed. Identification of such zonation is further hindered by the inherent lack of outcrop in the vicinity of the deposits. Results from both deposits, and regional rock sample suites, illustrate a distinct shift in the wavelengths of the Al-OH absorption feature in white micas from distal to proximal environments. The Al-OH absorption feature in white micas distal to ore is generally at >2000 nm (i.e., muscovite to phengite composition), whereas the absorption feature shifts to lower wavelengths (<2000 nm) in samples proximal to ore (i.e., paragonite composition). The VIRS data thus record subtle variations in alteration mineral chemistry that would otherwise be difficult to detect, and which may provide a directional indicator (vector) to favourable horizons.

This application note focuses on the analysis of heavy-metals, by Epsilon 5, in biological materials including vegetation, animal products and human hair. The chemical analysis of biological samples is an important measure of both human and environmental health. For the most part, this type of analysis has been done by wet-chemical, destructive techniques and essentially ignored by the field of XRF. However, the XRF technique provides high accuracy and precision with excellent detection limits (0.2 – 1 mg/kg).

Objectives: (i) to estimate mineralogical composition specifically clay mineralogy from the spectra, (ii) to qualitatively compare mineral concentrations calculated from XRF and XRD analyses, as well as mineral groups identified through SEM and microscopic petrography work with NIR results. NIR spectroscopy has been widely used in different scientific fields such as biology and medicine. However, it has been rarely used in mineral identification in finer sedimentary rocks specifically mudrocks. This work will allow us to verify the accuracy of this technique compared with XRF and XRD.

This application study was designed to demonstrate the performance of the Epsilon 5 EDXRF spectrometer and to show that the instrument meets the analytical challenge of analyzing Fe, Cu, Ni, As, Cd and Pb in edible oils. The long term stability and precision of the instrument is also demonstrated. The refining processes for products such as edible oils require the addition of catalysts in the correct proportions to optimize the processing reactions. These catalysts also contain elements such as Ni and Fe that must be removed from the oils during the later stages of the refining process.

In order to demonstrate the capability of the Epsilon 5 for the analysis of Pt, Pd and Rh in automotive catalysts and recycled slag materials, samples with concentrations of these elements previously determined by fire assay were used to create calibrations on the Epsilon 5. Accurate determination of platinum (Pt), palladium (Pd) and rhodium (Rh) concentrations in automotive catalysts is critical for developers and producers of automotive catalysts as well as those recycling the end products to reclaim these precious metals.

Operational impacts of Glen Canyon Dam on downstream resources of the Colorado River ecosystem have been the subject of intense study and discussions among adaptive management stakeholders that include Federal, State and tribal entities, as well as recreational businesses, environmentalists, power companies, and others ( U.S. Bureau of Reclamation 1995, Schmidt et al. 1998). The Grand Canyon Monitoring and Research Center (GCMRC within the U.S. Department of Interior) has identified the following key objectives for long-term monitoring of vital components of the ecosystem between Lakes Powell and Mead, while providing stakeholders with information for decision making: 1) lowest possible impact on critical resources; 2) highest possible return of integrated scientific information; and 3) long-term, repeatable data collection. Airborne hyperspectral image data are being considered as a possible long-term monitoring tool, which hold the promise of being able to fulfill the above requirements. The present study is a pilot project which is intended to explore the extent and variety of information, relevant for long-term monitoring, that can be extracted from images of both high spectral and spatial resolution. The Colorado River ecosystem in Glen Canyon and Grand Canyon poses special challenges for remote monitoring with respect to logistics required to support field-based studies and relative to spatial scales at which monitoring is needed. The narrow corridor of the river is confined by tall vertical cliffs, yet provides a habitat for a great variety of both native and non-native plant species. Riparian vegetation consists of stands that often cover only very small areas. Often, these stands consist of intermixed native and non-native species, and spectral signatures of the various plants sometimes display only subtle differences. Adequate change detection mapping requires both high spectral and high spatial resolution, as well as advanced processing algorithms that can detect subtle spectral differences. Change detection for evolving sand bars, rapids and debris fans is a critical part of the GCMRC's long-term monitoring program. Additionally, changes in the aquatic ecosystem's food base, such as abundance and distribution of algae and other benthic organisms are of great interest. Presently, the need to lower river stage for 2- 3 days during annual monitoring overflights for conventional aerial photography and traditional surface measurements introduces artificial and undesirable impacts on the ecosystem (e.g., Blinn et al. 1999). With hyperspectral imagery, we expect to map riparian vegetation, terrestrial sediment deposits, as well as sand bars and benthic organisms through some depth of water, obtaining greater compositional information than can be obtained through the use of aerial photography.

