Gene therapy is an area of rapid growth within the biopharmaceutical sector which uses genes to treat or prevent a disorder. The vectors which are used in gene therapy can be viral, non-viral and engineered vectors. In this work, we are focusing on a single viral vector used in gene therapy, recombinant adeno associated viruses (rAAV). In this application note rapid, accurate, and robust quantitative measurements of viral titer and full capsid content for AAV samples will be described. This study will demonstrate the capabilities of OMNISEC and ZS Ultra for analyses of rAAV5 samples at different levels of genome loading and sample heterogeneity. Screening for Aluminium Lakes: Analysis of Colorants in food Практическая аннотация

Aluminum food lakes are oil dispersible colorants manufactured by combining food dyes with alumina. Alumina absorbs the water-soluble colors and acts as a carrier vehicle for the dye. Aluminum consumption is a potential food safety concern. For example, the European Food Safety Authority (EFSA), in its opinion of 22 May 2008 (2) recommended lowering the tolerable weekly intake (TWI) for aluminum to 1 mg/kg body weight/week. Therefore quantification of aluminum in food dyes provides a way to monitor the potential for human intake. One of the challenges of developing a test protocol for dynamic light scattering measurements is getting the concentration of the sample right. Too low, and our scattering measurement fluctuates erratically as we become sensitive to particles entering and leaving the detection volume. The molecular characteristics of a macromolecule are important parameters to be determined as they all can affect the material’s physical properties and behavior in a final product as well as during processing. In order to measure and evaluate these parameters (molecular weight and molecular weight distribution, molecular size and structure) size exclusion chromatography (SEC), or gel permeation chromatography (GPC) as it is also known, is one of the most useful tools that can be used. SEC is a liquid chromatography method that separates macromolecules according to their size (hydrodynamic volume) thus allowing further macromolecular characteristics to be derived from analysis of these separated fractions using various detectors. The number of parameters derivable from this technique strongly depends on the number and type of detector used or included in the SEC system. Now you see me and now you don’t: GPC analysis of polysiloxanes Практическая аннотация

IntroductionGel permeation chromatography (GPC) is at the heart of polymer characterization. This characterization technique is implemented throughout the life of the material.This can include after initial polymerization, before and after purification, after post-polymerization modification, in the final formulation, after aging, failure and/or degradation. The primary focus of analyzing polymers using GPC is to characterize their molecular weights and molar mass distributions. There are many motivations behind determining the molecular weight including predicting how the products' properties will be affected and determining how well a reaction went. In addition to molecular weight, scientists also want to know about changes to the sample’s structure, branching, size, solution viscosity and compositional changes; the OMNISEC multi-detection GPC system provides this information with the highest level of sensitivity. The OMNISEC uses a combination of refractive index, light scattering, UV/vis spectrometer and viscometer detectors to provide a far more complete characterization than the relative molecular weight results provided by using only RI detection. Polysiloxanes are an interesting material to analyze by GPC, as if one tries to analyze the most common polysiloxane, polydimethylsiloxane (PDMS), in the standard organic solvent for GPC, THF, the peaks in the RI are extremely small and none existent in the light scattering detectors. However, when analyzed in toluene the PDMS signals in both of these detectors are intense. This is due to the refractive index increment (dn/dc) of the material (for further details on the dn/dc value please read this interesting blog post) Polysiloxanes are widely used for many applications as they are very chemically stable, unreactive, hydrophobic, have good electrical insulation and can form a variety of material types, such as elastomers, gels, lubricants, foams, and adhesives. Isothermal Titration Calorimetry (ITC) is a powerful and versatile technique that is widely used for measuring the binding affinity and thermodynamics of equilibrium association reactions. ITC is also an accepted, universal label-free assay technique that can be applied to any enzyme-catalyzed system, provided that the reaction is associated with a change in enthalpy – see the companion whitepaper and other review articles [1-8]. Using Isothermal Titration Calorimetry to Characterize Enzyme Kinetics Тематическая публикация

Enzymes are proteins that function as biological catalysts, which play crucial roles in the biochemical processes that occur in living organisms. Understanding how enzymes function, and how to activate or inhibit their activity, is a core research focus for biochemists. Since many drug targets are enzymes, the development of new therapies requires understanding of how the target enzyme binds and catalyzes its natural substrate. Several drugs are therapeutic enzymes, which are injected into the patient to treat genetic disorders characterized by missing or defective enzymes. Enzymes are also important in other industries including food science, biofuels, and detergents. Although enzymes were discovered in the mid-1800s, isolated and purified since the early 20th century, and cloned and expressed in recombinant systems since the 1970s, there is a continued need for efficient and detailed enzyme analysis to exploit the potential of enzyme-driven catalytic reactions. New enzymes are also being discovered. Reliable enzyme kinetics data are crucial to understand and control enzyme effectiveness and create next generation drugs. Isothermal titration calorimetry (ITC) techniques have been successfully applied to study enzyme kinetics and inhibition. ITC is a well-established, versatile technique that is widely used for measuring reaction thermodynamics. Microplastics, usually defined as pieces of plastic that are less than 5 mm in size, are a global environmental and potential health issue. Morphologically-Directed Raman Spectroscopy (MDRS®) can help research into the origin and fate of microplastics by enabling component specific data to be obtained from multi-component samples. A single automated measurement provides particle size and shape data for each different microplastic type chemically identified using Raman spectroscopy. Next to the information about the composition of a raw ore, heavy mineral concentrate or titania slag it provides useful information by the quantitative determination of the crystalline phases and the amorphous content. The analysis of the phases can help to optimize the concentration of ores and reduction of ilmenite concentrate. Traditionally quality control of heavy mineral concentrates and titania slag relies mainly on elemental, chemical, gravimetrical and magnetic analysis. Since the efficiency of concentration and reduction depends on the content of the different titanium and iron phases, such as ilmenite FeTiO3, rutile TiO2, anatase TiO2 or the various titanium oxides with different oxidation stages, a fast and direct analysis of the phases is required.The three case studies of this paper demonstrate how XRD and cluster analysis can be used for process optimization and quality control.The potential of XRD as an automatable, reliable tool, useful in the characterization of heavy mineral concentrates and titania slag will be demonstrated.

We present the use of Multi-angle Dynamic Light Scattering (MADLS®) for the measurement of nanoparticle concentration. We describe the theory of the method and its application to nanoparticles made of gold, silica and polystyrene, with diameters ranging from 30 to 400 nm, and demonstrate some of the limitations with particles of sizes 500 nm and above. We evaluate the method accuracy, linearity and reproducibility, as well as the operational nanoparticle concentration and size range. We show that the concentration working range depends on the material’s optical properties, size and concentration. Here it is shown that the level of accuracy that can be expected for the concentration of particles is typically within 50% of the nominal value across a range of materials and sizes and, for some samples, within 20%. The repeatability of the measurements, in terms of relative standard deviation, is typically below 30%. A linearity of within 40% across a concentration range of 3·108 to 2·1011 mL−1 for concentration measurements was also demonstrated by using gold nanoparticles and gravimetric dilutions for method validation. Overall, we show that MADLS® is a rapid and straightforward method for the reproducible measurement of particle concentration, as well as size, requiring minimal sample preparation, without the need to calibrate using a pre-determined concentration series, and applicable to a broad range of materials. These features make it an ideal tool to support both development and quality control of particle materials for a broad range of applications.

