Lipid nanoparticles (LNPs) hold great potential in treating, curing, and preventing a variety of challenging conditions, from genetic disorders to cancer. These vectors enable efficient delivery of therapeutic drugs such as RNA. Unlike virus vector manufacturing, LNPs can be produced using a cell-free production process, allowing for rapid scale-up. However, due to the complexity of their structure, LNP therapy presents challenges in development and manufacturing analysis. Accurate measurement and careful control of various key characteristics are required to evaluate the stability of LNPs. In addition, guidance and information provision for product design, production process optimization, and release specifications are necessary.
Traditional methods used to characterize the properties of conventional therapies, such as monoclonal antibody therapy, often do not suit the complexity and pace of new LNP-based therapies. Therefore, there is an urgent need for analytical packages that combine cross-disciplinary and multifaceted evaluation methods to complement traditional techniques.
In this seminar, we will introduce analytical instruments (DLS/MADLS, NTA/PTA, ELS, SEC-SLS) used to evaluate key LNP factors such as size, polydispersity, concentration, surface charge, and therapeutic drug information, including measurement principles and examples. We will also introduce the newly developed Sample Assistant accessory for automation and unmanned operation, essential functions for analytical instruments. By using the Sample Assistant, the previously tedious sample exchange during DLS measurements becomes automated, improving device operating rates and enhancing operator productivity by freeing up time for other tasks.
