Amorphous solid dispersions (ASDs) present a formulation option for poorly soluble drugs intended for oral administration. They work by molecularly dispersing the drug substance in a water-soluble polymer, thereby disrupting the intermolecular bonds that would otherwise form a crystal. Once the polymer dissolves, the drug molecule is also in solution and therefore available for absorption. However, not all drug substances are compatible with the polymer, the released molecule is in a transient supersaturated state, and the long-term stability of ASDs is not guaranteed.