The preferred orientation of crystallites constituting polycrystalline solid materials such as metals, ceramics or composites significantly influences the intrinsic properties of the respective solid. In material sciences this preferred orientation is commonly known as texture and describes the orientation of all crystallites in a sample with respect to a reference frame defined by the sample orientation. Using X-ray diffraction (XRD) the texture of a sample is usually measured using the approach developed by Schulz (1949) where the intensity of certain (hkl) reflections is measured as a function of the sample orientation. This is conventionally done by rotating (ϕ) and tilting the sample in axial plane (𝜒). Hence, point focus optics as well as an Eulerian cradle are required. In contrast to the conventional Schulz-technique, in the ω-texture approach different sample tilts in the equatorial plane are accessed by applying a ω-offsett to the goniometer while keeping the sample horizontal and rotating it in ϕ (Fig. 1). Therefore ω-texture allows measurements of complete pole figures in line focus without the need for an Eulerian cradle and point focus optics.

Determine particle density with Resonant Mass Measurement Техническая аннотация

The Archimedes' Resonant Mass Measurement technique measures a particle’s buoyant mass which can be used to calculate its mass and size. This requires a value for the density of the particle being measured, as well as the density of the fluid in which it is suspended, to be known. The density of a particle can be obtained using the Archimedes instrument even if its size is unknown. This is done by measuring the buoyant mass of the particle in different fluids of varying densities. This method has been shown to be very effective at accurately measuring lower density particles such as latex. Measuring particle density allows for the accurate calculation of mass and size and can give structural information for particles which are composed of more than one material.

Near-infrared (NIR) reflectance spectroscopy has been used to improve the efficiency of processes and quality of products in many industries. Mining and ore processing are under-utilizing the technology and could benefit from expanded application of NIR. NIR spectroscopy combined with chemometric predictive models are now being increasingly used as rapid techniques to quantitatively analyze mineral ores to provide real-time information that is needed for ore processing. While NIR is most commonly applied to analysis of organic materials, many inorganic minerals have significant NIR features that result from variations in their composition, thus providing the opportunity to create predictive models for these minerals.

Sample preparation by fusion for XRF analysis is all about knowing the exact weights of the sample and the flux (sample-to-flux ratio). The whole analytical chain, including the weighing step in sample preparation prior to fusion, is of crucial importance to get precise and accurate x-ray fluorescence (XRF) results. Consequently, the weighing method will affect the quality of the analytical results given by the spectrometer. In this study, the effects of different weighing methods on the precision (RSD) of the obtained XRF results are compared to determine the best weighing method for sample preparation by fusion in terms of comparable precisions in the XRF results.

Field Characterization of White Micas Using the ASD TerraSpec® Halo Тематическая публикация

The white micas are a diverse group of common fine-grained phyllosilicate minerals that include the true micas paragonite, muscovite, and phengite, as well as the K-deficient mica, illite. This group often serves as a useful exploration vector as they provide an indicator of geothermal and geochemical conditions at the time of their formation. Using the Al-OH scalar reported by the ASD TerraSpec® Halo mineral identifier it is possible to track geochemical conditions while the Illite Spectral Maturity (ISM) scalar (Doulblier et al. 2010) reported by Halo provides an indicator of thermal maturity. The white micas contained in the mineral library used by the ASD TerraSpec Halo mineral identifier are organized by composition. The true white micas are represented by the potassium white micas muscovite and phengite, and the sodic white mica paragonite. The muscovite-phengite series is characterized by increasing substitution of Mg+2 or Fe+2 and Si+4 for Al+3. The illites are classified similarly. Thus, a shift in mineralogy from muscovite and/ or illite (labeled as K-Illite in the Halo mineral library) to paragonite and/or paragonitic illite (labeled as Na-Illite in the Halo mineral library) is indicative of increasingly sodic conditions. Similarly, a shift in mineralogy to phengite and/or phengitic illite (labeled as Mg-Illite in the Halo mineral library) is usually indicative of increasingly magnesium-rich conditions. The ASD TerraSpec Halo mineral identifier is able to track geothermal gradients both by detecting mineralogy and through the reporting of the ISM scalar. With increasing metamorphic grade, smectites convert to illites, which then covert to muscovite (and other true micas). Within the illites, subtle geothermal trends are easily detected and mapped using the Halo’s ISM scalar which tracks the illite dehydration process as the illite converts to true mica.

Asphalt binder emulsions have proven to be an environmentally friendly, energy efficient, and cost-effective alternative to hot asphalt binder mixtures [1]. They are typically used for roadway resurfacing where they are employed as an adhesive for post-applied aggregate chip. They are typically composed of bitumen, water, and an emulsifying agent or surfactant and must give the required stability and application characteristics for optimum performance. For example, the structural layers must have adequate mechanical properties to make them less susceptible to early-life damage. The main component of asphalt emulsions is asphalt cement which constitutes 50-75 percent of the emulsion content. Its properties are critically important since they influence emulsion behavior and performance on the road [1]. Water is the next component and can affect the properties of the emulsion if minerals or other materials are present. One of the most influential components is the emulsifier or surfactant, and more specifically its chemical structure. The emulsifier can determine whether the emulsion is cationic, anionic, or non-ionic. Furthermore, the chemical compatibility of the emulsifier with the asphalt cement greatly affects the emulsions’ stability. The physical properties of asphalt emulsions can also be modified using additives such as polymers or fillers. There are many reasons to modify asphalt emulsions, including; improved resistance to permanent deformation at high temperature; increased low temperature flexibility; improved fatigue resistance; increased tensile strength; and reduced temperature susceptibility. Malvern Panalytical has several tools that can provide valuable metrics for production and characterization of asphalt emulsions. The two tools discussed in this application note are the Mastersizer and the Zetasizer., which can both be used to determine droplet/particle size, and in the case of the Zetasizer to measure the Zeta potential of emulsions also.

A comparison of weighing methods prior to fusion Практическая аннотация

Major investments are often made in state-of-the-art XRF equipment without knowing that the whole analytical chain, including the weighing step in sample preparation prior to fusion, is of crucial importance to get precise and accurate analytical results and consequently obtain estimated financial pay-offs. In fact, precision and accuracy of results enable the manufacturer to decrease the level of uncertainty associated with the concentrations of its products, and therefore avoid huge losses in revenue. The weighing step in sample preparation by fusion for XRF analysis is all about knowing the exact weights of the sample and the flux (sample-to-flux ratio). Consequently, the weighing method, the tolerance accepted as well as the analytical method to obtain this ratio will affect the quality of analytical results given by the spectrometer. There are many ways to weigh the sample and the flux prior to fusion: • Manual weighing (most widespread technique) • Automated weighing (with Claisse® LeDoser™ or LeDoser-12™ for example) • Weighing the sample and the flux directly in the platinum (Pt) crucible • Weighing the sample or the flux in another container, reusable or not, before transferring it into the Pt crucible • Pre-weighed flux vials • Weight correction on the XRF instrument (exact weight needs to be known) All these weighing methods affect the precision of the sample-to-flux ratio and consequently impact the final analytical results. In this application note, the effect of the different weighing methods on the precision (RSD) of the obtained XRF results are compared.