Recent evidence suggests that the chemical diversity of humid tropical forests exceeds that of all other terrestrial ecosystems combined, with cascading effects on spectroscopic patterns of tropical canopies acquired from new airborne and future space-based imaging spectrometers. To address this new frontier in spectroscopy, I developed the Carnegie Spectranomics Project (CSP), which seeks to quantify and understand linkages between chemical, spectral, and taxonomic patterns among tropical forest species. The CSP has collected and analyzed more than 3,500 species from C. and S. America, Africa, Asia, and the Pacific, showing that high-fidelity spectral patterns among species are driven by evolutionary processes that maximize chemical variation at site and global scales. This talk will highlight results from the CSP, and point to upcoming mapping flights using airborne high-fidelity imaging spectroscopy.

Soil reflectance spectra are responsive to a number of soil characteristics, including water content, mineral content, the presence/absence of organic material and the roughness or coarseness of the soil. To some extent, these components alter the spectral reflectance of soils independently, but there also appears to be some interaction among these effects. The model presented in this paper is a preliminary and relatively simple representation using the absorption and refractive properties of water along with absorption due to dissolved organic material to modifying the reflectance spectrum of a dry soil. The model is an attempt to capture the variability in reflectivity in a way that will lend itself to inversion. The model represents the effect of water in two ways: a thin layer of water on the soil particles alters the reflectance of the soil largely by reducing the magnitude of the reflectance; second, where water provides a more significant optical path (presumably with interstitial soil water) the spectral absorption characteristics of water dominate with some modification by the presumed presence of organic material dissolved in the water. Organic material can be expected to introduce absorption effects in the ultraviolet and blue portions of the spectrum. Taken together, these properties appear to account for a major portion of the spectral changes in soil reflectance; however, there are interesting and consistent discrepancies, at least in the current formulation of the model. The model and its characterization of soil reflectance is described and discussed.

It is important to understand the patterns of species distributions as they determine the functioning of ecosystems across a wide range of time and space scales from biogeochemical cycles and partitioning of the energy budget, to changes in biodiversity, habitat characteristics, and trophic structures. However, even high spatial resolution imagery has not been sufficient to identify and map individual species. Mapping large monotypic stands is challenging, unless the spectral properties of the species stand out from their neighbors due to differences in growth form or timing of phenological activity. Where it is possible, mapping of individual species is often restricted to limited areas of ecologically similar conditions, or restricted to analysis of single datasets and do not consider how species identification changes with season, location, and environmental conditions. In the past decade, with the development of environmental spectroscopy, both from field spectrometers and airborne imaging spectrometers, has allowed progress in identifying individual species from remote sensing data. There is now a growing literature on identifying species in many case studies. However, use of environmental spectroscopy for species identification needs understanding at a more fundamental level, especially the development of generalized methodologies and rules for detection and mapping, which is an area of active research today. Conceptually, we have yet to resolve how to identify unique spectral signatures for the estimated 400,000 extant plant species or groups of species (i.e., functional types). In contrast to geologic minerals which are often spectrally distinct, all land plants share a common basic metabolism and biochemistry, making identification uncertain because of the interactions with environmental conditions and phenological stages, in addition to the characteristic properties of individual species. These issues will be explored using examples from a wide range of habitats and site conditions, to develop a robust methodology.

In this application study, one half of a small shell was investigated in detail to explore its shape, mineral composition and special features. The example demonstrates the powerful combination of X-ray diffraction, X-ray imaging and Computed Tomography. With X-ray Computed Tomography(CT), a full 3D image of samples can be obtained non-invasively, allowing visualization and analysis of the full 3D microstructure.

The combination of phase analysis, density and microstructure using an X-ray platform allows to correlate meso- to nanoscopic features with material properties and to estimate the durability of concrete.
