Automated methods with OMNISEC

In my last post I described the steps I take when getting OMNISEC ready for analysis.  As I suggested at the end of that post, OMNISEC can automate many of those steps!  In this post, I’ll describe how I use automated methods with OMNISEC to help get my analyses ready.  At the end of the post is a video demonstration so you can see the automated methods in action for yourself! 

Types of automated methods

There are two types of automated methods: Instrument setup methods and Experiment settings methods.  An Instrument Setup Method controls instrument parameters such as flow rate, system temperatures, detector purges, and more.  An Experiment Settings Method allows you to set the options available under the Run Conditions on the Sequence tab, such as run length, column set and solvent descriptions, and more.

How to access the automated methods

You can easily access the automated methods using the Browse database buttons in the top left section on the Sequence tab, as shown below (1). Alternatively, you can add to and edit the automated method databases by going through the Tools section on the main toolbar.  There, you’ll see the databases for Instrument Setup and Experiment Settings Methods.

Creating an Instrument Setup Method

To create a new Instrument Setup Method, open the Instrument Setup Method database and click the Add button near the bottom right of the window.  In a new pop-up window, you can set which parameters you want to customize.  If you have been using the instrument already, the easiest way to populate an Instrument setup method is to click the Copy current instrument setup button in the bottom left. The current instrument settings will populate and then you can make additional adjustments as desired.

If the Set at Runtime? box is checked, the software will allow you to enter the desired setting at the start of the sequence, when you press Start.

The steps of an example Instrument Setup Method are listed below. It is important to remember that you ensure you have enough mobile phase to account for the Pre, Mid, and Post Setup Delays, as well as any detector purges. Please note that currently the detectors are purged three times each, but future software versions (v11.37+) will allow you to set the number of purges and let you choose if you want to zero the detectors.

  • Baseline monitor starts
  • Pump flow rates & temperature settings applied
  • Laser & UV-Vis PDA bulbs turned on
  • Autosampler primed
  • Mid-set up delay for baseline stabilization
  • RI, DP, & IP purges
  • RI detector zeroed
  • Post-set up delay for baseline stabilization
  • Sequence started

Creating an Experiment Settings Method

Similar to the Instrument Setup Methods, Experiment Settings Methods can be created by opening the Experiment Settings Method Editor and clicking the Copy Experiment Settings Setup button in the bottom left.

The Run length and post-run settings are the most important parameters, as they will affect how the sample(s) is run. The Solvent and Column Set Descriptions are for record keeping purposes and can be edited after the fact, if necessary.

Final thoughts

In conclusion, I hope the automated methods in OMNISEC make it easier for you to get your system ready and analyses started.  The Instrument and Experiment Setup Methods will handle all of your system preparation – all you have to do is click start and walk away!