Mining companies can lose millions of dollars if a slight bias is found in their whole ore analysis process. Their profitability then depends on the quality of their elemental analysis. This study shows that it is possible to use NaT flux coupled with an automatic fusion instrument to build a calibration strategy for XRF analysis that meets the ISO 9516-1 requirement for iron ore analysis. A calibration strategy based on CRMs prepared with NaT as flux is comparable to lithium-based flux for iron ore analysis. The developed method allows fast and repeatable analysis of iron ores in accordance with the currently accepted international norm. This method is a nice alternative to lithium-based flux used for sample preparation by fusion.

Comparing multi-detector GPC and APC for polyalphaolefin analysis Практическая аннотация

Polyalfaolefins (PAO) are a range of low molecular weight hydrocarbons, used in a number of lubricant applications. In particular, they are widely used in the industrial and automotive industries where they are used as base oils. The wide functional temperature ranges, high oxidative stability and high viscosity indices all contribute to their desirable qualities. As with so many polymers, the specific physical properties of PAO are strongly determined by their molecular weights. As a polymer of decene, a PAO sample will likely be a mixture of different decene oligomers and polymers, thus the final molecular weight distribution of a PAO sample will depend on polymerization parameters. As a lubricant, the primary tool for characterizing PAO samples will be a viscosity or rheology measurement, however, gel-permeation/size-exclusion chromatography (GPC/SEC) can be used to make measurements of PAO molecular weight distribution, which may offer insight into small differences between samples. Historically, GPC has been a slow technique with measurements taking 30-60 minutes to separate the sample. Additionally, the molecular weight result is determined by comparison to polymer standards of different structure and chemistry, making the result only relative. Despite this, the technique is a powerful tool for comparing samples and has become the gold-standard of molecular weight measurement in the polymer industry. The addition of advanced detectors such as light scattering and a viscometer can help overcome some of the limitations of GPC. Light scattering allows for the direct measurement of the polymer’s molecular weight, independent of its structure or chemistry providing what is often referred to as absolute molecular weight. The use of a viscometer allows for the measurement of some structural aspects including branching. In the last few years, further developments in GPC column chemistry have resulted in the development of Advanced Polymer Chromatography (APC) systems. These use smaller particle sizes to achieve superior resolution but generate higher backpressure. This also offers the benefits of significantly reduced run-time and solvent use. Previously, the narrow peaks generated by APC were incompatible with multi-detection because of the effects of band-broadening (also called dispersion) caused by entry and exit of the detector cells. This has now been overcome and it is possible to combine multi-detection with APC with multi-detection to perform absolute characterization of polymers at the timescales and resolutions of APC. As low molecular weight molecules which are mixtures of oligomers, PAO, are an ideal example of this. In this application note, the separation of PAO by multi-detector GPC and APC are compared and the benefits of multi-detector APC are discussed.

To guide soil fertility investment programmes in sub-Saharan Africa, better understanding is needed of the relative importance of soil and crop management factors in determining smallholder crop yields and yield variability. Spatial variability in crop yields within farms is strongly influenced by variation in both current crop management (e.g. planting dates, fertilizer rates) and soil fertility. Variability in soil fertility is in turn strongly influenced by farmers’ past soil and crop management. The aim of this study was to investigate the relative importance of soil fertility and crop management factors in determining yield variability and the gap between farmers’ maize yields and potential yields in western Kenya. In this study, 522 samples taken from the farms were analysed by diffuse reflectance spectroscopy, using an ASD FieldSpec FR spectroradiometer.

The increased demand for portable electronic devices, including mobile phones, laptops and new ‘wearables’, has required advances in battery technology to provide a low-cost, lightweight, long-lasting and stable power source. With fossil fuels dwindling and CO2 regulations becoming more stringent, battery technology is increasingly being used in applications such as renewable energy storage and electric vehicles, which require ever more lightweight, safe, high-power and fast-charging batteries. During discharge, lithium ions (Li+) move from the anode to the cathode, conducted via the electrolyte, with flow in the reverse direction occurring when the battery is charged. Anodes are typically graphite-based, while cathodes are often manufactured using lithium iron phosphate, lithium cobalt oxide, lithium-manganese oxide, lithium-nickel cobalt manganese oxide, etc. The cornerstones of battery performance are power, which impacts current and discharge characteristics, and energy storage capacity. Battery power is determined by the rate of reaction between the electrodes and the electrolyte, while storage capacity is a function of the volume of electrolyte within the cell. These properties are intrinsically linked to the intercalation structure and primary particle size of the electrode particles, which determine how well the mobile ions are taken up and released by the electrode1. Particle size distribution and particle shape also influence particle packing and hence the volume of electrolyte that can be accommodated within the interstitial voids of the electrode, which affects storage capacity. Since its establishment in 1920, Rand Refinery (close to Johannesburg, South Africa) has refined nearly 50,000 tons of gold – almost one third of all the gold mined worldwide. Also silver is refined to the same high benchmark standards. The refinement process starts at the smelter, which currently processes over 70 different types of material, including by-products from the mining industry, electronic waste, carbon fines and sludge. Chocolate is a complex multi-phase product consisting of fat, milk, cocoa powder and sugar, as well as other additive such as emulsifiers and flow modifiers. Of critical importance for both the sensory characteristics (texture) and processing of chocolate is its rheology or viscosity, which is influenced by many factors including particle characteristics such as size and shape, as well as fat composition and structure. To date the Casson model has been widely used to characterise the flow properties of Chocolate. This model is used to assess differences in chocolate flow properties that can be appropriated to the behavior during processing such as pump-ability and slump characteristics. This application note describes the texture analysis of a rolled copper specimen. The experimental data were measured on an X’Pert3 diffraction system equipped with a texture cradle. In polycrystalline solid materials, such as a wide class of metals and ceramics, usually the orientation of the crystallites, constituting the material, is not distributed randomly, like in an ideal powder specimen. In most cases a preferred orientation of the crystallites with respect to the sample reference frame is present. In materials science this preferred orientation is referred to as texture.

Multi-angle Dynamic Light Scattering (MADLS) offers improvements in particle sizing resolution, sensitivity and accuracy compared to traditional single angle Dynamic Light Scattering (DLS). Manufacturing industries using powder metallurgy (PM) techniques – including additive manufacturing (AM) and metal injection moulding – rely on a supply chain of closely specified, high-performance metal powders to produce precision components at a competitive cost. The physical characteristics of these powders – bulk properties and those of the individual metal particles – define their performance and are closely controlled during manufacture. Packing density and flowability are key bulk properties and are directly, though not exclusively, influenced by particle size. Accordingly, it is vital to identify analytical tools that can reliably set size specifications for metal powders, to validate quality and manage their use. Sieving has long been a favoured technique for routine particle sizing. However, it also has many limitations that are addressed by laser diffraction. In this article Dr Sarennah Longworth-Cook and Parminder Singh explore the importance of metal powder particle size, and compare the value of sieving and laser diffraction within this context.

The most common approach for measuring viscoelastic properties using a rotational rheometer is to perform small amplitude oscillatory shear (SAOS) testing within the samples linear viscoelastic region (LVER). An alternative approach for obtaining time dependent viscoelastic information is by means of a creep test in which a constant and unidirectional shear stress is applied to the sample and the resultant shear strain measured with time. While creep and oscillatory tests, and the results generated, are often considered independently they are in fact intrinsically linked. As with spectroscopic techniques such as NMR the time domain data can be transformed to the frequency domain using mathematical procedures such as the Fourier transform or equivalent algebraic procedures. One such algebraic procedure is based on an approach widely used in the field of microrheology for converting the mean square displacement to complex modulus which has also been adapted for converting creep compliance data also. In this technical note we demonstrate how such an approach can be used to generate comparable data to oscillatory testing but in a fraction of the time.

