Use DSC to select antibody with lowest propensity to aggregate. An important consideration during the engineering process of an antibody is both the inherent conformational stability of the antibody as well as long-term stability. Understanding these stability parameters and what trends they follow will be critical throughout the biotherapeutic development. Differential scanning calorimetry (DSC) is a potent tool in probing these qualities. As a stability indicating assay, DSC provides insights for the selection of stable protein construct candidates. In the case of multi-domain proteins, DSC uniquely allows the investigation of the stability of individual domains from multi-domain proteins like antibodies and Fc-conjugated proteins. In this application note, we compare stability data predicted by DSC of different engineered antibodies with SEC-HPLC data from accelerated stability studies. For the engineered proteins studied, there is a correlation between decreased thermostabilty based on DSC data, and greater aggregation formation during accelerated stability studies. This study demonstrates how DSC can be a useful tool in stability screening of engineered proteins.

See how DSC can be used to assess the effects of polyol excipients on antibodies. Differential scanning calorimetry was used to monitor polyol-induced increases in thermal stability across seven monoclonal antibodies. The increases in thermal stability were measured as a function of polyol concentration and orientation of hydroxyl groups. It was found that increases in thermal stability were linear with respect to alcohol concentration. Further, the increase in thermal stability was dependant on the transition temperature; antibodies with lower transition temperatures exhibited a greater increase due to the addition of polyols than antibodies with a higher transition temperature.

Nanoparticles can be difficult to produce, and are often synthesized in small quantities and dispersed in expensive solvents. To improve its shelf-life, milk undergoes a homogenization process. This process produces fat globules of a uniform, small size. During the homogenization process the size range of the fat globules is reduced from 0.1-15 μm in unprocessed milk to 1-2 μm in homogenized milk [1]. These smaller globules cannot form large enough clusters for creaming to occur, increasing the shelf-life of the milk. The creaming process is governed by Stokes’ Law and the relative densities of the fat and the other fluid components of the milk. Milk also contains casein micelles, in the size range of 0.05-0.25 μm. The micelles play a role in stabilizing the fat globules, especially after the homogenization process. The size of the fat globules and the proportion of free casein micelles are important parameters for monitoring the homogenization process and can be measured simultaneously by laser diffraction.

In this application note, we describe a novel method to rapidly measure the size and concentration of Adeno-Associated Virus (AAV), utilizing the Nanoparticle Tracking Analysis (NTA) technique and a gold labeling method. By labeling AAV capsids with small gold nanoparticles, we were able to increase the amount of light scattered by each capsid, enabling rapid NTA characterization to determine the virus concentration and aggregation state. NTA characterization of AAV was previously impossible without gold labeling.

IntroductionSmoothies have been around for the best part of a century although it was not until the development, commercialization and evolution of the blender between 1920 and 1950 that home-made smoothies (in the modern sense) became possible. Their popularity was further accelerated by the health food movement of the 1960s and more recently the commercialization of dedicated smoothie makers such as the NutriBullet and Nutri Ninja combined with a trend towards natural and nutritious foods have revolutionized the industry. In the Oxford English dictionary, a smoothie is defined as ‘a smooth, thick drink made with puréed fresh fruit and yogurt, ice cream, or milk’. However, there does not seem to be any fixed rules to what should go into a smoothie with common additions being vegetables, crushed ice, nuts, seeds and extra juice or water. What is widely accepted is that a smoothie should be as smooth and easy to consume as possible, aspects which are dependent on the particle size of the blended components and the viscosity of the dispersion. While the viscosity of the blend can be controlled most easily by the choice of ingredients and amount of water used, the particle size of a given set of blended ingredients is directly dependent on the performance of the blender. This is often the main criteria that consumers will use to judge the quality of a product – the taste test. Liposomes have been widely used as model membranes and in drug delivery, as their basic properties can easily be varied to target desired properties (lipid composition, concentration, water content, media (different buffers, varying ionic strength, pH, etc.) as well as lamellarity (unilamellar, large or small (Large Unilamellar Vesicles, LUVs, and Small Unilamellar Vesicles, SUVs), and multilamellar vesicles, MLVs)). In these studies, one must characterize the pure liposomes as well as their mixtures with other molecules (proteins, peptides, drugs), to ascertain the influence of the added component in the liposome phase behavior. DSC is a fundamental technique for the characterization of membrane behavior, providing all thermodynamic parameters for temperature-induced transitions. Furthermore, for mixtures of membranes with other components (drugs, proteins, peptides, polymers, etc.) DSC represents a first-line screening method, providing information on the effect(s) of the added components on membrane behavior.

