Grating-Coupled Interferometry (GCI): a new tool for label-free molecular interaction analysis of VLPs

By combining best-in-class sensitivity with crude sample robustness, normally only achieved with plate-based assays and a very fast transition capability, the Creoptix WAVE is revolutionizing the study of molecular interactions and changing the world of drug discovery.

Engineered around Grating-Coupled Interferometry (GCI), the WAVE system offers label-free quantification across the broadest range of compounds and sample types. The system can accurately characterize weak binder kinetics with off-rates of 5 sec-1 as well as high affinity antibodies. The WAVE’s microfluidics sustain pure serum, cell supernatant, larger particles and even crude membrane and VLP extracts.

The recently launched second generation of the WAVE system allows the characterization of a wide spectrum of analytes, ranging from small molecules to multiprotein complexes, large particles and even bacteria. In addition, the system is capable of detecting interactions within a remarkably wide affinity range (from mM to pM).

Here, we show examples of GCI applications in the field of VLPs and protein membrane extracts, including VLP capture from crude extracts and antibody binding kinetics on VLPs from crude preparations. Due to the comparably low amount of ligand and analyte needed, GCI is an excellent alternative for experiments where key components are scarce.