EPA Tier 3 Compliant Analysis of Ultra Low Sulfur Fuels Практическая аннотация

Introduction Environmental regulations set the maximum permitted sulfur level in car fuels at 10 mg/kg. Several different X-ray fluorescence technology test methods are available for analyzing the sulfur concentration, for example, ASTM D4294 (EDXRF) and ASTM D2622 (WDXRF). The environmental Protection Agency (EPA) Tier 3 regulation introduced in 2017, is XRF technology independent, with the condition that the performance of the instrument meets the Tier 3 accuracy and precision requirements. This application note shows that Epsilon 4 complies with the Tier 3 requirements in only five minutes measuring time.

Dynamic light scattering (DLS) is a technique that is widely used to measure dispersed particle size (specifically the hydrodynamic diameter) via direct measurement of the translational diffusion coefficient within the continuous phase. By utilizing the superior accuracy and resolution of multi-angle dynamic light scattering (MADLS®), it is now possible to determine the absolute particle concentration distribution using the Zetasizer Ultra. This provides the user with a fast assessment of the concentration of each population of particles present within their dispersion. Particle concentration measurements are achieved by recording the time averaged photon count-rate scattered by the sample to perform a transformation of the size distribution to derive the absolute number of particles present. In this technical note we describe the method and calculations used to determine particle concentration.

Multi-angle dynamic light scattering (MADLS®) available on the Zetasizer Ultra delivers an angular-independent particle size distribution (PSD) with increased resolution by combining scattering information from multiple angles. Visualizing structural changes in batteries using hard radiation and the GaliPIX3D detectorIn this data sheet, we present CT results obtained on alkaline batteries using an Empyrean diffractometer. Zn – MnO2 alkaline batteries were investigated by computed tomography using an Empyrean multipurpose laboratory diffractometer in combination with hard X-radiation (Ag anode) and the GaliPIX3D detector. The differences between the charged and the discharged state can be clearly observed and are in agreement with results obtained at synchrotron radiation facilities.

Polyvinyl chloride (PVC) is a common polymer used in countless applications from pipes, to flooring, and electrical insulation. As one of the world’s most widely produced polymers, it is very much a commodity polymer where tight margins mean little room for detailed analysis. However, even in this case, high level analytical tools can be used to characterize the polymer for appropriate grading. For example, the glossiness of a PVC sample has a significant effect on its finish and grade, and for which applications it can be used. Applying appropriate characterization tools can help grade different PVC batches, assigning them to appropriate applications to maximize the value of high quality batches, saving lower grades for others. In this application note, four PVC samples of known differences in glossiness were measured with both multi-detector GPC and rotational rheometry. The results both explain the differences in glossiness and the likely cause of those differences.

For virus development and production, it is important to know the virus concentration at different stages of the process, to optimize the clone used as well as the production yields. For example, instances such as clone screening, multiplicity of infection (MOI) optimization and adaptations of cell culture methods are instances when virus concentration, or virus titre as it is also commonly called, is of interest. Dynamic light scattering (DLS) is often used in virus development, in a screening function to separate good and stable samples from those with contaminants or aggregates. With the new multi-angle DLS (MADLS) based particle concentration measurement in the Zetasizer Ultra, it is now possible to get both size and size distribution as well as particle concentration per population, within a few minutes. In this application note, Allergan are kindly sharing data from their evaluation of the Zetasizer Ultra, examples of three adeno-associated virus (AAV) samples are shown. The concentration results are compared to results from capsid ELISA based virus titre assays.

Gel permeation / size exclusion chromatography (GPC/SEC) is the most well-known method for polymer and macromolecular analysis. As the technique has become more pervasive, there has been a push toward greater efficiency, by way of reduced run times and minimal solvent and sample consumption. The development of Advanced Polymer Chromatography (APC™) systems has accommodated these issues, by employing low volume columns and pumps with the ability to handle higher pressure generated by these columns. These features allowed for more efficient polymer analyses, but unfortunately, the technique still relies on the need for column calibration to provide relative molecular weight values. To determine if the efficiency of the ACQUITY APC unit and the power of the advanced detection of OMNISEC REVEAL were additive, two polymer samples with different structural features were analyzed. The goal of this experiment was to obtain enough characterization data to differentiate between linear and branched polystyrene samples – all in a matter of minutes.

Size-exclusion chromatography (SEC), also known as gel-permeation chromatography (GPC) is a well-known and widely used tool for measuring polymer molecular weight. Multi-detector SEC measurements including refractive index (RI) and light scattering (LS) can measure absolute molecular weight, independent of a molecules shape, structure, chemistry, or conformation. Polymer concentration is measured by the RI detector alone. The addition of a viscometer allows the measurement of intrinsic viscosity (IV), which is used to study conformation and branching, and can expose any structural changes that the polymer may undergo. Typical analytical SEC measurements can take approximately 25 to 45 minutes and consume 25 – 45 mL of mobile phase. This is time-consuming and can be expensive if working with organic solvents. Ultra- performance liquid chromatography (UPLC) systems use novel SEC column technologies with robust, small particles (<2 µm) to achieve better sample resolution using smaller columns. This increases productivity, while significantly reducing run-time and cost. An additional benefit of the reduced solvent use is to effectively make UPLC a ‘greener’ technology than traditional analytical SEC. The combination of multi-detector SEC and UPLC has, up to now, been challenging due to the significance of inter-detector band broadening (or dispersion); however, the latest developments in integrated multi-detector systems allows up to four detectors to be connected to UPLC systems with only a minimal loss of resolution due to band broadening. In this application note, Malvern Panalytical’s OMNISEC REVEAL multi-detector module was combined with a Waters ACQUITY™ APC™ system to offer the best of both worlds; high resolution-rapid multi-detector SEC characterization of a PLA-PEO-PLA drug delivery polymer. The molecular properties of a virgin sample were compared with two samples sterilized by radiation.

Use of NIR Spectroscopy in Mineral Identification in Shale Практическая аннотация

Objectives: (i) to estimate mineralogical composition specifically clay mineralogy from the spectra, (ii) to qualitatively compare mineral concentrations calculated from XRF and XRD analyses, as well as mineral groups identified through SEM and microscopic petrography work with NIR results. NIR spectroscopy has been widely used in different scientific fields such as biology and medicine. However, it has been rarely used in mineral identification in finer sedimentary rocks specifically mudrocks. This work will allow us to verify the accuracy of this technique compared with XRF and XRD.