Poly(lactic acid) is a biodegradable polymer derived from natural resources (corn starch), that has received significant attention in recent years. It is one of the most prevalent biodegradable polymers in the market place due to availability, low cost of production, and when derived from natural sustainable sources it is a truly renewable polymer. PLA is often used as a copolymer with glycolic acid to form PLGA and is routinely used with varying compositions of lactic and glycolic acids. Being a versatile polymer, it is used in a wide variety of applications from additive manufacture (3D printing) to disposable cutlery, biodegradable sutures, drug delivery, or as biodegradable packaging. It is widely recognized that the bulk properties of polymers are strongly dependent on their molecular properties. Most commonly, the strongest determinant of a polymer’s strength is assumed to be its molecular weight. However, in copolymers such as PLGA, it is also likely that copolymer composition will also strongly affect those properties. This application note uses two Malvern technologies to explore the relationship between the molecular and bulk properties of PLA and PLGA. Multi-detector GPC is used to measure molecular weight and intrinsic viscosity (which is dependent on copolymer composition), while rotational rheology is used to study melt-viscosity. The two results are then compared to study which molecular properties best correlate with melt-viscosity.

Polymer branching can have an enormous effect on the physical properties of the final synthesized polymer. Branching can disrupt crystallinity, increase chain entanglements, increase single chain density, and affect a variety of polymer properties such as glass transition temperature, modulus, toughness, etc. Understanding polymer branching and its effects on a polymer’s end-use properties can help avoid failures and improve specifications. In this article we take a look at how, from the starting point of an appropriate wet sensor, laser diffraction systems can be engineered to deliver to the needs of different process applications. A case study involving the control of an emulsification unit for the production of oil and water emulsions illustrates what can be achieved and the benefits delivered.

In this article we look at what zeta potential is and the potential role that on-line zeta potential monitoring can play in optimizing the water treatment process, specifically flocculation control. The applicability of zeta potential is illustrated via the experiences of Aurora Water, a utility responsible for the production of water treatment for the city of Aurora in Colorado, USA. One Aurora Water treatment facility is using zeta potential as both an off- and on-line tool for monitoring and controlling the water treatment process. Results show how on-line technology can reduce chemical usage costs and simplify process operation, delivering further gains in energy consumption.

