The PEG monolayer is an essential component of most mRNA-LNP formulations. PEG provides multiple vital functions, such as minimizing protein absorption and extending circulation times to help the vector protect and deliver nucleic acid therapeutic cargo to the desired target. However, the PEG monolayer can reduce the electrostatic repulsive forces between particles, resulting in lower colloidal stability of the drug product.

In this webinar, we will explore the importance of measuring LNP charge and how to measure zeta potential using Electrophoretic Light Scattering (ELS). Topics to be covered include ELS theory, what LNP components are charge carriers, how buffer excipients influence particle charge, and best practices when developing a zeta potential method.

This presentation is intended for any scientist involved in the development of LNP-based therapeutics, including LNP formulation scientists, LNP analytical development scientists, and LNP discovery scientists.

Speakers

  • Jonathan Mehtala, PhD - Team leader, Biologics Development, Malvern Panalytical

Vaccines And Nucleic Acid-Based Therapies Webinar Series

In this three-part webinar series, we will explore several areas critical to LNP vaccine and gene therapy development.

Topics include an overview of LNP structure and formulation, the common and advanced analytics used to characterize LNPs, and what LNP physicochemical properties are measured.

We will discuss in-depth the light scattering analytics used to measure LNP size, polydispersity, charge, and particle concentration.