Visible and near infrared reflectance spectroscopy is an accepted method for identification of a wide range of minerals including the phyllosilicates and carbonates. The combination of portable instrumentation and automated analysis methods enables real-time identification and characterization of these minerals at the wellsite. In sedimentary basins, the technique provides clay mineralogy, thermal maturity, and carbonate composition. The dominant phyllosilicate minerals found in sedimentary systems, including smectites, illites, kaolinite, and chlorite, have sufficiently unique spectral signatures to allow for both their identification and characterization. Kaolinite crystallinity is inferred from the shape of a doublet absorption feature. The progressive conversion of smectite to illite is tracked by monitoring the shape of a Al-OH absorption feature as well as the relative depths of water and Al-OH absorption features. Since oil generation takes place simultaneously with the diagenetic transformation of illite/smectite, substitution of NH4 for K in the interlayer sites of illite is often observed if hydrocarbons are present at the time of illitization. It is possible to determine the presence of ammonium illites since ammonium has unique absorption features near 2120 and 2020 nm. Reflectance spectra of chlorites typically have two hydroxyl absorption features (Fe-OH & Mg-OH) that shift in position with varying Fe/Mg ratios. Chlorites also exhibit a similar change in water to hydroxyl absorption feature depth with varying thermal maturity as do the illites. The Ca/Mg ratio of carbonates is determined from the position and shape of the CO3 absorption. The spectrum of the iron carbonate siderite, has a broad Fe2+ feature centered near 1200 nm and the CO3 is near 2335 nm. In addition to mineralogy, reflectance spectroscopy is also applicable to characterization of organics in sedimentary rocks. It has been used for analysis of kerogen in oil shales and bitumen abundance in oil or tar sands. And as with many minerals, thermal maturity of hydrocarbons may be inferred from systematic changes in the reflectance spectrum.

Visible and near infrared reflectance spectroscopy is an accepted method for identification of pathfinder minerals that serve as exploration vectors. The method is now widely used to assist in new deposit discovery as well as to provide key mineralogical information during mine development. The combination of portable instrumentation and automated analysis methods now allow for real-time identification of pathfinder minerals as well as the determination of associated geochemical and geothermal parameters. The reflectance spectrum of white micas and chlorite group minerals contain information related to composition, crystallinity and metamorphic grade. The wavelength positions of Al-OH, Fe-OH and Mg-OH absorption features are highly correlated to chemical composition, b unit cell dimension, and Kübler index values. For white micas, both the Nato- K ratio in interlayer sites and the degree of Fe or Mg substitution for Al in the octahedral layer influence the position and shape of the Al-OH absorption feature. The depth of absorption features associated with structural water in these minerals varies with thermal maturity. With the dehydration associated with increasing metamorphic grade, the ratio of the depth of the water absorption feature to the depth of Al-OH, Fe-OH and Mg-OH features decreases. Together, these parameters enable detailed mapping of geochemical and geothermal gradients, and have contributed to the discovery and development of a wide range of economic deposit types. Pathfinder minerals that serve as vectors to economic deposits are present many geologic systems. Rapid reflectance spectroscopy enables their identification, as well as the determination of their associated compositional, crystallinity, and thermal maturity parameters, and provides the means to map geochemical and geothermal gradients in these systems. In many cases, the relationship between the properties of these minerals and the proximity to the ore zone provides a very useful exploration vector.

Early detection of oak wilt disease Практическая аннотация

Initial work has begun on a long-term study of the application of spectroradiometric techniques to spectral discrimination of oak wilt disease in Quercus woodlands of central Texas. In situ hyperspectral data of the leaf, branch, and canopy level of the most commonly affected species will be collected, processed, and analyzed for discriminatory signatures of the disease in various stages of disease pathology. A controlled inoculation experiment will be conducted to provide the baseline spectral response in the absence of potentially disease-masking environmental factors such as drought, and other pathogens. Data obtained from the inoculation experiment and from the field will lead to the development of classification methods that can be applied to hyperspectral imagery that will be obtained over the study area in a scheduled acquisition from the Hyperion satellite. The ultimate goal of this research is to determine the feasibility of early disease detection through the application of hyperspectral technology.

The economy and efficiency of Waters’ ACQUITY APC combined with the powerful advanced GPC/SEC detectors of OMNISEC REVEAL allowed the complete characterization of the low molecular weight polystyrene and polycaprolactone samples. The individual oligomers of both samples could be observed and characterized separately due to the resolution of the APC columns and sensitivity of the OMNISEC REVEAL detectors – with standard 30 cm analytical GPC/SEC columns it is not possible to resolve the oligomers at all. The greatly reduced quantity of solvent and time required to obtain comprehensive characterization of samples using the APC-REVEAL system presents obvious benefits to both researchers and manufacturers working with polymeric materials.

Liposomes are nowadays frequently used as drug delivery systems, and as such requires that they are well characterized for manufacturing and batch release. This characterization includes their size and size distribution. For these parameters, dynamic light scattering (DLS) is commonly used to characterize size and size distribution and also nanoparticle tracking analysis (NTA) has previously been shown to be useful for measuring liposomes size and concentration.1 This application note will demonstrate how the multi-angle DLS (MADLS®) based particle concentration measurement in the Zetasizer Ultra is capable of measuring the particle concentration as well as size and size distribution of a range of dilutions of a liposome sample. A comparison will be made with NTA measurements of the same samples.

Dynamic Light Scattering (DLS) is a powerful and sensitive technique for characterizing particles or macromolecules in dispersion, due to its ability to resolve particle or molecular sizes ranging from sub nanometer to several microns. This sensitivity also means that DLS is a useful technique to characterize aggregated material, which may occur in far smaller quantities but is of a significant importance in many applications. Typically, however, the presence or dust (including filter spoil, column shedding, tracer aggregates or material from dirty lab ware), can have detrimental effects on the measurement of smaller sized particles and algorithms exist to suppress these effects. In this application note we present a new approach to handling DLS data which prevents the skewing of data for small particles whilst retaining insight into the presence of aggregates that may otherwise be lost, whereby the relative abundance and size of aggregates can be deduced.

Dynamic light scattering has been vastly explored as a tool for micelles characterization, not only for their size and stability, but also to determine the critical micellization concentration (CMC) and temperature (CMT)1-3. As a result of the unimers aggregation, as a function of the temperature for the case of this study, an increase in the scattered intensity is expected due to larger particles scattering substantially more photons. It’s been reported that for light-scattering techniques to be used reliably as CMC or CMT determination techniques, long correlation times are needed 1,4,5 which make this time-consuming and hence it’s not usually considered as a primary technique. We suggest in this application note, that DLS can be used in conjugation with other techniques to determine the CMT in a highly reproducible and time-efficient manner.

Dynamic Light Scattering (DLS) allows the hydrodynamic size of particles in dispersion to be measured by quantifying their diffusive motion. At particle sizes approaching the upper size range for DLS, sedimentation, thermal currents and number fluctuations can mean that the detected scattering no longer accurately describes purely diffusive motion, and particle size analysis becomes less accurate. Here we will discuss how the Low Volume Disposable Sizing Cell can improve data quality at large particle sizes and allow the entire size range of DLS to be explored without modification of the dispersant system.

When producing particles of a known size, the monodispersity of the samples in terms of both size and shape is important and often requires a high-resolution technique such as TEM to look at these sample properties. For example, Particle Works, a brand of Blacktrace Holdings Ltd, produce gold nanoparticles which must be highly monodisperse and have high batch-to-batch consistency, with CV values as low as 5% for the size distribution and 2.5% for the batch consistency. Currently, Particle Works use TEM as the primary technique for characterizing their samples for QC. However, TEM is a time consuming and expensive technique that requires an experienced user to perform analysis. In this study we show how the Zetasizer Ultra can be used to identify the majority of 'out of specification' batches of gold nanoparticles and therefore significantly reduce the amount of TEM analysis required.