The repeatability of the method is such that it allows accurate process control of cement production. The main component in industrial clinkers for the cement production is alite with the ideal composition Ca3SiO5, also written as C3S in the short cement notation. This compound can occur in crystallographic polymorphs. Their formation depends mainly on the burning conditions (temperature, burning time, cooling rates) and minor or trace elements in the crystal structure, introduced by the raw material and fuels. IntroductionDynamic Light Scattering (DLS), sometimes referred to as Photon Correlation Spectroscopy or Quasi-Elastic Light Scattering, is a technique classically used for measuring the size of particles typically in the sub-micron region, dispersed in a liquid. The sensitivity of some modern systems is such that it can also now be used to measure the size of macromolecules in solution, e.g. proteins Brownian MotionDLS measures Brownian motion and relates this to the size of the particles. Brownian motion is the random movement of particles due to the bombardment by the solvent molecules that surround them. The larger the particle or molecule, the slower the Brownian motion will be. Smaller particles are "kicked" further by the solvent molecules and move more rapidly. An accurately known temperature is necessary for DLS because knowledge of the viscosity is required (because the viscosity of a liquid is related to its temperature). The temperature also needs to be stable, otherwise convection currents in the sample will cause non-random movements that will ruin the correct interpretation of size. The velocity of the Brownian motion is defined by a property known as the translational diffusion coefficient (usually given the symbol, D). The Hydrodynamic DiameterThe size of a particle is calculated from the translational diffusion coefficient by using the Stokes-Einstein equation; where:- d(H) = hydrodynamic diameter D = translational diffusion coefficient k = Boltzmann's constant T = absolute temperature η = viscosity Note that the diameter that is measured in DLS is a value that refers to how a particle diffuses within a fluid so it is referred to as a hydrodynamic diameter. The diameter that is obtained by this technique is the diameter of a sphere that has the same translational diffusion coefficient as the particle. The particle translational diffusion coefficient will depend not only on the size of the particle "core", but also on any surface structure that will affect the diffusion speed, as well as the concentration and type of ions in the medium. Factors that can affect the diffusion speed discussed in the following sections. Ionic Strength of MediumThe ions in the medium and the total ionic concentration can affect the particle diffusion speed by changing the thickness of the electric double layer which is called the Debye length (K-1). Thus a low conductivity medium will produce an extended double layer of ions around the particle, reducing the diffusion speed and resulting in a larger, apparent hydrodynamic diameter. Conversely, higher conductivity media will suppress the electrical double layer reducing the measured hydrodynamic diameter. The performance of a DLS instrument is normally verified by measurement of a suitable polystyrene latex standard. If the standard needs to be diluted prior to measurement, then dilution in an appropriate medium is important. The International Standard on DLS (ISO22412:2008) says that dilution of any polystyrene standard should be made in 10mM NaCl. This concentration of salt will suppress the electrical double layer and ensure that the hydrodynamic diameter reported will be the same as the hydrodynamic diameter on the certificate or the expected diameter. For a 60nm monodisperse latex, dispersion in demineralised water rather than 10mM NaCl can result in 15% higher size reported. Colloidal gold is used in molecular recognition and targeting, electronics and nanotechnology. The appearance of the sample is determined by the size and shape of the gold nanoparticles present. Sizes less than 5 nm are yellowish in color, 5 nm to 20 nm tend to be reddish whereas particles greater than 100 nm are bluish in appearance. The molecular characteristics of a macromolecule are important parameters to be determined as they all can affect the material’s physical properties and behavior in a final product as well as during processing. In order to measure and evaluate these parameters (molecular weight and molecular weight distribution, molecular size and structure) size exclusion chromatography (SEC), or gel permeation chromatography (GPC) as it is also known, is one of the most useful tools that can be used. SEC is a liquid chromatography method that separates macromolecules according to their size (hydrodynamic volume) thus allowing further macromolecular characteristics to be derived from analysis of these separated fractions using various detectors. The number of parameters derivable from this technique strongly depends on the number and type of detector used or included in the SEC system. Additive Manufacturing (AM) is a term used to describe advanced production methods that convert a 3D model into a real part by the layer upon layer addition of material. Compared to traditional subtractive methods, AM is ideal for rapid prototyping and can enable the faster, cheaper and more efficient production of complex parts such as customized medical components or engine parts, often with improved strength and durability. As illustrated in Figure 1, Powder Bed Fusion (PBF) processes involve the controlled application of a thin layer of powder, followed by the use of a thermal source such as a laser to selectively fuse the powder particles together. This is repeated for each layer and the finished part is then extracted from the powder cake. The unused powder can then be recycled for the next part, usually by blending it with a certain amount of virgin powder. Thermoplastic polymers such as polyamide 12 (known commonly as nylon 12 or PA12) are well suited for PBF processes as they can be processed reliably due to their semi-crystalline nature providing a distinct melting point. Figure 1. Diagram showing a typical Powder Bed Fusion (PBF) process The ability to recycle unused powder gives PBF processes the potential to be very resource-efficient, but this efficiency depends strongly on the condition of the recycled powder, as it risks a deterioration in final part quality. This Application Note looks at how size exclusion chromatography (SEC), which determines the macromolecular characteristics of the feed material, and automated image analysis, which provides morphological information for the particles, can provide valuable insights to this active area of research. The process of identifying small molecule hits suitable for subsequent optimization into leads, and eventually candidates is a critical stage of the drug discovery workflow. Time-dependent inhibition (TDI) of CYP metabolism can generally be classified into three types: reversible, quasi-irreversible and irreversible. In general, TDI results from irreversible covalent binding or quasi-irreversible non-covalent tight binding of a chemically reactive intermediate to the enzyme that catalyses its formation. This results in loss of enzyme function and can cause clinically relevant drug-drug interactions (DDI). In some cases, reversible inhibition of a metabolite(s) generated in situ could give rise to TDI. TDI inhibitory effect lasts longer than reversible inhibition and persists after elimination of the parent drug from the body because CYP activity can only recover with new enzyme synthesis.

WEE1 regulates the G2/M cell cycle checkpoint via phosphorylation of CDK1 (aka Cdc2) at Tyr15, which inhibits CDK1/cyclin B kinase activity. The tumor microenvironment (TME) supports the development of pro-tumoral macrophages. These Tumour Associated Macrophages (TAMs), resemble M2-macrophages and are characterized by an immune-suppressive phenotype. In the TME, TAMs can induce proliferation and survival of tumor cells, facilitate angiogenesis, and suppress anti-tumor immune responses via expression of co-inhibitory molecules such as PD-L1 and cytokines such as IL-10 or TGF-β1. Therefore, TAMs are a highly attractive target for innovative cancer immunotherapies. Understanding whether candidate immunotherapies can reverse TAM-mediated immune suppression or reprogram TAMs to a pro-inflammatory phenotype is key to the development of effective cancer immunotherapy. Here we outline a suite of macrophage assays that can be adopted to determine whether candidate therapeutics can reverse human M2 macrophage-mediated immune suppression and repolarise M2 macrophages.