Adaptive Correlation: Better DLS data with less time and effort Практическая аннотация

Whilst being able to measure particles of below 1 nm size, DLS is preferentially sensitive to larger particles due to the 6th power relationship between particle radius and scattering intensity. This means that sample preparation typically needs to be scrupulous, especially for low scattering samples such a proteins and biological molecules. The contribution to contaminants such as dust and aggregates can be mitigated by filtering, however this may not always be practical or possible depending on the volume and fragility of the sample. Filtration of samples can also constitute a financial burden, both in terms of additional sample preparation time and consumables costs. This application note introduces a new DLS data capture process called Adaptive Correlation which helps address these issues. Adaptive Correlation uses a statistically driven approach to produce the best correlation data, which in turn gives more reliable DLS size data. It can reduce the need for filtering and give added confidence in you DLS results.

ASD systems are used to determine plant physiological status, including disease status, nutritional status related to nitrogen uptake and moisture balance. In the metal industry, some samples can be challenging to analyse. An inappropriate sample preparation method can lead to inconsistent analytical results and even render the samples impossible to analyse. For example, concentrates that contain partly reduced materials can cause problems when using lithium borate fusion. If this type of sample is not correctly oxidized during the sample preparation process, it can chemically alloy and attack the platinumware. Corn stover is regarded as the prime candidate feedstock material forcommercial biomass conversion in the United States. Variations in chemical composition of Zea mays cell walls can obviously affect biomass conversion process yields and economics. Mutant lines were constructed by activating a Mu transposon system. The cell wall chemical composition of 48 mutant families was characterized using near-infrared (NIR) spectroscopy. NIR data were analyzed using a multivariate statistical analysis technique called Principal Component Analysis (PCA). PCA of the NIR data from 349 maize leaf samples reveals 57 individuals as outliers on one or more of six Principal Components (PCs) at the 95% confidence interval. Of these, 19 individuals from 16 families are outliers on either PC3 (9% of the variation) or PC6 (1% of the variation), the two PCs that contain information about cell wall polymers. Those individuals for which altered cell wall chemistry is confirmed with wet chemical analysis will then be subjected to fermentation analysis to determine whether or not biomass conversion process kinetics, yields and/or economics are significantly affected. Those mutants that provide indications for a decrease in process cost will be pursued further to identify the gene(s) responsible for the observed changes in cell wall composition and associated changes in process economics. These geneswill eventually be incorporated into maize breeding programs directed at the development of a truly dual use crop.

Forests are a natural resource of major economic significance to Canada and many other countries. For example, Atlantic Canada forest industries exported over $3.9 billion in wood products and had a direct employment of 37,900 persons in 2003. Secondary and value added products, particularly that of Canadian northern bleached softwood kraft market pulps represent revenues reaching $7.5 billion in 2007. Canada leads the world in wood pulp exports. In addition to direct employment, the industry has an indirect effect through demand for goods and services and diversifies income sources for many communities. More recently, wood is recognized as a renewable resource and can sequester CO2. There is increasing emphasis on using wood for energy, bio-fuels and bio-chemicals production. Timely knowledge of wood characteristics is essential to ensure that the right fiber is directed to the appropriate wood manufacturing company at the right time and right cost to produce the right product. Agenda 2020 has identified that a number of key variables that need to be measured accurately along the wood processing, from trees to logs to lumber and to secondary processing such as pulp and paper products. Measurements of wood and processing parameters will allow process monitoring and control, leading to consistent high product quality at minimal costs and energy consumption. Reflectance spectroscopy, particularly visible-NIR spectroscopy, has shown much promise as an adaptable technology that would meet the requirements for fast, non-contact, and process capable as online and at-line analysers for monitoring wood and wood products properties. This paper will discuss the application of reflectance spectroscopy for measurements of standing tree wood properties, lumber properties during sawmilling operation, OSB flake properties and its use in panel production, and other pulp and paper applications. The paper will focus on adapting reflectance spectroscopy for on-line and at-line measurements for 24/7 operations.

Mineral physics dictates the appearance of rocks and soils across the electromagnetic spectrum. In the Visible/Near-Infrared (VNIR) and Short Wave Infrared (SWIR), many materials absorb radiation at specific wavelengths, allowing their identification by the position and character of absorption features. Electronic processes at wavelengths less ~1.0 micrometers allow identification of minerals containing Fe+3. Molecular vibrational features at wavelengths between ~1.0 and 2.5 micrometers are diagnostic of minerals containing anion groups such as Al-OH, Mg-OH, Fe-OH, Si-OH, CO3, NH4, and SO4. Small differences in absorption band position and shape are correlated with mineral compositional differences and variability. Imaging spectrometry has been used since the early 1980s to perform 2-dimensional mapping of mineral distribution based on spectroscopic characteristics. Geologic applications include areas such as lithologic mapping; exploration for precious and base metals; and oil, gas, and geothermal energy exploration. Field spectroscopy plays a critical role in the calibration, analysis, and validation of imaging spectrometer data. Spectral libraries have been measured for a variety of minerals. Imaging spectrometer datasets have been acquired around the world using airborne platforms and recent satellite systems provide spectral measurements for selected areas. Case histories will be presented demonstrating the link between laboratory, field, and imaging spectrometer data. Selected physics-based analysis methodologies will be discussed and the level of information available from the data will be demonstrated. Examples will include mineral identification and mapping in the context of hydrothermal alteration associated with active and fossil hot springs and mineral deposits. Specific mineral mapping examples will include hematite, goethite, and jarosite in the VNIR and identification and separation of calcite, dolomite, and calc-silicates along with phyllosilicates and sulfate minerals such as alunite and jaorsite using SWIR spectroscopy. Spectral variability caused by processes such as anion substitution in illite/muscovite, crystallinity in kaolinite-group minerals, and spectral mixing will also be discussed.

A wide range of factors affect the reflectance spectrum of a given soil sample. For example, variability can be introduced by the spectrometer itself, the experimental protocol and sometimes even by the operator. Unlike spectra of rock and minerals where easily recognized spectral features are sufficient for diagnostic assessment, it is more subtle spectral data that reveals the most characteristic information about a soil sample. Consequently, technical factors that alter the reflectance spectrum need to be identified and minimized and, if possible, eliminated or at least standardized. The need to minimize such factors in a robust way is important especially now that a worldwide soil spectral group has been established and soil spectral libraries are exchanged routinely between members. This paper summarizes a comprehensive and systematic study performed with this goal in mind. A group of 12 chemically defined soils were assessed spectrally by multiple operators using three different ASD spectrometers that require different protocols and measurement conditions. Despite the definition of strict protocols, for some soil samples there was great variation among the spectra generated by the three different ASD spectrometers that was exacerbated when more than one operator performed the spectroscopy. Therefore, we decided to investigate an approach taken from the wet chemistry discipline, whereby an internal and well-defined standard is included in every measurement cycle. Three different materials were examined for that purpose and a method was developed to normalize spectra to the spectra generated by a ‘mother’ spectrometer and protocol. We propose that this method enables the variable spectra of a given soil sample to be translated into a common denominator and thus should facilitate the exchange of spectral information among scientists worldwide and allow creation of a robust soil database for diverse applications.

Astronomers began the use of multi-wavelength studies of Solar System objects in the solar spectrum region, first by eyeballing the objects’ colors, such as for Mars, and quantitatively when the photographic process was developed in the last half of the 19th century. After the time of Galileo, astronomers came to emphasize gas and plasma studies, while solar system studies included dirt and rocks as subjects. I am more the dirt and rocks type and I will emphasize this history. I arrived on the scene in the 1960s when electronic detectors first began to trickle out to civilian scientists. These allowed far more accurate quantitative measurements, as well as measurements at infrared wavelengths. We, who had access to these new detectors, began using the astronomers’ telescopes to observe solar system objects almost at random during the nights we could get precious telescope time. My own early emphasis was to demonstrate that there are spectral expressions of rock-forming minerals (basalts) on the Moon. This seems primitive by today’s standards, but it is where we were in the1960s. Solar System studies grew from there, first with telescopes and then using spacecraft to carry our spectrometers to the objects. Spacecraft history developed slowly due to the dominance of geologists and picture taking over geochemists and the less visual spectroscopy. The delay of the launch of the Galileo spacecraft, due to the Challenger disaster, the first deep space mission to carry a visual-infrared imaging spectrometer, was a further setback. But we (and our graduate students) eventually succeeded at this. Today, we still find the telescope is an important tool but, nearly every spacecraft to Solar System object carries imaging spectrometers operating in the UV-visual-IR spectral region, where reflected solar radiation and emitted thermal radiation is detectable.

Environmental monitoring over land surfaces has various complexities caused by highly heterogeneous surface structures with vegetation layer, soil surfaces, and atmospheric disturbance. The vegetation biophysical parameters must be provided with sufficient accuracy to be used as an input of general circulation models, where the satellite remote sensing also plays an important role. Satellite remote sensing is the use of vegetation index (VI) as an intermediate variable to quantify status and amount of vegetation from satellite images. Various VI products currently exist and more VI products are expected to be available simultaneously, the estimation of biophysical parameters through VI has the be compatible by various sensors to ensure continuity of global environmental simulation over generations. Therefore, it is important to investigate the compatibility of VI products for new sensors with existing and old satellite sensors every time a new satellite will be launched. A soon to be launched optical sensor on board Japanese ADEOS-II platform, named GLobal Imager (GLI), is the focus of this study.

Operational impacts of Glen Canyon Dam on downstream resources of the Colorado River ecosystem have been the subject of intense study and discussions among adaptive management stakeholders that include Federal, State and tribal entities, as well as recreational businesses, environmentalists, power companies, and others ( U.S. Bureau of Reclamation 1995, Schmidt et al. 1998). The Grand Canyon Monitoring and Research Center (GCMRC within the U.S. Department of Interior) has identified the following key objectives for long-term monitoring of vital components of the ecosystem between Lakes Powell and Mead, while providing stakeholders with information for decision making: 1) lowest possible impact on critical resources; 2) highest possible return of integrated scientific information; and 3) long-term, repeatable data collection. Airborne hyperspectral image data are being considered as a possible long-term monitoring tool, which hold the promise of being able to fulfill the above requirements. The present study is a pilot project which is intended to explore the extent and variety of information, relevant for long-term monitoring, that can be extracted from images of both high spectral and spatial resolution. The Colorado River ecosystem in Glen Canyon and Grand Canyon poses special challenges for remote monitoring with respect to logistics required to support field-based studies and relative to spatial scales at which monitoring is needed. The narrow corridor of the river is confined by tall vertical cliffs, yet provides a habitat for a great variety of both native and non-native plant species. Riparian vegetation consists of stands that often cover only very small areas. Often, these stands consist of intermixed native and non-native species, and spectral signatures of the various plants sometimes display only subtle differences. Adequate change detection mapping requires both high spectral and high spatial resolution, as well as advanced processing algorithms that can detect subtle spectral differences. Change detection for evolving sand bars, rapids and debris fans is a critical part of the GCMRC's long-term monitoring program. Additionally, changes in the aquatic ecosystem's food base, such as abundance and distribution of algae and other benthic organisms are of great interest. Presently, the need to lower river stage for 2- 3 days during annual monitoring overflights for conventional aerial photography and traditional surface measurements introduces artificial and undesirable impacts on the ecosystem (e.g., Blinn et al. 1999). With hyperspectral imagery, we expect to map riparian vegetation, terrestrial sediment deposits, as well as sand bars and benthic organisms through some depth of water, obtaining greater compositional information than can be obtained through the use of aerial photography.

Cement is the key component of the concrete structures we see and use every day, from our cities and roads, to every bridge and every home. Cement manufacture is both expensive and energy intensive: energy consumption accounts for more than 50% of total manufacturing costs, with electrical energy accounting for a significant proportion of this – historically estimated to be 1% of the world’s electrical energy (circa 1966). However, electricity usage for cement manufacture has steadily been overtaken by that used to power the world’s data centres, which store the vast amounts of data generated over the internet. Waste oil with high chlorine concentrations is considered to be hazardous and must be treated differently than other waste oils. A common problem with the analysis of chlorine by EDXRF is the line overlap with sulfur due to the often high concentrations of sulfur in waste oils. The Epsilon 4 is equipped with a new high resolution SDD detector, which improves the separation of S Kα, S Kβ and Cl Kα peaks.

This data sheet demonstrates that the Epsilon 4 – a benchtop energy dispersive X-ray fluorescence (EDXRF) spectrometer – is exceedingly capable of assessing the purity of fluorite mineral samples that are prepared as fused beads. Fluorite (also called fluorspar) is the mineral form of calcium fluoride, CaF2 and is a widely occurring mineral found in large deposits. Fluorite is used as a flux in iron and aluminum smelting and in the production of certain glasses and enamels. The purest grades of fluorite are also a source of fluoride for the production of hydrofluoric acid, which is in turn used in the production of fluorine-containing fine chemicals and optically clear transparent fluorite lenses. In order to determine the individual applications for fluorite, the purity of the mineral must be quantified in an accurate way. Wet-chemical methods, including multiple digestion, precipitation and titration steps are traditionally employed for fluorite grading. These methods are notoriously time consuming and require the use of harmful chemicals by skilled laboratory staff. EDXRF analysis provides a useful alternative for classifying fluorite samples in a much faster, safer and cost-effective way.

X-ray fluorescence spectrometry (XRF) is widely used for the analysis of additives and wear metals in fresh and used lubrication oils. It provides fast, cost-effective, accurate and precise results with a minimum of sample preparation. On top of fundamental XRF benefits, Oil-Trace provides improved accuracy, savings on setup time, standards and calibration maintenance. Traditionally, different calibrations have to be prepared for different types of fresh and used lubrication oils. In recent years, new developments in the area of size exclusion chromatography have resulted in the introduction of Ultra High-Performance Liquid Chromatography (UHPLC or UPLC®) with Waters ACQUITY Advanced Polymer Chromatography (APC™) a breakthrough technology that is able to provide SEC analyses of polymeric materials with higher-resolution and higher-speed than conventional SEC.. However, the APC system is limited to conventional calibration, meaning it can only generate relative molecular weight information. In order to obtain comprehensive knowledge of the polymers under analysis, it is necessary to combine the high-resolution APC system with additional advanced detectors such as Malvern Panalytical's OMNISEC REVEAL. The combination of the OMNISEC-REVEAL multi-detector system with the APC System provides high quality, high-resolution and rapid sample analysis. The presence of the highly sensitive LS and RI detectors and the viscometer in the OMNISEC REVEAL system allowed for the calculation of absolute molecular weight, hydrodynamic radius and structure of the materials. In this study, the analyses of several polymers by Waters APC combined with the Malvern Panalytical OMNISEC REVEAL multi-detector system are presented and discussed in order to demonstrate the benefits of combining the two systems.