Compounds that are unstable in aqueous media may not be able to be maintained at therapeutically effective concentrations in vivo, and therefore have reduced chances of becoming a successful drug candidate. In dosing solutions, drugs may be prone to chemical degradation which may be caused by a variety of mechanisms: hydrolysis, oxidation, light-catalyzed degradation and others. In the gastrointestinal tract, drugs can also be subject to chemical degradation under different conditions. It is therefore important to determine the stability of a compound prior to its administration to animals for pharmacokinetic evaluation. Stability testing in buffer solutions at room temperature or 37ºC at acidic, neutral and basic pHs in early drug discovery allows timely identification of potentially weak candidates.

Limestone is the principal source of CaCO3, which is used as raw material in a wide range of applications. It is used as an additive in steel making, as main component for cement and, when purified, it even can be used as supplement for calciumrich animal feed or pharmaceutical fillers. Al2O3, SiO2, MgO and Fe2O3 are further considered as the main compounds of limestone, and are important in the use of limestone. The grade and application of the raw material is determined by the ratio between these main compounds, as well as the presence of other minor and trace compounds, like MnO, P2O5 and Pb. Dynamic light scattering has been vastly explored as a tool for micelles characterization, not only for their size and stability, but also to determine the critical micellization concentration (CMC) and temperature (CMT)1-3. As a result of the unimers aggregation, as a function of the temperature for the case of this study, an increase in the scattered intensity is expected due to larger particles scattering substantially more photons. It’s been reported that for light-scattering techniques to be used reliably as CMC or CMT determination techniques, long correlation times are needed 1,4,5 which make this time-consuming and hence it’s not usually considered as a primary technique. We suggest in this application note, that DLS can be used in conjugation with other techniques to determine the CMT in a highly reproducible and time-efficient manner.

Whilst being able to measure particles of below 1 nm size, DLS is preferentially sensitive to larger particles due to the 6th power relationship between particle radius and scattering intensity. This means that sample preparation typically needs to be scrupulous, especially for low scattering samples such a proteins and biological molecules. The contribution to contaminants such as dust and aggregates can be mitigated by filtering, however this may not always be practical or possible depending on the volume and fragility of the sample. Filtration of samples can also constitute a financial burden, both in terms of additional sample preparation time and consumables costs. This application note introduces a new DLS data capture process called Adaptive Correlation which helps address these issues. Adaptive Correlation uses a statistically driven approach to produce the best correlation data, which in turn gives more reliable DLS size data. It can reduce the need for filtering and give added confidence in you DLS results.

Liposomes are nowadays frequently used as drug delivery systems, and as such requires that they are well characterized for manufacturing and batch release. This characterization includes their size and size distribution. For these parameters, dynamic light scattering (DLS) is commonly used to characterize size and size distribution and also nanoparticle tracking analysis (NTA) has previously been shown to be useful for measuring liposomes size and concentration.1 This application note will demonstrate how the multi-angle DLS (MADLS®) based particle concentration measurement in the Zetasizer Ultra is capable of measuring the particle concentration as well as size and size distribution of a range of dilutions of a liposome sample. Polyvinyl chloride (PVC) is a common polymer used in countless applications from pipes, to flooring, and electrical insulation. As one of the world’s most widely produced polymers, it is very much a commodity polymer where tight margins mean little room for detailed analysis. However, even in this case, high level analytical tools can be used to characterize the polymer for appropriate grading. For example, the glossiness of a PVC sample has a significant effect on its finish and grade, and for which applications it can be used. Applying appropriate characterization tools can help grade different PVC batches, assigning them to appropriate applications to maximize the value of high quality batches, saving lower grades for others. In this application note, four PVC samples of known differences in glossiness were measured with both multi-detector GPC and rotational rheometry. The results both explain the differences in glossiness and the likely cause of those differences.

Multi-angle Dynamic Light Scattering (MADLS) offers improvements in particle sizing resolution, sensitivity and accuracy compared to traditional single angle Dynamic Light Scattering (DLS). By combining measurement data, sequentially collected, from three measurement angles it can provide improvements in resolution, typically from 3:1 to 2:1. It also provides improvements in accuracy by reducing signal to noise ratio, allowing lower smoothing coefficients to be used, and by reducing angular dependence of the final particle size result, as illustrated in the following Mie solution plot for a single protein macromolecule measured at 45, 90, and 173 degree scattering angles. This white paper provides a technical discussion on how MADLS is implemented in the Malvern Panalytical Zetasizer Ultra system and the advantages of combining data from multiple measurement angles as well as the practical considerations when employing MADLS on your nanomaterial and biomolecular samples.