Gisèle Usanase, a Masters Student at C2P2 laboratory (Chemistry Catalysis Polymers and Processes) based at University of Lyon describes her experience of working with the Kinexus and how it provided a quick and inexpensive means for quantifying co-monomer content in LLDPE. C2P2 is a research center with a major focus on fundamental chemistry, catalysis and chemical engineering to enhance the ability to produce organic, inorganic and composite materials, and to propose the processes used for this purpose. C2P2 brings together a range of unique skill sets and capabilities which enable them to perform high level research, based on original concepts, and backed up by a scientific approach. As part of her Masters project, Gisèle Usanase wanted to use rheology as a tool for probing the physical properties and composition of LLDPE and approached Malvern Panalytical for assistance with this project. Dr Frédéric Fraϊsse, Malvern Panalytical’s Rheology Specialist in France, was able to provide access to a Kinexus Ultra+ rheometer along with training and support so that Gisèle could complete her project.

Calcium carbonate is a versatile material used in applications covering fuels, agriculture, pharmaceuticals, and printing materials. The two methods of production are ground calcium carbonate (GCC) – where chalk,limestone, and dolomite are ground to the desired particle size – and precipitated calcium carbonate (PCC),where calcinated limestone is re-carbonized with carbon dioxide into calcium carbonate. Precipitated calcium carbonate is the method preferred by industry for the ability to create a product with a specified particle size, particle size distribution, surface area, shape, and surface chemistry. In paper and printing material production, calcium carbonate is used as a filler material that offers superior whiteness, brightness, gloss, and printability. It is also a more cost-effective solution when compared to GCC, kaolin clay, and wood pulp. Particle size distribution of PCC affects final coated paper whiteness and brightness through tightly controlled packing densities. In this application note we show how Insitec LPS 9 (On-line liquid process system) can provide real time particle size distribution data, which provides manufacturers with continuous particle size data allowing early detection of process upsets and reducing the need for protracted offline sample analysis .

Introduction This white paper outlines the main benefits of using hard radiation for material analysis studies. X-ray powder diffractometers are generally equipped with X-ray sources using Cu anodes. For specific applications, however, switching to shorter wavelength, ‘harder’ radiation, like obtained from X-ray tubes with Mo or Ag anodes, gives significant improvements over Cu, or even allows experiments that are impossible with Cu radiation, like experiments where a large Q range needs to be probed. Often, people make use of synchrotron radiation, but new developments on source, optics and detector technologies allow a variety of hard radiation experiments in the home lab. In this white paper we present six key advantages of hard radiation studies. Larger penetration depth For inorganic materials, the penetration depth of Cu K-alpha is often only a few micrometers (see Table 1). In order to extract information from a larger sample volume, hard radiation is an advantage in transmission experiments. Thanks to the larger penetration depth, it is possible to observe crystallographic changes in working devices, such as Li-ion batteries, and perform highpressure experiments with diamond anvil cells. Hard radiation allows using transmission geometry for inorganic samples. Table 1. Modern batteries like lithium-ion have revolutionized our day-to-day lives from smart mobile devices to pollution free electric cars and intelligent power management solutions. Batteries also hold the potential to being economical alternatives for mass energy storage to compliment renewable energy resources for power grid applications. Despite these successes, gaps in the battery technology remain in terms of safety as well as performance. Equally important is the cost reduction for their mass scale adoption in applications like electric vehicles. These core issues are driving not only the research in new battery materials, but also the improvement in production efficiency to minimize production costs. The imminent issue of rising pollution levels in large cities has generated a great amount of interest in electric vehicles across the globe, making it the main driver for the development of improved battery technology in terms of superior mileage, faster charging time, better safety, and at the same time at a reduced cost. The battery market is expected to grow at a compound annual growth rate of >10% over the next 10 years, primarily driven by the electrification of the transport sector. At the same time, new breakthrough technologies are expected to be commercialized which will boost the energy efficiency, power density and the safety of the rechargeable batteries. Electrodes used in lithium-ion batteries have a defining influence on their electrochemical performance and are typically manufactured by coating a metal foil substrate with a multi-component slurry made of active electrode particles and conductive additives suspended in a binder solution. Size and shape distribution of electrode particles influences not only the rheology of this slurry in terms of its stability and ease of application, but also defines the quality parameters of the resulting coat like thickness uniformity, packing density and porosity. These, in turn, impact key battery performance parameters, such as ion transfer rate and battery recharge time. This whitepaper provides an overview of how measurements of various parameters can help to optimize the properties of the slurries used to create electrodes for lithium-ion batteries. A key focus are the electrode materials that influence the performance of the manufactured battery cells. It describes the analytical techniques that support the optimization of these materials in research phase as well as in the quality control phase during the production process.

The application of atmospheric models to AVIRIS and other spectral imaging data to derive surface reflectance requires that the sensor output be calibrated to absolute radiance (Green et al, 1996; Chrien et al, 1996; Gao et al, 1993). Uncertainties in absolute calibration are to be expected, and claims of ±2% accuracy have been published (Chrien et al, 1996). Measurements of accurate surface albedos and cloud absorption to be used in radiative balance calculations depend critically on knowing the absolute spectral-radiometric response of the sensor. The Earth Observing System project is implementing a rigorous program of absolute radiometric calibration for all optical sensors (Butler and Johnson, 1996a) Since a number of imaging instruments that provide output in terms of absolute radiance are calibrated at different sites, it is important to determine the errors that can be expected among calibration sites. Another question exists about the errors in the absolute knowledge of the exoatmospheric spectral solar irradiance (Nekel and Labs, 1984).

Although there are more than ten known chromium minerals, only one is a source of commercial importance. This mineral is known as chromite and has the theoretical composition FeCr2O4 containing 68 % of chromic oxide ( Cr2O3 ), in which the proportions of Mg2+, Fe2+ and Cr3+, Al3+, Fe3+ may vary considerably. The main product generated by chromite is ferrochrome, a major player in the steel industry. In particular the stainless steel industry uses more than 90 % of the world’s chromite output. The mining and steel industries must assess the quality of the chromite ore to optimize the grade of their stainless steel production. As a result, the chemical analyses of the chromite ore as well as its final and waste products are mandatory. Metal analysis traditionally uses AA or ICP-OES to measure the metal contents in the ores and industrial products. However, the traditional dissolution method for chromite and ferrochrome is a multi-step, multi-acid digestion which requires the use of HNO3, HF and HClO4 and can take between 1 to 3 hours.

Since the 1990s, the automotive industry has become the largest consumer of platinum (Pt), palladium (Pd), and rhodium (Rh) on account of the arrival of catalytic converters in car exhaust systems. Approximately 40% of the total Pt, 74% of the total Pd and 80% of the total Rh were consumed by the auto catalyst industry in 2012. Malvern Panalytical has developed a simple sodium peroxide fusion method, for the dissolution of automotive catalytic converters substrates samples. Combined with a simultaneous ICP-OES, it yields good accuracy and precision for the determination of Pt, Pd and Rh contents in autocatalysts. The Claisse® fusion method is quick, allows total sample dissolution, eliminates the use of dangerous acids and demonstrates good accuracy, precision and recovery for Pt, Pd and Rh.
